creation and maintenance of a living biobank - how we do it

우리 손에는 1차 세포 배양(그림 2A&amp;B)의설립률은 대형 시리즈9에서12.9%였다. 새로운 외과 절제 된 표본에서 확장 가능한 종양 세포를 분리 하는 시도의 대부분은 성장 또는 초기 오염의 부족으로 인해 실패. 세포주 설립은 표준 배양 조건(DMEM, 10% FCS, 표준 배양 용기)에서 꾸준한 성장을 이한 3개 통로와 FACS 분석을 통한 상피 분화검증(10)에성공한 것으로 간주되었다. PDX종양(도 2C &amp; D)에서유래한 세포주들은 23.6%의 높은 확립율을 보였으며, 이는 또한 1차 절제된 종양9과는 대조적으로 반복적 시도의 가능성 때문이기도 하다. 그러나, 일부 혼합배양(도 2E)은섬유아세포 성장에서 해방될 수 없거나 섬유아세포 과성장(도2F)으로인해 손실되기도 한다.

PDX 생성 프로토콜의 변화를 고려하면, 마우스 균주 사용 및 또한 몇 년 동안 실험자, 이식에 사용할 수있는 종양 조직의 양에 큰 차이뿐만 아니라, PDX 생성의 전반적인 성공률을 제공하는 것은 사소한 일이 아니다. 최근 2명의 연구원(S.M 및 F.B.)에 의해 수행된 PDX 세대 실험의 아주 최근 시리즈에서, 대장 PDX에 대한 63%의 1차 성장률(그림 3A에서예시적인 히스토릭이 묘사될 수 있음) 및 췌장 PDX(그림3B)의경우 48%가 관찰되었다. 이식 부위에서 뮤린 또는 인간 림프종의 자라나는 것은 비교적 드물지만 성공적인 PDX 아웃성장을 모방할 수있습니다(도 3C). 조직 병리학 검사 외에도 PDX 모델과 기증자 환자 간의 일치는 짧은 탠덤 반복 (STR) 분석(그림 3D)에의해 정기적으로 확인되었습니다. 현재까지 바이오뱅크는 >50초및 >50초, 3차 및 6차 췌장암 세포주와 >150대장 및 19개의 췌장 PDX 모델로 구성된다.
