生体内蛍光顕微鏡は、生体組織の時空間的ダイナミクスを研究するために広く使用されています。研究者たちは、生物学的プロセスの特異的かつ敏感な蛍光シグナルへの変換を提供する遺伝的にコードされたセンサーを急速に開発しています - 広く利用可能な蛍光顕微鏡を使用して容易に記録することができます1,2.げっ歯類のほとんどの内臓は顕微鏡を用いて調べられてきたが、舌への応用は成功していない3.
味覚細胞のカルシウムイメージングに関する以前の研究は、舌組織を薄く切り離して、舌組織を薄く切り離して、味覚芽4、5、6を得るか、または味覚上皮を剥離して真菌形態の味覚芽7、8を得ることによって行った。これらのサンプルの調製は必然的に侵襲的であったため、神経の内在性、透水性バリア、血液循環などの自然微小環境は、大部分が摂動された。最初のインビタル舌画像化ウィンドウは2015年にChoiらによって報告されたが、流動性タスタント刺激によって引き起こされる動き及び光学的アーチファクトのために信頼性の高い機能的記録は達成できなかった。

このプロトコルでは、実験手順はμTongueを用いた 生体内 におけるマウス真菌形態味覚芽のカルシウムイメージングについて詳細に説明される。まず、人工唾液およびタスタント溶液の調製について説明する。第2に、準定常状態流を達成するためのマイクロ流体システムの設定が導入される。第三に、画像の取得を可能にするためにμTongueにマウス舌を取り付けるために使用される手順が描かれている。最後に、横運動アーチファクトの補正やレシオメトリーを含む画像解析の各ステップを指定します。このプロトコルはマウスの設備および2光子顕微鏡または同等の装置を用いてあらゆる実験室に容易に合わせることができる。

このプロトコルは、神経接続と血液循環がそのまま残っている自然な微小環境における味細胞の機能を観察することを可能にする。この技術を用いて、細胞間通信をインビボで調べることができる。まず、加圧流量灌流システムの貯留層を人工唾液およびタスタントで満たす。圧縮空気ラインをレギュレータ入力に接続し、流体送達システムの空気圧を1平方インチ当たり30~50ポンドに設定します。レギュレータの出力圧力を1平方インチあたり0.4ポンドに設定し、液体がチューブから流出し始めるかどうかを確認します。リザーバからマイクロタングの入力ポートにマニホールドを接続し、マイクロ舌の出力ポートをシリンジポンプに接続し、毎分約300マイクロリットルで液体を引き出して安定した状態を確立します。マイクロ舌の下に掛かる液滴の体積が一定であることを確認し、空気圧と流量を調整して所望のサンプル高さを得る。マイクロ流体システムが設定されたら、圧縮空気ラインを外し、マウスがin vivoイメージング用に準備されるまでシリンジポンプをオフにします。マウスに麻酔を投与した後、TRITC-dextranをレトロ軌道経路を通して静脈内注射し、イメージング中の血液循環を観察する。マウスをスピーヌ位置に置き、その頭に70%エタノールをスプレーします。鉗子を使って頭の皮を軽く持ち上げ、ハサミを使って約7平方ミリメートルの皮膚を切り取ります。頭皮の周りの毛をクリーニングした後、皮膚の下から骨膜を取り除き、頭蓋骨に瞬間的な接着剤を塗布します。次に、カスタマイズされたヘッドフィクサーを取り付けます。瞬間接着剤が硬化したら、ヘッドフィクサーの周りに歯の接着剤を適用し、青色光で照明によってそれを固めます。インスタント接着剤を使用してマウスの下唇をマイクロ舌の下のユニットに接着し、マウスの準備ボードにマウスを置き、マイクロ舌の端にある穴をポストに合わせることで、マイクロ舌の底部をホールドポストに固定します。ボードのヘッドフィクサーホルダーにマウスヘッドフィクサーを締め、マウスヘッドとデバイスの間の距離を調整します。