新生児細菌性髄膜炎は一般的な小児感染症である。K1カプセルを有する大腸菌(大腸菌)は、新生児細菌性髄膜炎を引き起こす最も一般的なグラム陰性病原体であり、全発生率1、2、3の約80%を占める。抗菌化学療法と支持ケアの進歩にもかかわらず、細菌性髄膜炎は依然として罹患率と死亡率が高い最も壊滅的な状態の1つである4.
新生児細菌性髄膜炎の発生は、通常、新生児の局所病変から末梢循環への病原性細菌の侵入によって引き起こされる細菌血症から始まり、続いて血液脳関門(BBB)を通して脳に浸透し、髄膜4の炎症をもたらす。菌血症の発症は、好中球やマクロファージ等を含む細菌と宿主免疫細胞との相互作用に依存する。白血球の50~70%を占める好中球は、細菌感染に対する防御の第1線である5,6である。細菌の侵入の間、活性化された好中球は、感染部位にリクルートされ、超酸化物アニオン、過酸化水素、ヒドロキシルラジカル、および一重酸素7を含む活性酸素種(ROS)を放出する。ROSは、細胞膜、核酸分子および細菌のタンパク質との酸化還元反応を受け、侵入菌8の傷害および死をもたらす。ミトコンドリアは、真核細胞におけるROS産生の主な部位であり、各種オキシダーゼ(例えば、ニコチンアミドアデニンジヌクレオチドリン酸(NADPH)オキシダーゼ複合体、リポキシゲナーゼ系、プロテインキナーゼCおよびシクロオキシゲナーゼ系)の産生を仲介する。好中球における主要な抗菌機構を表すROSの生産のリアルタイム測定は、細菌と宿主の相互作用の間に宿主防御を研究するための有用な方法である。
このプロトコルでは、髄空大腸菌 に感染した好中球における時間依存的ROS産生を、蛍光ROSプローブDHEで定量し、リアルタイム蛍光マイクロプレートリーダーによって検出した。この方法は、病原体と宿主の相互作用の間に他の哺乳動物細胞におけるROS産生の評価にも適用され得る。

大腸菌は、新生児髄膜炎を引き起こす最も一般的なグラム陰性細菌である。新生児細菌性髄膜炎の発生は、純粋に菌血症から始まり、血液中の細菌が血液脳関門を貫通して髄膜炎を引き起こす。好中球は細菌感染に対する防御の第一線である。細菌の侵入の間、活性好中球はROSを放出する。ROSは、侵入病原体を破壊する宿主細胞の主要な細菌性メカニズムを表す。好中球中のROSの産生を測定することは、細菌と宿主の相互作用の間に宿主の防御を開始するための有用な方法である。滅菌ピペットでプレートから単一の細菌コロニーを取り出し、凹面フラスコにリファンピシンを含む5ミリリットルの脳心臓注入媒体に入れます。培養シェーカーで17時間90RPMで、37摂氏で細菌培養をインキュベートする。末梢血サンプルを2000RPMで5分間遠心分離する。遠心分離後、血液サンプルは3つの層に分けられる。下から上へ、赤血球層、白血球層、および血漿層が挙げられる。白血球層を滅菌ピペットを用いた新しいチューブに吸引する。3倍の赤血球のリシスバッファーを追加します。.混合物を十分にブレンドし、室温で5分間置きます。チューブを2000 RPMで5分間遠心分離します。上清を完全に吸引し、廃棄する。赤血球のリシス手順を1〜2回繰り返し、堆積物が白くなるまで繰り返します。2ミリリットルPBSで沈めを再懸濁して細胞を洗浄します。その後、チューブを1000 RPMで5分間遠心分離する。1500万個のセルに対して15マイクロリットルの磁気細胞選別バッファーで沈殿物を再懸濁します。15マイクロリットルの磁気マイクロビーズを加え、十分に混ぜます。4セングレードで30分間インキュベートします。正常な条件下では、成人の人間はマイクロリットル血液あたり約4〜10,000個の白血球を有する。したがって、5ミリリットルのヒト末梢血における全白血球のコストは、5000万未満です。そして15マイクロリットル磁気選別バッファーの50マイクロリットルの磁気ビーズは十分である。好中球の正常な割合は、全白血球の50〜70%である。したがって、5ミリリットルの血液から約1000万個の好中球を得ることができます。チューブに1000万個の細胞につき2ミリリットルの磁気選別バッファーを加え、遠心分離機を4°1,200 RPMで10分間洗浄します。磁気カラムと分離棚を組み立てます。上清を完全に捨て、最大1億個の細胞に対して500マイクロリットルの磁気選別バッファーで堆積物を再懸濁させる。カラムを3ミリリットルの磁気選別バッファーでリンスします。細胞懸濁液をカラムに塗布して、磁気ビーズによって標識された好中球をカラムに取り付けられるようにします。非特異的な細胞を3回、カラムリザーバーが空になると3回の間、3ミリリットルのバッファーを加えて洗い流した。磁気セパレータからカラムを取り外し、新しいチューブに入れます。5 ミリリットルの磁気ソート バッファーを列に追加します。プランジャーを使用して磁気標識されたセルを押し出します。チューブを1,200RPMで5分間遠心分離する。上清を完全に吸引し、1ミリリットル培養培地で沈降液を再懸濁させた。カウンタを含むセルの数を確認します。DHE蛍光プローブを含む培養培地中の細胞濃度を1ミリリットル当たり200万に調整した。DHEプローブをロードするために30分間インキュベーターに好中球を入れます。セルサスペンションを、ウェルあたり200マイクロリットルの96ウェルブラックマイクロプレートに割り当てます。マイクロプレートリーダーの電源を入れます。温度を摂氏37度に設定し、蛍光読み出しモード、運動読み取り型を選択し、蛍光波長を設定する。プレートフォーマットを選択します。読み取り領域を決定します。5分ごとに1時間の蛍光強度を測定します。各読み取り値の前にプレートを3秒間振ります。インキュベーターからマイクロプレートを取り出します。PMA または E44 株を 3 回繰り返して追加します。プレートをマイクロプレートリーダーに入れ、スタートボタンを押してすぐにアッセイを開始します。本稿のプロトコルの概要を用いて、好中球をヒト末梢血から単離し、蛍光プローブDHEを装填した。PMAを添加することにより、好中球中の細胞内ROSは、20分から40分に有意な増加を示し、60分でピークに達した。しかし、私はE44株、ROSの生産はすぐに発生し、依存して時間を増加させると思います。E44株によるROSの発現は、PMAによって引き起こされるものと同様である。好中球におけるROS産生の検出は、好中球のバクテリア機構のさらなる研究に寄与し得る。このプロトコルでは、好中球中のROS産生をリアルタイムに検出する簡単な方法を提供します。この単純な方法は、宿主と細菌の相互作用の間に他の宿主細胞タイプのROS産生を監視するためにも使用することができる。
