Skeletspieren zijn een van de meest voorkomende weefsels in het menselijk lichaam en ondersteunen belangrijke lichaamsfuncties zoals voortbeweging, warmtehomeostase enmetabolisme 1. Historisch gezien zijn diermodellen en tweedimensionale (2D) celkweeksystemen gebruikt om biologische processen en ziektepathogenese te bestuderen, evenals voor het testen van farmacologische verbindingen bij de behandeling van skeletspierziekten2,3. Hoewel diermodellen onze kennis van skeletspieren in gezondheid en ziekte sterk hebben verbeterd, is hun translationele impact belemmerd door hoge kosten, ethische overwegingen en verschillen tussen soorten2,4. Bij het wenden tot menselijke celgebaseerde systemen om skeletspieren te bestuderen, zijn 2D-celkweeksystemen gunstig vanwege hun eenvoud. Er is echter een beperking. Dit formaat slaagt er vaak niet in om de cel-cel en cel-extracellulaire matrixinteracties samen te vatten die van nature in het lichaam voorkomen5,6. In de afgelopen jaren zijn driedimensionale (3D) skeletspiermodellen naar voren gekomen als een krachtig alternatief voor hele diermodellen en conventionele 2D-kweeksystemen door de modellering van fysiologisch en pathologisch relevante processen ex vivo7,8mogelijk temaken. Inderdaad, een overvloed aan studies hebben strategieën gerapporteerd om menselijke skeletspieren te modelleren in een bioartificiaal 3D-cultuurformaat1. Een beperking voor veel van deze studies is dat actieve kracht wordt gekwantificeerd na het verwijderen van de spierweefsels van de kweekplatforms en de hechting aan een krachtomvormer, die destructief is en daarom beperkt is tot het dienen als eindpunttest9,10,11,12,13,14,15,16,17,18 ,19,20,21. Dit protocol beschrijft een gedetailleerde methode om menselijke spiermicrotissues (hMMCT's) te fabriceren in het skeletspieren (Myo) microTissue Array deviCe To Investigate forCe (MyoTACTIC) platform; een 96 well plate apparaat dat de bulkproductie van 3D skeletspier microtissuesondersteunt 30. De MyoTACTIC-plaatfabricagemethode maakt het mogelijk om in één gietstap een 96-put polydimethylsiloxaan (PDMS) kweekplaat en alle bijbehorende putkenmerken te genereren, waarbij elke put een relatief klein aantal cellen nodig heeft voor microtissuevorming. Microtissues gevormd binnen MyoTACTIC bevatten uitgelijnde, gestreepte en multinucleated myotubes die reproduceerbaar zijn van goed naar put van het apparaat, en bij rijping kunnen reageren op chemische en elektrische stimuli in situ30. Hierin worden de techniek om een PDMS MyoTACTIC-cultuurplaatapparaat te vervaardigen uit een replica van polyurethaan (PU), een geoptimaliseerde methode om onsterfelijke menselijke myogene voorlopercellen te implementeren om hMMT's te fabriceren, en de functionele beoordeling van gemanipuleerde hMMT-krachtgeneratie en calciumbehandelingseigenschappen geschetst en besproken.

assessing functional metrics of skeletal muscle health in human skeletal muscle microtissues

Driedimensionale (3D) in vitro modellen van skeletspieren zijn een waardevolle vooruitgang in biomedisch onderzoek omdat ze de mogelijkheid bieden om skeletspierreformatie en -functie te bestuderen in een schaalbaar formaat dat vatbaar is voor experimentele manipulaties. 3D-spierkweeksystemen zijn wenselijk omdat ze wetenschappers in staat stellen om skeletspieren ex vivo te bestuderen in de context van menselijke cellen. 3D in vitro modellen bootsen aspecten van de inheemse weefselstructuur van volwassen skeletspieren na. Hun universele toepassing wordt echter beperkt door de beschikbaarheid van platforms die eenvoudig te fabriceren, kosten- en gebruiksvriendelijk zijn en relatief grote hoeveelheden menselijke skeletspierweefsels opleveren. Bovendien, aangezien skeletspieren een belangrijke functionele rol spelen die in de loop van de tijd in veel ziektetoestanden wordt aangetast, is een experimenteel platform voor microtissue-studies het meest praktisch wanneer minimaal invasieve calciumtransiënte en contractiele krachtmetingen direct binnen het platform zelf kunnen worden uitgevoerd. In dit protocol wordt de fabricage van een 96-well platform dat bekend staat als 'MyoTACTIC', en de massale productie van 3D menselijke skeletspiermicrotissues (hMMTs) beschreven. Bovendien worden de methoden voor een minimaal invasieve toepassing van elektrische stimulatie gerapporteerd die herhaalde metingen van de skeletspierkracht en calciumbehandeling van elke microtissue in de loop van de tijd mogelijk maakt.

