過去数十年にわたり、光ピンセットは、立体構造変化と酵素速度論の同時操作と測定の顕著な可能性により、単一分子レベルでタンパク質相互作用のメカノケミストリーを解明するための貴重なツールでした1,2。特に、細胞内の分子モーターが及ぼす力の範囲の力を適用および測定する能力と、サブナノメートルの構造変化を測定する能力により、光ピンセットは、モータータンパク質の化学力学的特性とその機械的制御を解明するためのユニークな単一分子ツールになりました。
超高速フォースクランプ分光法(UFFCS)は、3ビーズ形状の負荷下での分子モーターの高速速度論を研究するために開発された、ピンセットに基づく単一分子力分光法です(図1a)3,4。UFFCSは、モータータンパク質への力の加わるタイムラグを光ピンセットの物理的限界、すなわちシステムの機械的緩和時間まで短縮し、ミオシンの実行開始後(数十マイクロ秒)に迅速に力を加えることを可能にします3。この能力を利用して、高速骨格3および心臓5筋ミオシンの初期の機械的イベントを調査し、パワーストロークの負荷依存性、弱結合状態と強結合状態、および生化学的(Pi)および機械的(パワーストローク)イベントの順序を明らかにしました。

図1aに示すように、このシステムでは、単一のアクチンフィラメントが、二重光ピンセット(「ダンベル」)の焦点に閉じ込められた2つのポリスチレンビーズの間に吊り下げられています。不均衡な正味力F = F1-F 2は、高速フィードバックシステムを介してフィラメントに課され、フィラメントは、正味の力が反対方向に反転するユーザー定義の反転点に到達するまで、一方向に等速で移動します。モータータンパク質がフィラメントと相互作用していないとき、ダンベルは単一のモータータンパク質が付着している台座ビーズにまたがる三角形の波状(図1b、下のパネル)で自由に前後に動きます。相互作用が確立されると、ダンベルによって運ばれる力はモータータンパク質に非常に急速に伝達され、モーターはミオシンがアクチンから分離するまで、相互作用時にフィードバックシステムによって加えられた力の強さと方向の下でステップすることによってフィラメントを移動させ始めます。トラップされたアクチンフィラメントの極性に依存するモーターのステッピングによって生成される変位であるため、負荷は、加えられた力の方向に応じて、補助的、つまりモーター変位と同じ方向に押す(図1b上のパネルを押す)、または抵抗性、つまりモーターの変位に対して反対方向に引っ張る(図1bの引っ張り)のいずれかになります。 上パネル)は、加えられた負荷の強度と方向性の両方によって運動処理能力の化学力学的調節を研究することを可能にする。

超高速フォースクランプ分光法は、レーザーピンセットに基づく単一分子技術であり、最高の時間分解能で負荷がかかった状態でのミオシンモーターの化学力学を調べることを可能にします。この技術により、力の方向性を完全に制御しながら、モーター相互作用のすべての段階で力を一定に維持することにより、力の生成における非常に迅速なイベントを調査できます。適切な調整を行うことにより、このプロトコルは、キネシンやダイニンなどの他の種類の処理モーターに簡単に適用して、強制的に規制を明らかにすることができます。音響光学デフレクターの1セットをリニアトランスレータに取り付けた後、アイリスを取り、対物レンズの背面開口部サイズに合わせて絞りを調整します。対物レンズを、ねじ付き対物レンズハウジングを中心としたアイリスに置き換えます。圧電結晶によってブロックされたビームの部分がアイリスの後に見えるまでトランスレータをレーザービームに向かって動かし、ビームがアイリス開口部を完全に満たすまでトランスレータを少し後方に回します。酢酸ペンチル中でシリカビーズを調製するには、まず1.2ミクロンの10%固体シリカビーズ20マイクロリットルを1〜1.5ミリリットルのアセトンでボルテックスおよび30秒間の超音波処理で希釈する。超音波処理後、遠心分離によってビーズを収集し、2回目の洗浄のために1ミリリットルの新鮮なアセトンにビーズペレットを再懸濁する。2回目のアセトン洗浄後、同じ遠心分離条件下で1回の洗浄につき1ミリリットルの新鮮な酢酸ペンチルでビーズを3回洗浄し、ペレットを100マイクロリットルの1%ニトロセルロースおよび900マイクロリットルの酢酸ペンチルに再懸濁する。次に、純粋なエタノールに浸したラボティッシュを使用して、24 x 24ミリメートルのガラスカバーガラスを注意深く拭いてから、きれいなピンセットを使用してガラスをエタノールで直接すすぎ、カバーガラスを穏やかな窒素流下で乾燥させます。調製したシリカビーズストックを30秒間ボルテックスして超音波処理し、エタノール洗浄した24 x 60ミリメートルのカバーガラスを使用して、2マイクロリットルのシリカビーズ溶液を小さいカバーガラスの表面に塗りつけます。ビーズが乾いている間に、示されているように26 x 76ミリメートルのスライドをきれいにし、スライドの長い端にテープで留められた3ミリメートルの両面テープで2本の太さ60〜100ミクロンの線を配置します。