In dit multi-step retinale inductieprotocol werden hPSC's stap voor stap begeleid om het retinale lot te verkrijgen en zelf georganiseerd in retinale organoïden met gelamineerde NR en RPE. Tijdens de differentiatie hebben hPSC's alle belangrijke stappen van de menselijke retinale ontwikkeling in vivosamengevat , van EF, OV en RPE tot retinale laminatie, waarbij alle subtypen retinale cellen worden gegenereerd, inclusief retinale ganglioncellen, amacrinecellen, bipolaire cellen, staaf- en kegelfotoreceptoren en ...

Retinale degeneratieve ziekten (ED's), zoals leeftijdsgebonden maculaire degeneratie (AMD) en retinitis pigmentosa (RP), worden gekenmerkt door de disfunctie en dood van fotoreceptorcellen en leiden meestal tot onomkeerbaar verlies van het gezichtsvermogen zonder effectieve manieren om te genezen1. Het mechanisme dat aan deze ziekten ten grondslag ligt, is grotendeels onbekend, deels als gevolg van een gebrek aan menselijke ziektemodellen2. In de afgelopen decennia zijn er aanzienlijke vorderingen geboekt in de regeneratieve geneeskunde door middel van stamceltechnologie. Veel onderzoekers, waaronder wijzelf, hebben aangetoond dat menselijke pluripotente stamcellen (hPSC's), inclusief menselijke embryonale stamcellen (hESCs) en door de mens geïnduceerde pluripotente stamcellen (hiPSC's), kunnen differentiëren in alle soorten retinale cellen, zelfs mini-retinale weefsels door verschillende differentiatiebenaderingen3,4,5,6,7,8,9,10, 11, biedt een enorm potentieel in ziektemodellering en celtherapie12,13,14.
Het inductieproces van hPSC's naar retinale cellen is echter zeer gecompliceerd en tijdrovend met een lage herhaalbaarheid, wat onderzoekers met rijke ervaring en hoge vaardigheden vereist. Tijdens het complexe en dynamische inductieproces zullen een aantal factoren de opbrengst van retinale weefselsbeïnvloeden 15,16,17. Ook variëren verschillende inductiemethoden vaak aanzienlijk in timing en robuuste expressie van retinale markers, wat de monsterverzameling en gegevensinterpretatie kan verstoren3. Daarom zou een eenvoudig protocol van retinale differentiatie van hPSC's met stapsgewijze begeleiding in trek zijn.
Hier, op basis van onze gepubliceerde studies18,19,20,21,wordt een geoptimaliseerd retinale inductieprotocol beschreven om retinale organoïden (RO's) te genereren met rijke kegelfotoreceptoren van hPSC's, waarvoor geen supplement van retinoïnezuur (RA) nodig is. Dit protocol richt zich op de beschrijving van de meerstappenmethode om neuraal netvlies en RPE te genereren. EB-vorming is het essentiële onderdeel van de vroege inductiefase. Zowel de grootte als de platingdichtheid van EB's zijn kwantitatief geoptimaliseerd, wat de opbrengst van retinale weefsels wetenschappelijk verbetert en de herhaalbaarheid bevordert. In het tweede deel van de inductie organiseren optische blaasjes (OVs) zichzelf in de therapietrouwcultuur en vormen zich ERRO's in de suspensiecultuur; de tijdscursussen en efficiëntie van dit deel variëren aanzienlijk in verschillende hPSC-lijnen. De rijping en specificatie van retinale cellen in RA's vindt voornamelijk plaats in het middelste en late stadium van inductie. Zonder de toevoeging van RA kunnen volwassen fotoreceptoren met zowel rijke kegels als staafjes worden geproduceerd.
Het doel van dit protocol is om elke stap kwantitatief te beschrijven en te detailleren voor onervaren onderzoekers om te herhalen. Verschillende hPSC-lijnen zijn met succes geïnduceerd in RO's door dit protocol met een robuuste opbrengst van kegelrijke retinale weefsels en een hoge herhaalbaarheid. HPSC-afgeleide RO's met dit protocol kunnen de belangrijkste stappen van retinale ontwikkeling in vivosamenvatten en op lange termijn overleven, wat downstream-toepassingen vergemakkelijkt, zoals ziektemodellering, geneesmiddelenscreening en celtherapie.

retinal organoid induction system for derivation of 3d retinal tissues from human pluripotent stem cells

