精密切断肺スライス法は、局所的バイアスなしに3次元の構造と空間的方向を維持し、血管および気道収縮性測定のための生きた代理モデルとして役立つことができる。タイムラプスイメージング、ウェスタンブロット、RNA解析と組み合わせて、精密切断肺スライスは、多種多様な障害の根本にあるシグナル伝達カスケードの包括的な理解に寄与し、肺血管疾患における病態生理学のより良い理解につながります。まず、安楽死させた成体マウスをスーピーヌの位置に置き、足と鼻を粘着テープでテーブルにテーピングして所定の位置に固定します。70%エタノールでマウスの腹側表面をスプレーします。鉗子を使用して膀胱の位置でマウスの腹部の皮膚を持ち上げ、手術用はさみを正中線で切断し、頚部領域に優れた位置に移動して気管を露出させる。その後、ピンセットを使用して下層筋膜層を持ち上げ、外科用はさみを使用して内臓や経帯を露出させ、再び優れた部分に劣って切断する。心臓と肺を露出させるために正中線で胸骨をカットします。肝臓と腹部の区画から慎重に横隔膜を解剖します。腸の下に下の大静脈を見つけて、それをカットします。右心室を見つけた後、25〜30ゲージの蝶の針を使用して、0.5ミリリットルの1%分画ヘパリンを右心室に注入し、PBSの10〜15ミリメートルでゆっくりとゆっくりと安定して洗い流します。喉頭を見つけた後、鈍い先端ピンセットを使用して周囲の筋膜および組織を解剖し、手術縫合糸を気管の下に置き、外科的結び目をゆるく結ぶ。弦よりも優れた気管に小さな穴を開け、20ゲージの鈍い針でカニューレートします。外科的結び目を使用して針と気管の周りの縫合線を締めます。その後、気管にアガロースの2.5〜4ミリリットルを注入し、肺の膨張を監視します。アガロース注射後、肺に冷たいPBSを注ぎ、アガロースを固める。その後、外科的縫合糸よりも優れた気管を切断し、癒着および筋膜を除去することによって、肺と心臓を隔体から解剖する。固化後、DMEMの中の組織を氷の上のペトリ皿に入れ、すぐにビブラームマシンを使用して1つのローブをスライスする。肺のスライスを準備するには、内側側を上に向けておいて、スーパーグルーでサンプルをプラットフォームに取り付けます。次に、サンプルコンテナーに PBS を入力します。周囲のPBSとサンプルを冷たく保つために氷で容器を満たします。ビブラートメをオンにします。厚さを300マイクロメートル、周波数を100ヘルツ、0.6ミリメートルに振幅、毎秒5マイクロメートルに調整します。アレンレンチを使用して、ブレードホルダーを安全な位置に回し、顎を開いてブレードを挿入します。その後、アレンレンチで顎を締めます。サンプルを箱の上に置き、PBSに沈み込むかどうかを確認します。ボックスを正しい位置にスライドさせて、安全であることを確認します。ブレードホルダーをスライスする適切な位置にします。ビブラートメブレードをブロックの端まで持ち上げ、サンプルの上部に収まるまで手動でブレードを下げます。設定を確認し、ビブラートを実行します。新鮮なスライスを切断した後、PBSからサンプルを取り出し、1回限りの抗ミコティック抗生物質を含むDMEMの10ミリリットルで滅菌ペトリ皿に入れます。ペトリ皿にすべてのスライスを入れた後、ブレードを完全に引き込み、アレンレンチを使用してブレードを安全な位置に上げます。サンプルは摂氏37度でDMEMに保存してください。そして、顕微鏡のスライドの上にビブラートメのスライスを置きます。余分な媒体を取り除くためにクリーニングワイプを使用してください。サンプルを位相コントラスト顕微鏡に置き、10~20倍の倍率で血管を識別します。その後、60ミリモルの塩化カリウムまたは1つのマイクロモル・インドセリン-1などの500マイクロリットルの血管収縮剤を加え、スライドを完全に沈水させます。30~60秒間録画してビデオを観察し、キャプチャします。実験を開始する前に、小さなポリスチレンフォームボックスに1リットルの液体窒素を充填し、液体窒素を容器に1.5ミリリットルマイクロ遠心分離チューブ2本入れます。モルタルボウルに4~5個の新鮮なビブラートメスライスを入れます。スライスの上に5〜10ミリリットルの液体窒素を注ぎ、液体窒素が蒸発する前に素早く害虫を使用してスライスを粉末に粉砕します。冷やされたスチール製の実験室スクーパーを使用して、2つのチルドマイクロ遠心分離チューブに粉末をすくい、RNA分離用のRNA抽出試薬500マイクロリットルまたはタンパク質リシス用のRIPAバッファー500マイクロリットルを添加して粉末をライスします。代表的な生存率実験では、PCLSスライスの色変化がゼロ日から1日目、9日目から10日目まで検出されました。ソリューションは当初青く、一晩でピンク色になり、生存率を示しました。色の変更は通常 1 ~ 4 時間以内に発生しますが、時間が長くなる場合があります。ビブラート体組織の562と613ナノメートルの波長の吸光度の間の日次差をここに示す。ここでは、内皮1および塩化カリウムを用いて、ビブラートドームで調製した肺組織における血管構造の収縮を可視化することができる。タモキシフェン注射の1週間後に細胞内でのtdTomato標識の保存は、これらの肺スライスで実証される。追加の実験やイメージングの前に循環からすべての血液を除去するために、肺のクリアとインフレステップをうまく行うことが最も重要です。血管疾患に加えて、PCLSは気道関連疾患の生理生物学の研究に適用することができ、臨床的に関連するシナリオでヒト肺組織でさらに試験することができる。
