骨格筋は硝酸塩の主要な貯蔵庫として機能し、亜硝酸塩に変換された後、運動中にNOの発生源として機能します。ここでは、筋肉やその他の組織でこれらのイオンを測定するための方法論を紹介します。まず、蒸留水中で890ミリモルのフェリシアン化カリウムと118ミリモルのn-メチルマレイミドを組み合わせて硝酸塩保存または停止溶液を調製し、溶液中に結晶が残らないようにしてから、非イオン性界面活性剤を1対9の比率で加え、泡立ちを避けるために穏やかに混合します。停止液を蒸留水で1対9の比率で希釈し、希釈した停止液をホモジナイズチューブに加えます。次に、以前に単離して凍結したラットの筋肉組織を氷上で解凍します。組織が溶けたら、骨格筋から残りの脂肪と結合組織を取り除きます。骨格筋の一部を切り取り、ガーゼで拭いて乾かします。適量の骨格筋組織の重量を量り、次に、停止液を含むホモジナイズチューブに予備重量組織を置きます。回転式ホモジナイザーで均質化するには、事前に計量された骨格筋と事前に測定された停止液を含むM型チューブを機械に入れます。各サンプルを2回ホモジナイズし、各ホモジナイズ後、チューブをすぐに氷の上に5分間置き、冷却します。均質化後、チューブを短時間遠心分離します。チューブを氷上に戻し、ボルテックスする前に適切な量のメタノールを15秒間加えます。サンプルを再度ホモジナイズし、氷上で30分間インキュベートしてから、サンプルを遠心分離し、上清を吸引して、亜硝酸塩と硝酸塩レベルの測定に進みます。ビーズ均質化の場合は、骨格筋組織をビーズ含有チューブに入れ、装置で利用可能な最高速度で45秒間2回ホモジナイズします。各均質化の後、すぐにチューブを氷上に5分間置き、冷却し、次にチューブを遠心分離し、チューブを氷上に戻し、適切な量のメタノールを加えてから15秒間ボルテックスします。サンプルをもう一度45秒間ホモジナイズし、氷上で30分間インキュベートします。サンプルを遠心分離し、上清を吸引し、亜硝酸塩と硝酸塩レベルの測定に進みます。粉砕機ベースの均質化では、希釈した停止液を含むチューブを準備し、チューブの重量を計量して記録します。ドライアイスで粉砕機ツールを30分間冷却した後、液体窒素で冷やしたピンセットを使用して、事前に計量した組織サンプルを粉砕機に移し、少量の液体窒素を加えて組織が液体窒素温度であることを確認します。液体窒素の95%が気化した後、破砕工具を組織の上に置き、しっかりと押してサンプルのつぶれを感じ、木槌を使用して破砕工具を3〜5回叩きます。試料全体が粉砕されたら、液体窒素冷却スプーンおよびピンセットを用いて、希釈停止液を含む予め秤量されたチューブに破砕組織を直接移す。次に、チューブを15秒間ボルテックスしてから、チューブを開いて、蓋に詰まった組織がないか確認します。ある場合は、それを取り除いてから、もう一度渦を巻いてください。サンプルを2〜3秒間短時間遠心分離します。チューブを再度計量し、この新しい重量から元のチューブ重量を差し引いて組織の重量を計算します。チューブを氷の上に置きます。適量のメタノールをチューブに加え、チューブを15秒間完全にボルテックスしてから氷上で30分間インキュベートし、サンプルを遠心分離し、上清を吸引し、亜硝酸塩と硝酸塩のレベルの測定に進みます。回転式ホモジナイザー、ビーズホモジナイザー、および粉砕機を用いて調製したラット骨格筋ホモジネートの硝酸レベルおよびラット骨格筋ホモジネートは類似していた。興味深いことに、亜硝酸塩レベルについては、粉砕機によって調製されたホモジネートサンプルが最も高い値を示し、他の2つのサンプル値とは統計的に異なっていました。ビーズホモジナイザーを使用して20、50、および200ミリグラムの組織から調製した大殿ホモジネートの亜硝酸塩と硝酸塩のレベルを比較すると、硝酸塩レベルも亜硝酸塩レベルも筋肉サンプルサイズ間で有意差がないことが示されました。異なるげっ歯類の脚骨格筋組織の硝酸塩レベルを比較すると、大臀筋は他の3つの筋肉組織よりも約2倍高い硝酸塩レベルを持っていることが明らかになりました。しかし、これらの差は統計的有意性に達しなかった。硝酸塩レベルと同様に、大殿筋の硝酸塩濃度も他の3つの筋肉組織よりも高く、腓腹筋サンプルの硝酸塩濃度よりも有意に高かった。私たちの方法は比較的簡単で、サンプル調製中に硝酸塩と亜硝酸塩を保存しながら、小さなサンプルに適しています。
