ユビキチン(Ub)の共有結合は、基質タンパク質の内部リジン残基(複数可)に、ユビキテリメンテーションと呼ばく、真核生物における翻訳後の最も重要な修飾の1つを表す。ユビキチン活性化酵素(E1酵素)、ユビキチン結合酵素(E2酵素)、ユビキチンリガス(E3酵素)、時にはユビキチン鎖伸長因子(E4酵素)を含む3つの酵素クラスの順次カスケードによって媒介される。ここでは、ユビキチンリガーゼ活性の評価、E2-E3対の連携、および基板選択を可能にする、ユビキタス化アッセイのための インビトロ プロトコルが提供される。協力E2-E3ペアは、E3リガーゼのフリーポリユビキチン鎖および/または自動ユビキトリメンテーションの生成を監視することによってスクリーニングすることができます。基質のユビキタス化は、E3リガーゼの選択的結合によって定義され、 インビトロ 反応のウェスタンブロッティングによって検出することができる。さらに、E2〜Ub排出アッセイが記載されており、機能性E2-E3連携の直接評価に有用なツールである。ここで、ユビキチンのE3依存性転写は、対応するE2酵素からフリーリジンアミノ酸(模倣基質のユビキタス化)またはE3リガーゼ自体の内部リジン(自動ユビキトリメンテーション)に続く。結論として、E3リガーゼ触媒機能に対処するために高速かつ簡単に実行できる3つの異なる in vitro プロトコルが提供されています。

このプロトコルの全体的な目標は、E2酵素特異性、基質選択およびユビキチン伝達を含むE3リガーゼ機能の重要な重要な側面を分析することである。この技術の主な利点は、すべての真核生物源からのタンパク質を組み換えて発現し、特別な装置を必要とせずにインビトロで研究することができるので、その広い適用性です。リジン放電技術を初めて使用する場合、酵素濃度などのアッセイパラメータを最適化して、選択したE3リガーゼの理想的な排出条件を特定することが重要です。インビトロオートユビキセイ法を実行するには、異なるE2酵素の機能能力をテストするピペッティングスキームを設定し、すべてのユビキタス反応と1つの追加反応のために氷上のマスターミックスを準備します。次に、適切なチューブにE2酵素のマイクロモルを1つ加え、指示された条件下で2時間PCRサーマルサイクラーにサンプルをインキュベートします。インキュベーション後、各反応にSDSサンプルバッファーを加え、数回ピペットで混合します。その後、すぐに摂氏95度で5分間サンプルを沸騰させ、変性タンパク質を摂氏20度で保存します。インビトロ基質のユビキタス化アッセイを行うために、すべての反応に対してピペッティングスキームを調製する。1つの反応に必要なマスターミックスの量を計算した後、氷上のすべての反応のためにマスターミックスを準備し、各チューブにマスターミックスを追加します。それぞれのE3リガを個別に追加します。次に、サンプルを PCR サーマルサイクラーに 2 時間インキュベートします。インキュベーションの最後に、各反応に2回SDSサンプルバッファーを加え、ピペットで数回混合し、サンプルを摂氏95度で5分間沸騰させます。リジン放電アッセイを行うために、充電反応のためのピペッティングスキームを設定し、摂氏37度で15分間充電反応をインキュベートします。充電反応を停止するには、1ミリリットル当たり1.8単位の最終濃度にアピローゼを加え、室温で5分間インキュベートします。インキュベーションの最後に、EDTAを30ミリモルの最終濃度に加え、二重蒸留水を使用して反応量を30マイクロリットルに調整します。E2のユビキタスをE3で排出するには、実験の時点に対応する5つのチューブを設置し、各チューブに6.7マイクロリットルの非還元サンプルバッファーを追加し、停止した充電反応の6マイクロリットルをタイムポイントゼロチューブから取り除き、総体積を26.7マイクロリットルに調整します。放電反応をセットアップするには、二重蒸留水、ユビキリメンテーションバッファー、BSA、リジン、E3リガーゼおよび荷電E2を各反応に加える。選択した期間の後、各放電反応から20マイクロリットルのサンプルを対応するサンプルチューブに移します。その後、すぐにサンプルを渦し、10分間摂氏70度に置きます。この代表的なウェスタンブロット分析では、非アクティブなCHIPは自動ユビキティで表されていませんでした。しかし、E3非依存性ユビキチン製品は、非アクティブCHIPの存在下およびCHIPの不在時に形成された。野生型CHIPはUBE2DおよびUBE2Eファミリーのタンパク質と組み合わせるとオートユビキテニケーション化され、フリーポリウビキチン鎖はUBE2Dファミリーのタンパク質と協力して製造されたが、UBE2Eファミリーのタンパク質とは組み合わされなかった。UBE2N/V1によるユビキチンの結合は、遊離ユビキチン鎖の形成を指示した。UNC-45Bの自己ユビキタス化およびユビキタス化は、野生型CHIPでは観察されたが、CHIPの不活性変異体では観察されなかったため、UNC-45BはCHIPの保存基質として機能することを示している。チップの触媒活性をリジン放電アッセイを用いて分析した場合、未充電のUBE2D2酵素は分子量17キロダルトン、単一ユビキチン分子を有するUBE2D2は分子量約26キロダルトンであった。時刻0では、全ての荷電E2収率が観察された。非アクティブなCHIPの存在下で、UBE2D2の微弱なE3リガーゼ非依存性放電が検出されたが、CHIPの自動ユビティリメンテーションは検出されなかった。野生型CHIPの存在下では、UBE2D2の排出は速く、60分以内に完了し、CHIPのオートユビキトリメンテーションはユビキチンを独自のリジン残基に移すことを示した。限られたE2能力のために、できるだけ早く働き、すぐにSDS-PAGEおよびウェスタンブロッティングが続く排出反応を進める。これらのインビトロのユビキタス性プロトコルに従って、特異的なユビキタスリンケージタイプは、リンケージ特異的抗体を用いてウェスタンブロットを探査することによって同定することができる。
