直径12nmの金ナノ粒子(Auナノ粒子)を、テトラクロロイ酸の溶液を3.0g(3.7mmol)に速やかに注入して合成した 3.6 mL)のオレイラミン(技術グレード)と3.0mLのトルエンを、トルエン147mLのオレイラミンの5.1g(6.4 mmol,8.7 mL)の沸騰溶液にする。反応液を沸騰させ、2時間混合しながら、反応混合物の色は透明から薄い黄色、薄いピンク色に変化し、次いでゆっくりと濃赤色に変化した。その後、熱を消し、溶液を1時間室温まで徐々に冷却した。その後、金ナノ粒子を回収し、遠心分離機を用いて溶液から分離し、3回洗浄した。トルエンの10 mL部分に金ナノ粒子をボルテックスして分散させ、その後40mLのメタノール部分を加えて遠心分離機で回転させることで金ナノ粒子を沈殿させる。その後、溶液をデカンデーして、残りの副産物および未反応の出発物質を除去した。真空環境で金ナノ粒子を乾燥させることで、固体の黒色ペレットが生成されます。後で使用するために長期間(最長1年)保存し、トルエンなどの有機溶媒に再溶解することができます。

この金ナノ粒子合成プロセスの目的は、直径約12 nm.in の金ナノ粒子を作り出す事です。この金ナノ粒子合成プロセスは、平松博樹とフランクE.Osterloh.化学薬品、テトラクロローウ酸、オレイラミン、トルエン、メタノールの合成と精製に必要な研究記事に記載されているプロセスの修正版です。窒素グローブボックスに化学反応ガラス製品と供給物をセットアップし、洗浄を開始します。磁気攪拌棒がガラス反応容器の中にあることを確認してください。約200mlのトルエンをガラス反応容器に注ぎます。トルエンを付したガラス反応容器を攪拌加熱マントルに置き、コンデンサーチューブを反応容器に送ります。ゆっくりと水をオンにして監視し、コンデンサーチューブに静かに流れ込んでいくようにします。必要に応じて、水バルブをわずかに開閉して、水の流れを調整します。水がコンデンサーチューブの底部の入口ポートを通って、凝縮器チューブを上に、そしてコンデンサーチューブの上の出口ポートを流れるようにします。グローブボックスに新鮮な窒素を継続的に流し込み、グローブボックスをパージします。グローブボックスにわずかな真空を引っ張って、グローブボックスを連続的に換気します。攪拌および加熱マントルに磁気攪拌機でトルエンを加熱し、攪拌を開始します。トルエンが穏やかな沸騰に近づくのを許可します。トルエンの引火点に近づいたり超えたりしないでください。沸騰し始めたら火を弱めます。トルエンを沸騰させ、30分間蒸発させて反応ガラス製品をきれいにします。蒸発したトルエンは凝縮器の管で冷却し、凝縮し、反応容器に戻って滴下する。30分後、ヒーターをオフにし、トルエンが蒸発しなくなるまで数分間冷却します。トルエンが冷却した後、コンデンサーチューブを持ち上げ、クランプで標準を使用して反応容器の上に吊り下げます。反応容器からトルエンを400に注ぎ ml.beaker.Be 誤って磁気攪拌棒を注ぎ出さないよう注意してください。反応容器を加熱し、マントルをかき混ぜる上に戻します。400 ml.glassビーカーでトルエンを回してビーカーをきれいにします。汚れた/使用されたトルエンを可燃性廃棄物ボトルに注ぎます。新鮮なトルエンでビーカーを再び清掃し、使用したトルエンを脆弱な廃棄物ボトルに捨てます。8.7mlで147mlまたは約150mlのトルエン、または約9mlのオレイラミンの沸騰液を作る。400 ml.グラスビーカーを使用して、147 mlのトルエンを測定します。147mlのトルエンをガラスビーカーから反応容器に注ぎます。注意:オレイラミンは有毒で腐食性であるため、注意して処理してください。オレイラミンをこぼさない 5 ml.小ガラスの卒業したシリンダーを慎重に測定し、8.7 ml のオレイラミンを注ぐ。例えば、最初の4 ml.on4 ml、次に4.7 ml.慎重にガラスの卒業したシリンダーから反応容器にオレイラミンを注ぎます。ガラスコンデンサーチューブをガラス反応容器に下ろします。水が冷却、凝縮、およびトルエンとオレイラミン蒸気を収集するために凝縮管の外チャンバーを通って流れていることを確認します。