次にヘッドフィクサーホルダーを使用して、マウスヘッドを約45度滑らかに回転させます。マウスをボードに固定した後、プラスチック製のピンセットを使用して舌を軽く引きます。次に、インスタント接着剤を使用して、舌の腹側をマイクロ舌の下の単位の上側に取り付けます。舌を湿らせた状態に保つには、濡れた綿棒で表面を拭き取り、舌の露出した表面に人工唾液に浸した組織を置きます。湾曲したワッシャーをポストに置き、マイクロ舌の下部を保持し、マウスの準備を顕微鏡の段階に移します。マウスの露出した舌を顕微鏡の目的領域の近似中心の下に置き、ステージのダイナミックレンジから逸脱しないようにし、ネジを締めてステージ上にマウスボードをしっかりと取り付けます。マウスの下に加熱パッドを置き、体温を36.5~37.5°Cの間で維持します。マウスの口の中に薄いねじれた紙を入れ、舌の表面に置かれた湿ったティッシュを取り除き、準備したマイクロ舌をマウスの舌の上に置き、舌の表面が画像窓から見えるように、マウスの気管に液体が入るのを防ぎます。最小の圧縮圧力で両端を静かにねじ込むことによってマイクロ舌を固定します。画像の取得を開始するには、顕微鏡ソフトウェアを開き、920ナノメートルの2光子レーザーをオンにします。マイクロ舌のイメージングウィンドウに目的を下げ、目的を浸漬して、マイクロスコープに水浸漬目的を取り付けます。カメラモードでは、水銀灯の青い光を点灯して舌の表面を照らします。近似焦点面を見つけるには、Z軸を調整し、フィリフォーム乳頭から自己蛍光信号を検索します。次に、XとY調整ノブを使用して、味覚芽を見つけます。マルチフォトンモードに切り替え、励起波長、発光フィルタセット、スキャンモード、フレームサイズを設定して画像取得します。画像窓の中央に味覚芽を使用して、味覚芽の約3分の2の高さで味覚芽を囲む血管を見つけ、血液循環を視覚化します。血流が詰まっている場合は、固定ネジを少し緩めて血流を再開します。Z軸を調整して、味覚細胞の数が十分にある味覚芽のZ平面を見つけ、80秒間2〜6ヘルツでカルシウムイメージングを進めます。イメージングが開始された後、流体システムのリザーバをオンにして、20秒間の味覚溶液を提供します。味覚刺激の20秒後、人工唾液に戻って貯水池を切り替えます。イメージング中は、セッションの間に約3〜4分待って、舌を湿らせ、前のセッションから残っているタスタントを洗い流すために人工唾液を一貫して提供します。Pirt-GCaMP6f-tdTomatoマウスの舌表面は、自己蛍光フィリフォーム乳頭で覆われ、味覚芽がまばらに広がっています。黄色のフィリフォーム乳頭は500〜550ナノメートルの光検出器を使用して捕獲され、深さ約25マイクロメートルまでの舌の表面から観察することができます。GCaMP信号を緑色に500~550ナノメートルフィルターで検出し、tdTomato信号を赤色で、607~670ナノメートルフィルタで検出し、味覚細胞を表す。比メトリック分析用にtdTomato信号を取得します。味覚芽を囲む血管は、447〜460ナノメートルフィルタセットを使用して、構造的に味覚芽を支えるコラーゲン結合組織に設定された500〜550ナノメートルフィルタを使用して取得されます。この代表的な例では、カルシウムイメージングを用いたインビボ味スクリーニングでは、各味細胞は破線で区切られる。今回の試験では、細胞2は甘味とうま味の両方のタスト剤に反応した。そして、細胞3は低塩と高塩味の両方に反応した。この味覚芽の細胞のどれも酸味に反応しませんでした。本実験の目的は、自然環境における細胞機能の味を観察することであるため、調製工程で蛍光を血管に送達し、実験中にその循環を確認することが重要である。
