차세대 염기서열 분석(NGS)은 환자 치료와 암 연구 모두에 점점 더 많이 활용되어 치료를 안내하고 과학적 발견을 촉진합니다. 조직은 종종 제한적이며 가변 종양 함량을 가진 작은 표본이 일상적으로 사용됩니다. 따라서 종양 적합성과 완전성은 의미있는 데이터를 얻는 데 장벽으로 남아 있습니다. 종양 백분율이 낮은 샘플은 실제 변이체와 시퀀싱 아티팩트를 구별하는 데 어려움을 야기할 수 있으며 종종 NGS1에 적합하지 않습니다. 20% 미만의 낮은 종양 함량의 경우의 종양 농축은 재현 가능한 시퀀싱 데이터를 생성하고 저주파 변이체가 놓치지 않도록 충분한 물질을 산출하는 데 도움이 되는 것으로 나타났다2,3. 그러나 제한은 사용 된 플랫폼과 생성 된 데이터의 계획된 사용에 따라 다릅니다.

자동 해부술은 종양 함량이 낮은 사례에서 종양 함량을 풍부하게하는 방법으로 확인되었으며 현재 FFPE 임상 표본에 사용하기 위해 판매되는 자동화 된 조직 해부 장비의 의도 된 기능을 최대 10 μm 두께로 테스트하고 확장했습니다. 이 연구는 자동화 된 해부가 연구 목적으로 FFPE 및 신선한 냉동 인간 또는 동물 조직 절편에 최대 20 μm 두께로 적용될 수 있음을 보여줍니다. 이 프로토콜은 또한 종양 함량이 낮은 조직 및/또는 의미 있는 거대 해부가 어렵거나 실현 가능하지 않은 중첩되고 분산된 종양이 있는 경우 종양 농축을 위한 디지털 주석을 달고 해부를 자동화하는 접근법을 시연하고 NGS에 충분한 핵산의 품질과 수율을 보여줍니다. 따라서 자동 해부는 종양 농축을 위해 중간 수준의 정밀도와 증가 된 처리량을 제공 할 수 있으며 다른 관심 영역을 풍부하게하거나 연구 또는 임상 질문에 답하기 위해 다른 플랫폼과 결합 할 수도 있습니다.
