Het doel van dit protocol is om het genereren van pancreasplakken en de procedures die nodig zijn om de plakjes te gebruiken in functionele en immunologische studies te verklaren. Er zijn echter verschillende kritieke stappen die essentieel zijn voor het weefsel om levensvatbaar en nuttig te blijven tijdens de beschreven experimentprotocollen. Betrokkenheid van de alvleesklier is pathognomonisch voor ziekten zoals pancreatitis, T1D en T2D1,2,3. De studie van de functie in geïsoleerde eilandjes omvat meestal het verwijderen van de eilandjes uit hun omgeving4. De methode voor het snijden van levend pancreasweefsel is ontwikkeld om de studie van pancreasweefsel mogelijk te maken met behoud van intacte micro-omgevingen van eilandjes en het vermijden van het gebruik van stressvolle isolatieprocedures voor eilandjes5,6,7. Pancreasweefselplakken van menselijk donorweefsel zijn met succes gebruikt om T1D te bestuderen en hebben processen van bètacelverlies en disfunctie aangetoond naast immuuncelinfiltratie8,9,10,11,12,13. De methode voor het snijden van levend pancreasweefsel kan worden toegepast op zowel muis- als menselijk pancreasweefsel5,6,8. Menselijke pancreasweefselplakken uit orgaandonorweefsels worden verkregen door een samenwerking met het Network for Pancreatic Organ Donors with Diabetes (nPOD). Muissegmenten kunnen worden gegenereerd uit verschillende muizenstammen.
Dit protocol zal zich richten op niet-obese diabetische recombinatie activerende gen-1-null (NOD. Rag1-/-) en T-celreceptor transgeen (AI4) (NOD. Vod1-/-. AI4 α/β) muisstammen. KNIKKEN. Rag1-/- muizen zijn niet in staat om T- en B-cellen te ontwikkelen als gevolg van een verstoring in het recombinatie-activerende gen 1 (Rag1)14. KNIKKEN. Vod1-/-. observing islet function and islet-immune cell interactions in live pancreatic tissue slices

Levende pancreasweefselplakken maken de studie van eilandjesfysiologie en -functie mogelijk in de context van een intacte micro-omgeving van eilandjes. Plakjes worden bereid uit levend menselijk en muis pancreasweefsel ingebed in agarose en gesneden met behulp van een vibratome. Deze methode zorgt ervoor dat het weefsel levensvatbaarheid en functie behoudt, naast het behoud van onderliggende pathologieën zoals type 1 (T1D) en type 2 diabetes (T2D). De slice-methode maakt nieuwe richtingen mogelijk in de studie van de pancreas door het onderhoud van de complexe structuren en verschillende intercellulaire interacties die de endocriene en exocriene weefsels van de pancreas omvatten. Dit protocol demonstreert hoe kleuring en time-lapse microscopie van levende endogene immuuncellen in pancreasplakken kunnen worden uitgevoerd, samen met beoordelingen van de eilandjesfysiologie. Verder kan deze benadering worden verfijnd om immuuncelpopulaties te onderscheiden die specifiek zijn voor eilandcelantigenen met behulp van belangrijke histocompatibiliteit complex-multimeerreagentia.

