그러나 hPSCs에서 망막 세포에 이르는 유도 과정은 매우 복잡하고 반복성이 낮은 시간이 소요되며, 풍부한 경험과 높은 기술을 가진 연구원이 필요합니다. 복잡하고 역동적인 유도 과정에서 여러 가지 요인이 망막 조직의수율(15,16,17)에영향을 미칩니다. 또한, 상이한 유도 방법은 종종 샘플 수집 및 데이터 해석3을혼동할 수 있는 망막 마커의 타이밍과 견고한 발현에 상당히 차이가 있다. 따라서 단계별 지침이 있는 hPSC에서 망막 분화의 간단한 프로토콜이 수요가 있을 것입니다.
여기서, 당사의 발표된 연구18,19,20,21을기반으로, hPSC로부터 풍부한 콘 광수용체를 가진 망막 오르가노이드(ROs)를 생성하는 최적화된 망막 유도 프로토콜이 설명되어 있으며, 이는 레티노산(RA)의 보충제를 필요로 하지 않는다. 이 프로토콜은 신경 망막 및 RPE를 생성하는 다단계 방법의 설명에 중점을 둡니다.
