התפשטות גרורתית למוח היא ביטוי שכיח והרסני של סוגים רבים של סרטן. בארצות הברית לבדה, כ-200,000 חולים מאובחנים עם גרורות במוח מדי שנה. הושגה התקדמות משמעותית בשיפור תוצאות ההישרדות של חולות עם סרטן שד ראשוני וממאירויות סיסטמיות; עם זאת, הפרוגנוזה העגומה עבור חולים עם גרורות קליניות במוח מדגישה את הצורך הדחוף לפתח סוכנים טיפוליים חדשניים ואסטרטגיות נגד מחלה קטלנית זו. היעדר מודלים ניסיוניים מתאימים היה אחד המכשולים העיקריים המעכבים את התקדמות ההבנה שלנו של גרורות מוחיות, ביולוגיה וטיפול. במאמר זה אנו מתארים מודל עכברי של גרורות במוח שנוצרו באמצעות הזרקת וריד זנב של קו תאים אנדוגני מוגבר HER2 שמקורו בסרטן שד דלקתי (IBC), צורה נדירה ואגרסיבית של סרטן השד. התאים סומנו בלוציפראז גחלילית ובחלבון פלואורסצנטי ירוק כדי לנטר גרורות במוח, וכומתו עומס גרורתי על ידי דימות ביולומינסצנטי, סטריאומיקרוסקופיה פלואורסצנטית והערכה היסטולוגית. עכברים מפתחים גרורות במוח באופן יציב ועקבי, מה שמאפשר חקירה של מתווכים מרכזיים בתהליך הגרורתי ופיתוח בדיקות פרה-קליניות של אסטרטגיות טיפול חדשות.