Retinale degeneratieve ziekten zijn de belangrijkste oorzaken van onomkeerbare blindheid zonder effectieve behandeling. Pluripotente stamcellen die het potentieel hebben om te differentiëren in alle soorten retinale cellen, zelfs mini-retinale weefsels, houden enorme beloften in voor patiënten met deze ziekten en vele mogelijkheden in ziektemodellering en geneesmiddelenscreening. Het inductieproces van hPSC's naar retinale cellen is echter ingewikkeld en tijdrovend. Hier beschrijven we een geoptimaliseerd retinaal inductieprotocol om retinale weefsels met een hoge reproduceerbaarheid en efficiëntie te genereren, geschikt voor verschillende menselijke pluripotente stamcellen. Dit protocol wordt uitgevoerd zonder de toevoeging van retinoïnezuur, wat de verrijking van kegelfotoreceptoren ten goede komt. Het voordeel van dit protocol is de kwantificering van EB-grootte en platingdichtheid om de efficiëntie en herhaalbaarheid van retinale inductie aanzienlijk te verbeteren. Met deze methode verschijnen alle belangrijke retinale cellen sequentieel en vatten ze de belangrijkste stappen van retinale ontwikkeling samen. Het zal downstream-toepassingen vergemakkelijken, zoals ziektemodellering en celtherapie.

Het retinale inductieproces in dit protocol bootst de ontwikkeling van het menselijk foetaal netvlies na. Om de retinale differentiatie te initiëren, werden hPSC's gedissocieerd in kleine klonten en gekweekt in suspensie om de vorming van EB's te induceren. Op D1 vormden de geüniformeerde celaggregaten of EG's (figuur 1C). Het cultuurmedium werd geleidelijk overgezet naar NIM. Op D5 werden EB's op de ECM-gecoate kweekschalen geplakt. Hier beschrijven we een geoptimaliseerd retinale organoïde inductiesysteem, dat geschikt is voor verschillende menselijke pluripotente stamcellijnen om retinale weefsels met een hoge reproduceerbaarheid en efficiëntie te genereren.