熱と溶液をかき混ぜ、溶液が穏やかな沸騰に近づくできるようにします。注射液の調製を開始する:150 mg.のテトラクロロウリン酸、3.6 mlのオレイラミン、および3mlのトルエン。テトラクロロウリン酸を空気や湿気から保護している実験室フィルムまたはシールを取り除きます。テトラクロロウ酸は、水、水分、湿度に非常に敏感です。テトラクロロウリン酸粉末を空気と水に暴露しないようにあらゆる努力をする必要があります。テトラクロロウリン酸容器は、窒素グローブボックスに開いて保管する必要があります。金属スパチュラを使用して、容器からテトラクロロウ酸粉末をマイクロバランス上のガラスバイアルに堆積し、150mgのテトラクロロウ酸粉末の測定重量にします。注意:オレイラミンは有毒で腐食性であるため、注意深く取り扱ってください。5 ml.小ガラスの段階的なシリンダーを使用して、3.6 ml.のオレイラミンを測定します。5ml.小ガラスの段階的なシリンダーから3.6mlのオレイラミンをテトラクロローリン酸なしで20mlガラスバイアルに慎重に注ぎます。慎重に注ぎ、5 ml.小ガラスの卒業シリンダーにトルエンの3 mlを測定します。5ml.小ガラスの段階的なシリンダーから3mlのトルエンを慎重にオレイラミンと共に20mlガラスバイアルに注ぎます。PTFEで裏打ちされたキャップを、内部にオレイラミンとトルエンを入れた20 ml.グラスバイアルに戻します。閉じたガラスバイアルを振って渦巻き、オレーランミンとトルエン溶液を混ぜ合わせます。150mgのテトラクロロウリン酸粉末を、オレイラミンとトルエン溶液でガラスバイアルに慎重に注ぎます。PTFEで裏打ちされたキャップをガラスバイアルにねじ込みます。テトラクロロウ酸、オレイラミン、トルエンを使用して閉じたガラスバイアルを振って旋回し、溶液を混ぜ合わせます。溶液を振り続け、完全に混合されていることを確認します。テトラクロロウリン酸、オレイラミンおよびトルエン溶液は、濃い赤色に変わるはずです。水が凝縮管の底部にゆっくりと流れ込み、凝縮器管の上部に流れ出していることを確認してください。必要に応じて、水バルブを慎重に開閉して、水の流れを調整します。反応容器内のオレイラミンおよびトルエン溶液が穏やかな沸騰していることを確認し、トルエンとオレイラミンが凝縮管に蒸発することを確認します。磁気スターラーがオンになっていることを確認します。クランプ付きのスタンドを使用して、コンデンサーチューブを反応容器の上に上げて、ガラス製品を支えます。ゴムバルブを使用する前に、バルブとガラスパイプハットでゴムバルブを操作する方法を知っていることを確認してください。ゴムバルブを絞りながら上部バルブを押して、ゴムバルブを収縮します。長い段階のガラスピペットの先端をテトラクロローリン酸、オレイラミン、トルエン注射液を用いて20ml.グラスバイアルに慎重に入れます。ゴム球の下側バルブを軽く押し、テトラクロロウリン酸、オレイラミン、トルエン注入液をガラスピペットにゆっくりと引き込みます。ガラスピペットの先端を反応容器の開口部に慎重に入れ、迅速にテトラクロロウリン酸、オレイラミンおよびトルエン溶液を反応容器に注入する。金ナノ粒子が核化して成長し始めると、色は約1分以内に赤から黄色に変わるはずです。コンデンサーチューブを反応容器に下ろします。溶液を2時間穏やかに沸騰させると熱します。金ナノ粒子が大きくなるにつれて、数分間にわたって、色は白から黄色、ピンク、赤に変わるはずです。数分後、溶液はピンクまたは明るい赤色に変わります。数分後、ソリューションはさらに赤くなります。溶液は2時間煮るほど暗い赤くなり続けます。2時間加熱した後、ヒーターをオフにします。金ナノ粒子溶液を1時間自然に冷却します。ヒーターがオフになっていて、溶液が冷却されていることを確認します。コンデンサーチューブを通して水を流すのを止めます。メタノールは、金ナノ粒子を洗浄し、未反応の化学試薬や副産物を除去するために使用されます。50 ml.円錐形遠心分離チューブのそれぞれに35mlのメタノールを注ぎます。その後、メタノールを用いた50ml.円錐遠心管のそれぞれに12mlの金ナノ粒子溶液を注ぎます。