Dit protocol levert levende pancreasweefselplakken op die geschikt zijn voor zowel functionaliteitsstudies als immuuncelopnamen. Ons protocol zal helpen bij het beantwoorden van de impact die immuuncelinteracties hebben op de eilandjesfunctionaliteit. De methode voor het snijden van levend pancreasweefsel maakt het mogelijk om de fysiologie en functie van eilandjes te bestuderen met behoud van de weefsels die ten grondslag liggen aan pathologieën en de inheemse micro-omgeving. Deze methode zou inzicht kunnen geven in de ziekteprocessen van pancreasziekten, zoals pancreatitis, diabetes type I en diabetes type II.Plaats om te beginnen de blokken op de monsterhouder zodanig dat ze de bladbreedte niet overschrijden, zodat het blad zo min mogelijk naar voren kan bewegen wanneer de trilling langzaam beweegt. Breng een lijn superlijm aan op de monsterhouder, maak een dunne laag met behulp van het uiteinde van de lijmdispenser en draai vervolgens de tissueblokken op de lijm zodat de kant dicht bij het weefsel naar boven is gericht. Duw de blokken voorzichtig naar beneden en laat de lijm drie minuten drogen.Bevestig vervolgens de plaat met een schroef aan de trilpan en pas de hoogte en afstand van het blad aan, zodat het blad over de lengte van blokken beweegt en er nauwelijks boven komt. Duw de blokken voorzichtig met de tang om te controleren of de lijm is opgedroogd en vul vervolgens de vibratome-lade met een gekoelde extracellulaire oplossing totdat het mes is bedekt. Stel de trilling in om plakjes van 120 micrometer dikte te maken en start het, observeer vervolgens de plakjes die van de weefselblokken komen.Til de plakjes op wanneer ze van het blok drijven door er een verfkwast of tang onder te plaatsen en plaats de plakjes in Krebs-Ringer bicarbonaatbuffer met drie millimolar D-glucose en trypsineremmer. Leg de platen met de plakjes op een wip en incubeer bij kamertemperatuur gedurende een uur bij 25 RPM. Als de plakjes langer bewaard moeten worden, plaats de plakjes dan in 15 milliliter plakcultuurmedium en plaats het bord in een couveuse.Incubeer de bereide plakjes bij 37 graden Celsius voor studies op dezelfde dag of breng de plakjes die 's nachts bij 24 graden Celsius zijn gekweekt over naar 37 graden Celsius gedurende ten minste één uur vóór het experiment. Schakel de microscoop ten minste een uur voor de opname in en breng de incubator van de podiumtop in evenwicht tot 37 graden Celsius. Bevestig de petrischaal met de glazen bodem met het plakje op het podium.Richt de microscoop door het 10X-objectief in de brightfield-modus in te stellen en lokaliseer de eilandjes met behulp van de bright field-modus door te zoeken naar oranjebruin gekleurde ovalen in de slice. Schakel de microscoop over naar confocale beeldvorming door op de CS-knop op de touchscreencontroller van de microscoop te drukken. Als u de eilandjes wilt bekijken met behulp van reflectie, schakelt u de 638-laserdetector en PMT-detector in.Stel het laservermogen in tussen één en 2% en schakel de inkepingsfilters uit. Gebruik de 405 nanometer laser en PMT detector om de nucleïnezuurvlek te observeren. De hybride detector voor de CD8 antilichaamdetectie en de 488 nanometer laser en hybride detector om het insulinetetrameer te bekijken.Centreer het interessante eilandje in het gezichtsveld met behulp van de X- en Y-knoppen van de podiumcontroller. Zodra een interessant eilandje zich bevindt, schakelt u over naar het 20X-objectief en zoomt u in, zodat het eilandje het grootste deel van het frame in beslag neemt. Neem een z-stack van het eilandje en zoek vervolgens de beste optische secties waar de cellen leven en eventuele omliggende immuuncellen in focus zijn.Stel de microscoop in om op te nemen in XYZT-modus en optimaliseer de instellingen om elke 20 minuten een z-stack van de geselecteerde stappen op te nemen gedurende een periode van enkele uren. De eilandjes in een plak pancreasweefsel werden gevisualiseerd met behulp van brightfield en gereflecteerde lichtmicroscopie. De insuline in de eilandjes verhoogt de granulariteit en veroorzaakt absorptie van grote hoeveelheden gereflecteerd licht, waardoor de zichtbaarheid van de eilandjes toeneemt.De levensvatbaarheid van een levend menselijk pancreasweefsel werd beoordeeld door middel van kleuring. De levende cellen zijn aangegeven in groen en de dode cellen in blauw. Een andere positieve indicator voor levensvatbaarheid is waarneembare basale activiteit.Wanneer een levende pancreasschijf werd geladen met een celdoorlatende calciumindicator, werd glucosestimulatie waargenomen in zowel muis- als menselijke pancreasplakken. De fluorescentie van individuele cellen tijdens glucosestimulatie werd ook gekwantificeerd. Intacte en zieke eilandjes werden waargenomen met behulp van dithizone-kleuring en gereflecteerde lichtmicroscopie.De zieke eilandjes beginnen granulariteit te verliezen als gevolg van immuuncelinfiltratie en celdood. Immuuncelpopulaties kunnen worden geïdentificeerd met behulp van CD8-antilichamen en insulinetetrameerkleuring. Co-kleuring geeft aan dat de cellen effector T-cellen zijn die specifiek gericht zijn op het insulineantigeen.Het meest kritieke om te onthouden in deze procedure is om de plakjes in oplossingen proteaseremmer te allen tijde te houden. Na deze procedure kunnen tal van functionaliteiten in immuuncelexperimenten worden uitgevoerd, waardoor het effect van immuuncelinteracties en eilandjesfunctie kan worden bestudeerd.
