Ved specifikt at målrette den metriske rumlige informationsproces giver denne metriske test et nødvendigt værktøj til at forstå hukommelsesmangel efter TBI. Forekomsten af neurologiske underskud som opmærksomhed, udøvende funktion og visse hukommelsesunderskud efter moderat traumatisk hjerneskade (TBI) er mere end 50 procent1,2,3,4,5,6,7,8. TBI kan føre til alvorlige funktionsnedsættelser i rumlige kortsigtet, langsigtet, og arbejdshukommelse9. Disse hukommelsesnedsættelser er blevet observeret i gnavermodeller af TBI. Gnavermodeller har gjort det muligt at udvikle teknikker til at teste hukommelsen, hvilket giver mulighed for dybere undersøgelser af virkningen af TBI på hukommelsesbehandling i neurale hukommelsessystemer.
To tests, der er relateret til henholdsvis topologisk og metrisk fysisk informationsbehandling, hjælper med måling af rumlig arbejdshukommelse (SWM). Den topologiske test afhænger af ændring af størrelsen af miljøområdet eller relaterede rum af forbindelse eller kabinet omkring et objekt, mens den metriske test vurderer ændringer i vinkler eller afstand mellem objekter10,11. Goodrich-Hunsaker et al. tilpassede først den menneskelige topologiske test for rotter10 og anvendte den metriske opgave at adskille rollerne for parietal cortex (PC) og dorsal hippocampus i rumlig informationsbehandling11. Tilsvarende gurkoff og kolleger evalueret metriske, topologiske og tidsmæssige bestilling hukommelse opgaver efter lateral væske slagtøj skade9. Der er en sammenhæng mellem skader på visse områder af hjernen og svækkelse af metriske eller topologiske hukommelse. Det er blevet foreslået, at metriske hukommelsesnedsættelser er relateret til læsioner i bilateral dorsal dentat gyrus og cornu ammonis (CA) sub-region CA3 af hippocampus, og at topologisk hukommelse svækkelse er relateret til bilaterale parietale cortex læsioner10,12.
Formålet med denne protokol er at vurdere det rumlige hukommelsesunderskud i en rottepopulation via en metriske opgave. Denne metode er et velegnet alternativ til at undersøge mekanismer af SWM efter hjerneskade, og dens fordele omfatter den relative lethed af gennemførelsen, høj følsomhed, lave omkostninger ved reproducerbarhed, muligheden for dynamisk observation og et lavt stressmiljø. Sammenlignet med andre adfærdsmæssige opgaver som Barnes labyrint13,14, Morris vand navigation opgave15,16,17,eller rumlige labyrint opgaver18,19, denne metriske test er mindre kompliceret. På grund af dens lette implementering kræver den metriske test en kortere og mindre stressende træningsperiode og finder sted over kun 2 dage9:1 dag til beboelse og 1 dag til opgaven. Desuden er vores foreslåede test lettere at udføre end andre lav stresstest, såsom den nye objektgenkendelse (NOR) opgave, og kræver ikke den ekstra dag med habituation20.
Dette papir giver en enkel model til evaluering af SWM efter hjerneskade. Denne vurdering af swm efter TBI kan bidrage til en mere omfattende undersøgelse af dens patofysiologi.

Svækkelser til sensorisk, kortsigtet og langvarig hukommelse er almindelige bivirkninger efter traumatisk hjerneskade (TBI). På grund af de etiske begrænsninger af humane undersøgelser giver dyremodeller egnede alternativer til testbehandlingsmetoder og til at studere mekanismerne og relaterede komplikationer af tilstanden. Eksperimentelle gnavermodeller har historisk set været de mest anvendte på grund af deres tilgængelighed, lave omkostninger, reproducerbarhed og validerede tilgange. En metriske test, som tester evnen til at huske placeringen af to objekter på forskellige afstande og vinkler fra hinanden, er en teknik til at studere svækkelse i rumlig arbejdshukommelse (SWM) efter TBI. De betydelige fordele ved metriske opgaver omfatter muligheden for dynamisk observation, lave omkostninger, reproducerbarhed, relativ brugervenlighed og miljø med lav stress. Her præsenterer vi en metriske testprotokol til måling af værdiforringelse af SWM hos voksne rotter efter TBI. Denne test giver en mulig måde at evaluere fysiologi og patofysiologi af hjernens funktion mere effektivt.