Het inductieproces van hPSC's naar retinale cellen is ingewikkeld en tijdrovend. Dit geoptimaliseerde protocol kan retinale weefsels genereren met een hoge reproduceerbaarheid en zonder kosten. Het voordeel van dit protocol is de kwantificering van EB-grootte en platingdichtheid om de efficiëntie en herhaalbaarheid van retinale inductie van hPSC's aanzienlijk te verbeteren.Met deze methode verschijnen alle belangrijke retinale cellen sequentieel en vatten ze de belangrijkste stappen van retinale ontwikkeling samen. Het zal ziektemodellering en celtherapie voor retinale degeneratieve ziekten vergemakkelijken. We demonstreren de procedure en zijn met Yuanyuan Guan, een promovendus, en Bingbing Xie, een technicus van het laboratorium.Begin met het bereiden van 50 ml ECM-oplossing door 1 ml van de derde 50 maal voorraadoplossing toe te voegen aan 50 ml DMEM. Voeg vervolgens 1 ml van deze bereide ECM-oplossing toe aan elke put van een plaat met zes putten en incubeer deze gedurende een uur bij 37 graden Celsius en 5% koolstofdioxide. Bereid hPSC-onderhoudsmedium voor volgens de instructies van de fabrikant en verwarm het gedurende 30 minuten voor op kamertemperatuur.Giet een cryogene injectieflacon met hPSC's uit een tank met vloeibare stikstof door deze gedurende 30 seconden in een waterbad bij 37 graden Celsius te incuberen. Desinfecteer het zorgvuldig met een 75% alcoholspray en plaats het in een bioveiligheidskast. Breng de celsuspensie van de injectieflacon over naar een buis van 15 millimeter.Voeg vervolgens druppelsge voor druppelsgevoudig 5 ml voorverwarmd onderhoudsmedium toe aan de buis met behulp van een pipet van 5 ml, terwijl u de buis voorzichtig schudt om de hPSC's te mengen. Centrifugeer de buis gedurende vijf minuten op 170 maal G. Verwijder voorzichtig het grootste deel van het supernatant met behulp van een pipet van 1 ml, waarbij ongeveer 50 microliter supernatant achterblijft om te voorkomen dat de cellen verloren gaan.Hang de pellet opnieuw op met 1 ml onderhoudsmedium door op en neer te pipetteren. Verwijder ECM uit de voorgecoate putten en voeg 1,5 milliliter onderhoudsmedium toe aan elke put. Verdeel vervolgens 0,5 milliliter celsuspensie in elke put.Schud de plaat voorzichtig om de hPSC's gelijkmatig te verdelen. Plaats de plaat in een incubator bij 37 graden Celsius en 5% koolstofdioxide gedurende ten minste 24 uur om de celtrouw te bevorderen. Verander elke dag het medium en doorloop hPSC's bij 80% confluentie.Start op dag-0 de differentiatie door de 80% confluentiecellen uit één put van de zesputplaat te verwijderen. Verzamel de cellen met EDTA-dissociatieoplossing zoals beschreven in het tekstmanuscript. Verwijder de EDTA-oplossing en voeg 1 ml onderhoudsmedium toe dat 10 micromolaire fluvastatine bevat om de celdissociatie te stoppen.Verzamel de cellen met een pipet van 1 ml. Breng de celsuspensie over naar een 100 millimeter ultralaag bevestigde petrischaal en voeg 9 ml onderhoudsmedium met 10 micromolair fluvastatine toe aan de schaal. Schud de schaal twee keer voorzichtig om de cellen gelijkmatig te verdelen.Doe het vervolgens in de incubator bij 37 graden Celsius en 5% koolstofdioxide. Nadat de cellen gedurende ten minste 24 uur zijn gekweekt, observeert u ze onder de microscoop. Voeg 9 ml onderhoudsmedium en 3 ml NIM toe aan een buis van 15 ml.Breng de celculturen over in een centrifugebuis van 15 ml en voeg 10 ml van het voorverwarmde NIM-mengsel toe aan de schaal. Centrifugeer de buis gedurende 3 minuten op 60 maal G om de aggregaten te verzamelen. Verwijder vervolgens het supernatant met een pipet van 5 ml en laat ongeveer 500 microliter achter om te voorkomen dat cellen verloren gaan.Voeg 2 ml van het mengsel toe aan de buis en breng de suspensie terug naar dezelfde petrischaal. Schud de schaal voorzichtig om de celaggregaten gelijkmatig te verdelen. Zet de schaal vervolgens terug in de couveuse.Verwijder op dag 5 ECM uit de voorgecoate gerechten en voeg 10 ml voorverwarmde NIM toe aan elk gerecht. Haal de schaal met Ebs eruit en controleer de kwaliteit van EB's onder de microscoop, zorg ervoor dat ze helder en rond zijn. Verzamel alle EB's in een buis van 15 ml en laat ze 5 minuten bezinken.Verwijder vervolgens het grootste deel van het supernatant en laat ongeveer 2 ml medium achter. Na het tellen van de EB's, zaai ze druppelsgevoudigd met een dichtheid van ongeveer 2-3 EB per vierkante centimeter in gecoate schalen met 10 ml NIM. Schud de gerechten voorzichtig om de EB's gelijkmatig te verdelen en leg ze minstens 24 uur in de couveuse.Gebruik een wolfraamnaald of een naald met een spuit van 1 ml om de morfologisch identificeerbare optische blaasjes mechanisch los te maken, samen met het aangrenzende retinale pigmentepitheel op dag 28 tot 35. Kweek ze in schorsing. Doe 50-60 optische blaasjes in elke 100 millimeter laagaanhechtingscultuurschaal, met 15 ml RDM voor retinale organoïdevorming.Vervang het medium elke 2 tot 3 dagen tot dag-42. Om de retinale differentiatie te initiëren, werden hPSC's gedissocieerd in kleine klonten en gekweekt in suspensie, die sinds dag-1 EB's vormden. Op dag 5 werden EB's op de ECM-gecoate kweekschalen geplateerd en de cellen migreerden geleidelijk uit de EB's.Op dag-16 werd het inductiemedium vervangen door RDM, waardoor de neurale netvliesdomeinen zich vormden en geleidelijk uit de schotel staken, evenals celvormende optische blaasjesachtige structuren omringd door RPE-cellen. Tijdens dagen-28 tot 35, retinale organoïden bestaande uit het neurale netvlies, bevestigd aan een RPE-bol. Aan de ene kant vormden zich cellen.Naarmate retinale differentiatie en specificatie vorderden, produceerden hPSC's subtypen van neuraal netvlies die geleidelijk in lagen werden opgesteld en de architecturale kenmerken van een inheems menselijk netvlies nabootsen. Retinale ganglioncellen werden voor het eerst gegenereerd uit retinale voorlopers en hoopten zich op in de basale kant van het neuro-netvlies. Met dit protocol ontwikkelden retinale organoïden zich tot zeer volwassen fotoreceptoren met zowel rijke staafjes als kegeltjes.Fotoreceptorcellen bevonden zich aan de apicale kant, terwijl amacrinecellen, horizontale cellen, bipolaire cellen en muller gliacellen zich allemaal in de tussenlaag van het neurale netvlies bevonden. De belangrijkste punten van dit protocol zijn het creëren van een hoge kwaliteit van Ebs en het zaaien ervan met de juiste dichtheid.