各遠心チューブに金ナノ粒子溶液を注ぎ込みながら、誤って磁気攪拌棒を注ぎ出さないよう注意してください。35 ml のメタノールを含む 50 ml.円錐型遠心分離管のそれぞれに 12 ml の金ナノ粒子溶液を注ぎ込んだ後、各遠心管は約 47 ml.の溶液を持つ必要があります。残りの金ナノ粒子溶液を遠心管の間に均等に分配する。キャップを50 ml.遠心分離チューブにねじ込んで閉じ、キャップを締めます。50 ml.円錐形遠心分離管を、黄金のナノ粒子溶液とメタノールを窒素グローブボックスから取り出します。遠心管は、同量の金ナノ粒子溶液を有し、均等に重み付けされ、バランスを取る必要があります。50 ml.遠心分離管のうち6本を金ナノ粒子を入れ、遠心分離機に入れる。遠心分離機の蓋を閉じ、金ナノ粒子を回転させる設定を入力します。RCF:1000.タイマー:5分遠心分離機の回転を開始します。金ナノ粒子を含む最初の6本の遠心管を回転させた後、遠心分離機からそっと取り出します。金ナノ粒子ペレットを乱さないで、チューブをラックに入れないように注意してください。遠心力は溶液中の金ナノ粒子を引き下げ、メタノールとトルエンからそれらを分離します。金ナノ粒子は、各遠心分離管の底部のペレットに沈殿します。金ナノ粒子を含む最後の6本の遠心管を遠心分離機に入れる。遠心分離機の蓋を閉じ、金ナノ粒子を回転させる設定を入力します。RCF:1000.タイマー:5分回転を開始します。遠心管の回転が終わったら、遠心分離機からそっと取り出し、チューブラックに入れ。慎重に煙のフードにすべての遠心管を持ち歩きます。各遠心分離チューブから溶液を可燃性廃棄物にゆっくりと静かにダンプします。邪魔をしないように注意し、チューブの底に金ナノ粒子ペレットを注ぎ込まないように注意してください。最初のメタノールリンスサイクルが完了しました。各遠心管に10mlのトルエンを注ぎ込み、第2のメタノールリンスサイクルを開始します。ボルテックス各遠心管は、10 ml.トルエン溶液全体に金ナノ粒子を再懸濁し、分散させる。遠心分離管の底面を確認して、黒い残留物が再懸濁されていることを確認します。各遠心管に14mlのメタノールを注ぎます。10 mlのトルエンが各遠心チューブに既に入っている場合、チューブあたり50mlの溶液が存在します。キャップを50 ml.遠心分離チューブに戻し、キャップがしっかりとつかっていることを確認します。遠心分離機の遠心管を回転させて、各チューブの底部にあるペレットに金ナノ粒子を集める。以前と同じ遠心分離機の設定を使用します。メタノールとトルエンを前のように可燃性廃棄物に慎重に注ぎ出します。第2のメタノールリンスサイクルが完了しました。3回目および最後のリンスサイクルでは、トルエンでのボルテックス、メタノールでの洗浄、遠心分離、慎重に溶媒を注ぐ場合と同じプロセスに従ってください。最後のトルエンとメタノールリンス溶液を注ぎ出した後、金ナノ粒子を乾燥させる必要があります。推奨される手順は、金ナノ粒子を真空乾燥させることです。50 ml.遠心分離チューブのキャップを緩めて、チューブがまだ覆われているようにしますが、溶剤は蒸発してチューブ内から脱出することができます。真空負荷ロック内に金ナノ粒子を持つチューブのラックを置きます。外側の負荷ロックドアを閉めてシールし、バルブを真空ポンプに開けて、負荷ロックの真空を引っ張り始めます。真空圧計の中央部に適度な真空圧力が届くまで真空をポンプします。黄金のナノ粒子を、数分間、約半分のゲージの中程度の真空圧でロードロックに残します。低圧までポンプダウンせず、真空中に長時間放置したり、リガンドが剥離したりすること。金ナノ粒子が数分間真空下にあった後、金ナノ粒子を乾燥させ、残りの溶媒を蒸発させ、負荷のロックを窒素でパージして、負荷のロックが大気圧に達するまで窒素を除去します。50 ml.遠心分離チューブを金ナノ粒子と共にロードロックから取り出し、ヒュームフード内の金色ナノ粒子の乾燥を検査します。金ナノ粒子ペレットが乾燥した後、キャップを遠心管にねじ込みます。密閉キャップの周りにラボフィルムをラップし、内部の金色ナノ粒子ペレットで遠心管を密封します。遠心管に金ナノ粒子のラベルを付け、2の内部に配置します。