Betydningen af sammenligninger mellem grupper blev bestemt ved hjælp af Mann-Whitney-testen. Statistisk signifikans af resultaterne blev anset for at være på P < 0,05, mens statistisk høj relevans blev målt til P < 0,01.

Traumatisk hjerneskade er almindeligt forbundet med hukommelsesnedsættelse. Her præsenterer vi en protokol til vurdering af spacial arbejdshukommelse efter traumatisk hjerneskade via en metriske opgave. De vigtigste fordele ved metriske opgaver omfatter muligheden for dynamisk observation, billig reproducerbarhed, relativ nem implementering og miljø med lav stress.For at evaluere den neurologiske sværhedsgrad score efter parasagittal væske slagtøj skade, placere den voksne Sprague Dawley rotte i midten af en 50 centimeter diameter cirkel, tre gange og overvåge dens evne til at forlade cirklen. For at teste for tab af refleks, placere dyret på ryggen i håndfladen og give en score på en, hvis rotten er i stand til at rette sig selv. For at teste for hemiplegi skal du evaluere rottens manglende evne til at modstå tvungen positionering.For at teste den refleksive bøjning af bagbenet skal dyret hæves ved halen. For at teste evnen til at gå lige, placere rotten på gulvet. For at bedømme tabet af at søge adfærd i prostration, placere rotten i et nyt miljø og observere, om rotten bevæger sine whiskers, sniffs eller løber.Derefter teste lemmer reflekser for placering på venstre og højre forben efterfulgt af venstre og højre bagben. For at teste balancen skal du udføre en strålebalanceringsopgave ved at placere rotten på en 1,5 centimeter bred stråle for en 20, 40 og større end anden session. Kør derefter en strålevandringstest ved hjælp af tre bjælker af forskellig bredde.Før du udfører den metriske opgave, skal du placere en diameter på 200 centimeter, en centimeter tyk, sort cirkulær platform på et 80 centimeter højt bord og placere to objekter i midten af platformen, 68 centimeter fra hinanden. Installer et kamera med den nødvendige computersoftware til hentning, lagring og behandling af data i en højde af 290 centimeter fra gulvet. En dag før analysen placerer rotten på platformen i 10 minutter for at vænne dyret til det nye miljø.På dagen for opgaven skal du placere rotten på enden af platformen, lige langt fra objekterne i en 15 minutters habituationsperiode og begynde at optage. I slutningen af den eksperimentelle habituationsperiode overføres rotten til et individuelt bur i fem minutter og rengør platformen med fem til ti procent alkohol. Reducer afstanden mellem objekterne til 34 centimeter, og placer derefter rotten på platformen i fem minutter, mens den registrerer dens udforskningsaktivitet.Rengør platformen igen, før du fortsætter med det næste dyr. Før du analyserer videofilerne, skal du indsætte softwarehardwarenøglen, starte videosporingssoftwaren og åbne den forudindstillede skabelon. Når du har kontrolleret, duplikeret og omdøbt indstillingerne for testkontrol og registrering af arenaen i hovedfeltet, skal du højreklikke på musen for at vælge grabbaggrund.Vælg placeringen af den videofil, der skal bruges som baggrundsbillede, i menuen Gennemse. Hent billedet, og markér de undersøgte områder og zoner for at kalibrere billedet til analyse. Udfør den samme opsætning for indstillingerne for prøvekontrol og registrering.Når du har downloadet listen over videofiler til analyse fra prøvelisten, skal du tilføje videoerne og angive placeringen med de nødvendige indstillinger. Vælg derefter anskaffelse, og start prøveversionen, og eksporter alle data til et regneark. Kontrolgruppen viser ingen neurologiske underskud, 48 timer efter den falske skade.Efter TBI er neurologiske underskud betydeligt større hos både mandlige og kvindelige skadede dyr. Under den metriske opgave er objektudforskningstiden betydeligt kortere for de mandlige TBI-rotter end for de han-sham-opererede rotter. Det samme mønster ses for de kvindelige TBI og kvindelige sham-opererede rotter.Efter den metriske opgave kan vi udføre andre metoder til hukommelsesvurdering, såsom Barnes labyrinten og topologiske og vand labyrint opgaver. Den metriske opgave giver også mulighed for yderligere forskning i hukommelsesnedsættelse i sammenlignelige modeller af neurologiske skader, såsom modeller af diffus axonal hjerneskade og slagtilfælde.