清潔なピンセットを使用して、ビーズをスライドチャンバーの内側に向けてビーズコーティングされたカバーガラスをテープの上に置き、チャンバーに15ミリモルのリン酸バッファーと浮遊ポリスチレンビーズ溶液を満たします。次に、チャンバーをシリコングリースで密封します。明視野カメラのピクセル対ナノメートル比を較正するには、視野の中心から約5ミクロンのカバーガラス表面上のシリカビーズの最初の画像を取得します。ステージを移動してビードを視野の中心に向かって10ミクロン移動させ、2番目の画像を撮影します。ビーズの2つの位置間のピクセル距離を測定して、カメラのキャリブレーション定数を計算します。トラップ位置を較正するには、1つの浮遊粒子を1つのトラップにトラップし、音響光学偏向器を0.2メガヘルツステップで動かし、粒子の画像と各ステップの偏向器の対応する周波数を取得します。その後、同様にして第2トラップについてキャリブレーションを繰り返す。トラップのパワーと剛性を校正するには、各トラップに単一の粒子をトラップし、音響光学偏向器を使用して両方のトラップを0.2メガヘルツステップで変位させ、象限フォトダイオード検出器とそれに対応する音響光学偏向器の周波数で両方のトラップの粒子のブラウン運動を記録します。記録されたブラウン運動のパワースペクトルにローレンツ関数を当てはめて、キャリブレーション定数を取得します。測定用のサンプルを組み立てるには、カバーガラス表面にシリカビーズが付いた新しいチャンバーにビオチン化BSA1ミリリットルあたり1ミリグラムを追加します。5分後、チャンバーをABバッファーで洗浄し、ストレプトアビジン1ミリリットルあたり1ミリグラムをチャンバーに加えます。5分後、実証されたようにチャンバーを洗浄し、2マイクロモルのカルモジュリンを添加したM5Bバッファーに3ナノモルのビオチン化ミオシン-5B重メロミオシンを追加します。5分後、AB緩衝液中の2マイクロモルカルモジュリンを添加したビオチン化BSA1ミリリットル当たり1ミリグラムでチャンバーを3回洗浄し、最終洗浄後3分間待ちます。反応混合物を流し、シリコングリースでチャンバーを密閉します。サンプルを測定するには、チャンバーを顕微鏡ステージに置き、浮遊アルファアクチニンビーズに焦点が合うまで対物レンズを動かします。1つのトラップのスイッチを入れ、1つのビーズをトラップします。最初のトラップが占有されたら、トランスレータを動かしてトラップされたビードをカバーガラス表面の近くに配置して、複数のビーズをトラップしないようにし、2番目のトラップにビードをトラップします。サンプルを長距離トランスレータに通して、溶液内に少なくとも5ミクロンの長さのアクチンフィラメントを配置します。フィラメントが発見されたら、サンプルを移動して、トラップされたビードがフィラメントの一方の端に近づくようにします。フィラメントを取り付けたら、ビーズの距離をおおよそのフィラメント長に調整し、ステージを動かして、バインドされていないビード方向に流れを作ります。フィラメントは流れによって引き伸ばされ、最終的に2番目のビーズに結合します。結果として生じる複合体は呼ばれます ダンベル「アクチン-ミオシン接触を確立するには、2つのトラップを静かに分離し、フィラメントを約3ピコニュートンまでのプリテンションレベルに設定します。ダンベルの剛性を調べるには、1つの音響光学偏向器の周波数を変更し、その結果、位置信号を介して後続のビードにモーションが伝達されることを確認することにより、トラップを三角波で振動させます。ステージを動かしてダンベルを台座シリカビーズの近くに配置し、アクチンフィラメントがシリカビーズ表面に接触するようにダンベルの高さを調整します。この代表的な位置記録に見られるように、ミオシンモーターがアクチンフィラメントと相互作用していないとき、トラップされたビーズは溶液の粘性抗力に対して等速で移動する。ミオシンモーターがフィラメントと相互作用し始めると、移動するフィラメントによって運ばれる力が非常に急速にタンパク質に伝達され、システム速度はゼロに低下し、ステッピングイベントは実行の終わりまで一定の力の下で発生します。力はフィードバックシステムによって正の方向から負の方向に切り替わり、ビードがユーザーが設定した振動範囲の端に達すると力の方向が切り替わります。ミオシンがフィラメントを結合して正の方向に変位させると、ビーズを振動範囲の上端に向かって押します。これが補助力の下で起こると、ミオシンのランは振動エッジでの力方向反転によって中断され、ランの長さがダンベル振動の振幅に制限されます。最適な空間分解能と時間分解能を達成するには光学系の正確な位置合わせが必要であり、加えられた力の値を正確に決定するには正確なキャリブレーションが必要です。この技術は、DNA中の転写因子のスライディングおよびターゲット検索、ならびに微小管フィラメント上に誘導するメカノトランスダクションタンパク質間の動的相互作用の研究に適応されています。
