多発性硬化症(MS)は、炎症、脱髄、および軸索喪失を特徴とする中枢神経系に影響を与える神経炎症性疾患である。この病気の予後は治療の進歩があっても変動し、若者の神経障害の最も一般的な原因の1つである。MSの診断は、磁気共鳴画像法(MRI)2、3による特徴的な病変の臨床症状および可視化の基準に基づいている。

異なるアミロイドPETトレーサー(11C-PIB、18 F-florbetaben、18 F-florbetapir、18 F-flutemetamol)を使用してミエリンを定量化し、疾患の進行および治療応答に関する重要な情報を提供し、神経炎症の干渉なしに脱髄および再髄鞘化プロセスの同定を可能にする。 アミロイドPET画像投射は、活動患者8における早期白質損傷によって説明することができる非活動患者と比較して、活性MS患者におけるトレーサー取り込みの減少を示した。下方アミロイドトレーサー取り込みもフォローアップ研究における認知機能の低下と関連しており、この技術が疾患および臨床転帰の病態生理を研究するための貴重なツールであることを示す9。
リゾレシチン(LPC)ラットモデルは、多発性硬化症の化学的誘導モデルであり、そこで注入された毒素、LPCは、炎症の増加をもたらすマクロファージの高応答を誘導し、その結果、脱髄10、11。脱髄は約4週間で急速に逆転し、げっ歯類の脱髄および再髄鞘化プロセスを評価するための良いモデルになります。このモデルは、PETイメージングを用いて既に評価されており、結果は良好であり、事後分析エッセイ12と相関している。
