기판 단백질의 내부 리신 잔류물, 유비쿼터스라고 불리는 공정에 유비퀴틴(Ub)의 공유 부착은 진핵 생물에서 가장 중요한 번역 후 수정 중 하나를 나타냅니다. 유비퀴틸화는 유비퀴틴 활성화 효소(E1 효소), 유비퀴틴-컨쥬게이팅 효소(E2 효소), 유비퀴틴 리구아제(E3 효소) 등 3가지 효소 클래스의 순차적 캐스케이드에 의해 매개되며, 때로는 유비퀴틴 사슬 연공인자(E4 효소)에 의해 매개된다. 여기서, 유비퀴틸레이션 분석용 체외 프로토콜이 제공되어 E3 유비퀴틴 리가제 활성, E2-E3 쌍 간의 협력 및 기판 선택의 평가를 가능하게 한다.