Hierin worden methoden beschreven om een 96-well PDMS-gebaseerd MyoTACTIC-kweekplatform uit een PU-mal te gieten, om arrays van hMMT-replicaweefsels te fabriceren en om twee aspecten van hMMT-functie te analyseren binnen de generatie van kweekapparatuurkracht en calciumbehandeling. Ons protocol beschrijft gedetailleerde methoden om een 3D-microtissuecultuur van menselijke skeletspieren te fabriceren en te analyseren. Spiermicrotissues kunnen worden toegepast op studies van elementaire spierbiologie, ziektemodellering of voor het testen van kandidaat-moleculen. Het belangrijkste voordeel van deze techniek is dat het in-situ beoordeling van contractiele kracht en calciumtransiënten mogelijk maakt.Daarom kunt u longitudinale studies van de spierweefselfunctie uitvoeren. De procedure wordt gedemonstreerd door Brennen Musgrave en Heta Lad, afgestudeerde studenten van mijn laboratorium. Plaats twee tot drie uur voor het zaaien van de cel een gedeelte van zes goed milde tactiekplaten in een celkweekschaal van 10 centimeter.Bereid elke individuele myotactische cultuur goed voor door 100 microliter van een 5% pluronische F-127-oplossing toe te voegen. Gebruik het deksel om de putjes af te dekken en breng een paraffinefilm aan om de schaal af te sluiten. Centrifugeer de schotel gedurende één minuut op 1.550 maal G in een centrifuge die is uitgerust met een platenspinneradapter.Bewaar de kweekschaal bij vier graden Celsius totdat de cellen klaar zijn om te zaaien. Ontdooi dan langzaam 150 microliter aliquot keldermembraanextract en 110 microliter aliquot trombine op ijs in de kweekkap. Werk in de celkweekkap en voeg 700 microliter 0,9% zoutoplossing toe aan een microcentrifugebuis van 1,5 milliliter met zeven milligram fibrinogeen in poedervorm.Plaats de buis drie tot vijf minuten in een celkweekincubator van 37 graden Celsius zonder vortexing. Verwijder en veeg voorzichtig met de buis en draai vervolgens de opgeloste oplossing in een microcentrifuge voordat u deze terugbrengt naar de kweekkap. Filtreer de fibrinogeenoplossing met een spuit van één milliliter die is uitgerust met een spuitfilter van 0,22 micrometer.Breng vervolgens de opgeloste fibrinogeenoplossing over op ijs naast het keldermembraanextract en trombine aliquots. Bereid en verwarm de weefselgroeimedia bij 37 graden Celsius, die na het zaaien van de weefsels in de kweekputten worden geïntroduceerd. Verwijder de celkweekplaten uit de incubator enspirateer de kweekmedia.Was de cellen vervolgens eenmaal door vijf milliliter DPBS aan elke kweekplaat toe te voegen. Zuig de DPBS op en maak de cellen los door een milliliter van 0,25% trypsine EDTA toe te voegen aan elke kweekschaal. Plaats de plaat gedurende drie minuten in de celkweekincubator, houd de trypsine vast door drie milliliter wasmedium aan de kweekschaal toe te voegen en breng de cellen vervolgens over naar een conische buis van de juiste grootte.Pellet de cellen door gedurende 10 minuten te centrifugeren op 400 maal G. Zuig de media zorgvuldig op zonder de celkorrel te beschadigen en suspensuleer de cellen vervolgens opnieuw in één milliliter van het wasmedium. Tel de cellen met behulp van een hemocytometer en trypan blauwe kleurstof onder heldere veldmicroscopie.Om zes weefsels te zaaien, bereidt u voldoende cellen en extracellulaire matrix voor acht weefsels voor, waardoor verliezen en bubbelvorming worden verantwoord. Breng 1,2 miljoen cellen over naar een nieuwe conische buis en verhoog vervolgens het volume tot 10 millimeter met wasmedium. Pellet de cellen door gedurende 10 minuten