石灰化大動脈弁疾患(CAVD)は、西洋集団において最も一般的な弁膜心疾患であり、1歳の65歳以上の高齢者の2.5%近くに影響を及ぼす。CAVDは600万人以上のアメリカ人に影響を及ぼし、正常な血流を損なうリーフレットの機械的特性の変化に関連している-1,2.現在、疾患の進行を止めたり、ミネラル退縮を活性化したりする薬理学的治療法はない。CAVDを治療するための唯一の有効な治療法は、手術または経カテーテル大動脈弁置換術3による大動脈弁置換術である。したがって、新しい薬理学的標的を特定するためには、バルブの鉱物化につながる分子メカニズムを調査することが不可欠です。実際、非治療大動脈狭窄症は、左心室機能不全および心不全4のようないくつかの有害な結果を有する。

単離されたマウス弁細胞の使用は、トランスジェニックマウスからの遺伝子組み換え細胞の使用における弁石灰化につながるシグナル伝達経路を調査するために不可欠である。2段階の消化は、迅速かつ効率的な方法です。このプロトコルの最も困難な部分は、8週間マウスからの大動脈弁の分離です。大動脈弁をよりよく視覚化するために、古いマウスで練習する方が良いです。実験を開始する前に、70%エタノールですべての手術器具とワークスペースを洗浄し、滅菌し、手術器具を30分間オートクレーブします。最初のセットアップが準備ができたら、エタノールを使用して8週齢の安楽死マウスの胸部および腹部領域を洗浄することから始める。はさみを使用して、腹部とマウスの胸部を開き、左心房と左心室の間を小さな外科用ハサミで切る。冷たいPBSの10ミリリットルを浸透させることによって心臓から血液を除去し、心臓を切断し、上昇大オルタの3ミリメートルを維持する。体型顕微鏡では、心室の中央で心臓を水平に切断し、左心室を大動脈に向かって切断し、大動脈弁を慎重に解剖することによって大動脈弁を解剖する。小さな35ミリメートルのティッシュ培養皿に一緒にバルブをプールします。抗生物質を補った5ミリリットルの10ミリモルコールドHEPESで75ミリリットルの細胞培養皿で単離されたバルブを洗浄し、連続揺れで摂氏37度で30分間コラゲラーゼタイプ1の5ミリリットルでインキュベートします。インキュベーション後、チューブを150倍Gで5分間遠心分離し、10ミリモルHEPESの2ミリリットルで1回、30秒間高速で渦巻き状でペレットを洗浄します。チューブから35ミリメートルの培養皿に結果の懸濁液を収集し、慎重に新鮮なチューブに組織断片を転送するために薄いピンセットを使用しています。その後、35分間連続撹拌下で37°Cでコラゲレーターーゼタイプ1の5ミリリットルで15ミリリットルのチューブにペレットをインキュベートします。1ミリリットルのピペットと遠心分離機を150倍Gで摂氏4度で5分間再懸濁して細胞を分離する。分離したペレットを完全なDMEMの2ミリリットルに再懸濁し、摂氏4度で5分間Gの150倍の遠心分離を行い、細胞を2回洗浄します。めっきの場合は、ペレットを完全な培地の1ミリリットルに再懸濁し、最小量の培地で6つのウェル細胞培養皿の1つのウェルに加えます。プレートを摂氏37度で5%の二酸化炭素で乱さないようにしてください。3日間のインキュベーションの後、顕微鏡下で細胞を観察することによって組織の破片に近い成長を確認する。1,000個の細胞が見える場合は、オートクレーブされたピンセットで組織の破片を慎重に取り除き、培地を変更します。70%の合流度に達した後に細胞をトリプシン化し、75ミリリットルの組織培養皿に移す。アッセイを開始する前に、バイオセーフティフードを70%エタノールで洗浄し、DMEM培地を摂氏37度に温めます。次に、完全なDMEMで6つのウェルプレートに条件ごとに100,000個の細胞をシードし、摂氏37度で培養します。24時間後、上清培地を石灰化培地に交換し、37°Cで7日間細胞をインキュベートし、3日目に培地を交換します。7日後、アルセナゾIII試薬を用いて、650ナノメートルで吸光度を記録することにより、96ウェルプレート中のカルシウム濃度を測定します。代表的な分析では、培養3~5日後に免疫蛍光染色で各種バルブ細胞の型が同定された。マウスVICの発現ビメンチンおよびαSMAは、バルブ細胞の有意なマーカーである。さらに、CD31免疫蛍光染色は、VIC培養における内皮細胞からの汚染がないことを確認した。リン酸リッチな石灰化培地を用いてVICを培養すると、細胞内の石灰化を刺激し、カルシウム節に対する赤陽性染色でさらに確認した。プロトコルは異なったマウスからの3から5つの弁のプールに基づいている。ごみの交尾と3つの異なる生物学的複製の使用は、発見を検証するために重要です。マウス弁細胞の使用は、トランスジェニックマウスから細胞を単離することによって大動脈狭窄につながる分子経路を理解するために不可欠である。このプロトコルは、以前石灰化大動脈弁の鉱物回帰における受容体への経路の影響を調べるのに使用されてきた。