te centrifugeren op 400 maal G.Bereid 150 microliter ECM-mengsel in een microcentrifugebuis van 1,5 milliliter en bewaar het mengsel op ijs tot gebruik. Zuig de media op uit de conische buis met de cellen en zorg ervoor dat de celkorrel wordt vermeden. Veeg krachtig met een gehandschoende vinger aan het uiteinde van de buis totdat de pellet verschijnt als een celbrij.Breng 120 microliter van de ECM-oplossing over naar de buis met de celslurp en pipetteer voorzichtig op en neer om de cellen in de ECU volledig opnieuw op te suspensie om een enkele celsuspensie te genereren. Vermijd het creëren van bubbels en plaats vervolgens de cel-ECU-suspensie op ijs tot gebruik. Plaats de gekoelde schaal van 10 centimeter met de myotactische putjes bovenop een ijspak in de celkweekkap en zuig de pluronische F-127-oplossing uit elke put op.Laat de resterende pluronische F-127-oplossing loskomen uit het poreuze PDMS en bezink op de bodem van de put door de putten vijf minuten op ijs te laten zitten en vervolgens opnieuw op te zuigen. Pipetteer de ecm-suspensie van de cel voorzichtig om de cellen opnieuw op te schorten en breng vervolgens 105 microliter van de suspensie over in een verse, voorgekoelde buis van 1,5 milliliter door deze dicht bij de bovenkant vast te pakken. Voeg 0,84 microliter van 100 eenheden per milliliter trombine-stamoplossing toe aan de 105 microliter cel-ECM-suspensie.Pipetteer snel en grondig om te mengen, waarbij de introductie van bubbels wordt vermeden. Voeg 15 microliter van het cel-ECU-mengsel toe aan elke put zonder de pipet in de bodem van de put te drukken. Spreid met twee lichte bewegingen de celophanging achter elke paal in de put.Plaats het deksel op de kweekplaat van 10 centimeter en breng het gedurende ongeveer vijf minuten over naar een weefselkweekincubator van 37 graden Celsius. Voeg 200 microliter voorverwarmde weefselgroeimedia toe aan elke myotacticus ruim nadat het cel-ECM-mengsel is gepolymeriseerd. Plaats het deksel op de schaal van 10 centimeter terug en bewaar het in de couveuse tot de differentiatie.Gedurende een kweekperiode van 14 dagen vertoonden myoblasten de aspecten van inheems weefsel door zichzelf te organiseren in de milde tactiek om een 3D-microtissue te vormen met multi-nucleated dwarsgestreepte myotubes. Myotubes hebben sarcomere striations, die werden gevisualiseerd door immunostaining voor sarcomerische alfa-actinine. Om goed uitgelijnde myotubes te bereiken, moeten technische fouten zoals bubbelvorming tijdens het pipetteren of schadelijke pluronische coating tijdens het aspirateren worden vermeden.Een representatieve verplaatsing van de myotactische putplaatpost als reactie op laag- en hoogfrequente elektrische stimulatie, wordt hier getoond, wat aangeeft dat myobuizen reageren op verschillende elektrische stimuli en dienovereenkomstig samentrekken. Kwantificering van de postverplaatsing maakt conversie naar absolute contractiele kracht van de HMMTs mogelijk. Calciumtransience op HMMT's gemaakt van GCaMP6-getransduceerde myoblasten werden ook geanalyseerd als reactie op lage en hoogfrequente stimulatie.Kwantificering van fluorescerende intensiteit kan worden gebruikt als meting voor de calciumverwerkingseigenschappen van HMMT's. De meeste mensen die dit weefselzaaien voor de eerste keer uitvoeren, worstelen met het toevoegen van de cel extracellulaire matrix aan de putten en met bubbelvorming. We raden aan om de techniek te oefenen op enkele reserve microtissue kweekputten met een zeer eenvoudige viskeuze oplossing voor de eerste poging met cellen.
