Aminopeptidase N (APN), et måneskinsenzym, der tilhører metalloproteinase M1-familien, fungerer som en tumormarkør, receptor og signalmolekyle via enzymafhængige og enzymuafhængige veje 1,2. Ud over at spalte de N-terminale aminosyrerester af forskellige bioaktive peptider til regulering af deres biologiske aktivitet spiller APN en vigtig rolle i patogenesen af forskellige inflammatoriske sygdomme. APN deltager i antigenbehandling og præsentation af trimmede peptider, der binder tæt til større histokompatibilitetskomplekserklasse II-molekyler 2,3. APN udøver også antiinflammatoriske virkninger ved at binde med G-proteinkoblede receptorer, der deltager i multipel signaltransduktion, modulerer cytokinsekretion og bidrager til Fc gammareceptormedieret fagocytose i immunresponset 4,5,6,7.
Som en bredt distribueret membranbundet exopeptidase er APN rigelig i svinetyndtarmens slimhinde og er tæt forbundet med receptormedieret endocytose 1,5,8. APN genkender og binder spikeproteinet i den overførbare gastroenteritisvirus til celleindgang og interagerer direkte med FaeG-underenheden af enterotoksinfremkaldende Escherichia coli F4 fimbriae for at påvirke bakteriel adhærens med værtsceller 9,10,11. APN er således et potentielt terapeutisk mål i behandlingen af virale og bakterielle infektionssygdomme.
Siden udviklingen af hybridomteknologi og andre strategier til produktion af monoklonale antistoffer (mAbs) i 1975 har mAbs været meget udbredt i immunterapi, lægemiddelafgivelse og diagnose12,13,14. I øjeblikket anvendes mAbs med succes til behandling af sygdomme, såsom kræft, inflammatorisk tarmsygdom og multipel sklerose12,15. På grund af deres stærke affinitet og specificitet kan mAbs være ideelle mål i udviklingen af antistof-lægemiddelkonjugater (ADC) eller nye vacciner16,17. APN-proteinet er kritisk for selektivt at levere antigener til specifikke celler og kan fremkalde et specifikt og stærkt slimhindeimmunrespons mod patogener uden interferens, herunder lav proteinekspression, enzyminaktivitet eller strukturelle ændringer 5,8,18. Derfor viser terapeutiske produkter baseret på APN-specifikke mAbs løfte mod bakterielle og virale infektioner. I denne undersøgelse beskriver vi produktionen af APN-specifikke mAbs ved hjælp af hybridomteknologi og ekspression af anti-APN rekombinante antistoffer (rAbs) ved hjælp af prokaryote og eukaryote vektorer. Resultatet indikerer, at APN-proteinet blev genkendt af både rAbs udtrykt i pIRES2-ZSGreen1-rAbs-APN-CHO-celler og hybridomafledte mAbs.

Dette gør APN til en attraktiv kandidat i udviklingen af vacciner, der selektivt målretter mod slimhindepitelet. I denne undersøgelse sammenlignede vi produktionen af APN-specifikke monoklonale antistoffer (mAbs) ved hjælp af traditionel hybridomteknologi og rekombinant antistofekspressionsmetode. Vi etablerede også en stabilt transfekteret kinesisk hamsterovarie (CHO) cellelinje ved hjælp af pIRES2-ZSGreen1-rAbs-APN og en E. coli-ekspression BL21 (DE3) stamme, der huser pET28a (+)-rAbs-APN-vektoren . Resultaterne viser, at antistoffer udtrykt i pIRES2-ZSGreen1-rAbs-APN-CHO-celler og mAbs produceret ved hjælp af hybridomer kunne genkende og binde til APN-proteinet. Afledte monoklonale antistoffer anvendes primært i diagnostiske terapeutiske immunreagenser, hvilket fremhæver behovet for at producere stabile og pålidelige antistoffer i begrænset produktion fremstillet. De metoder, der er beskrevet her, viste, at rekombinant antistofproduktion kunne øge monoklonal antistofproduktionseffektivitet og minimere arbejds- og tidsrelaterede omkostninger. Til at begynde med injiceres intraperitonealt 100 mikrogram APN-protein i de udvalgte voksne hunmus for et sidste antigenboost. Efter tre dages injektion opsamles milten fra mus og vaskes med DMEM to gange for at fjerne blod og fedtceller.Filtrer miltcellesuspensionen ved hjælp af et 200 mesh kobbergitter for at fjerne vævsaffald og høste miltceller ved hjælp af centrifugering for at fjerne miltmembranen. Frø musens SP20 myelomceller i en 25 kvadratcentimeter kolbe indeholdende fem ml DMEM suppleret med 6% føtalt bovint serum og kultur cellerne ved 37 grader Celsius og 6% kuldioxid atmosfære for at opretholde cellelevedygtighed. Efter fem til seks dages dyrkning skal cellerne nå 80 til 90% sammenløb efter genoplivning og fremstå runde, lyse og klare under mikroskopet.En dag før hybridisering skal du samle makrofager fra musenes peritoneale hulrum. Dyrk pET28a plus rekombinante antistoffer aminopeptidase NBL21 transformerede bakterier i nærvær af 0,4 millimolær beta-d-1-thiogalactopyranosid i en orbital shaker ved 37 grader Celsius i 10 timer.Frø 100 mikroliter 0,5 gange 10 til den femte kinesiske hamsters ovarieceller pr. Brønd i en 96-brøndplade til inkubation ved 37 grader Celsius og en 6% kuldioxidatmosfære i 18 til 24 timer. Når cellerne når 80 til 90% sammenløb, fortyndes pIRES2-zs Green one rekombinante antistoffer aminopeptidase og plasmid med Opti-MEM til en endelig koncentration på 0,1 mikrogram pr. Mikroliter. Efter fem minutter blandes de 50 mikroliter fortyndet pIRES2-zs Green, et rekombinant antistof, aminopeptidaser og plasmid med en mikroliter lipofectamin 2000 og 49 mikroliter Opti-MEM.Efter 20 minutters inkubation tilsættes 100 mikroliter af blandingen til hver brønd i en 96-brøndplade indeholdende CHO-celler. Fire til seks timer efter transfektion skal mediet udskiftes med DMEM F12 suppleret med 10% FBS. Efter 48 timers inkubation tilsættes 400 mikrogram pr. Brønd i en 96-brøndplade og kultur i 37 grader Celsius, 6% kuldioxidinkubator.I repræsentativ mikroskopisk analyse syntes alle hybridomasceller runde, lyse og klare. De oprensede monoklonale antistoffer med tunge 50 kilodalton og lette 25 kilodaltonkæder blev bekræftet af SDS-PAGE og blev fundet i de rensede ascites. Titre af disse anti-APN monoklonale antistoffer i kultursupernatanter og ascites er vist her.Monoklonal antistofisotyping med mus afslørede, at antistoffer afledt af kloner 5B31, 5B36, 3C48, 5C51 og 6C56 havde immunoglobulin G2B-underklasser, mens APN 2A20 var et immunglobulin G2A kappa-type antistof og monoklonalt antistof APN 3FD9, 3F10 og 10F3 tilhørte immunoglobulin M-type og forarbejdede kappa-lyskæder. De fleste af disse monoklonale antistoffer viste AV-værdier på over 50%, hvilket indikerer, at de målrettede mod forskellige epitoper i APN, mens APN 5C51-antistoffet genkendte antigene epitoper som dem, der genkendes af APN 3C48, 5B31 og 6C56 monoklonale antistoffer. Det amplificerede APN 5B36 VHVL-gen blev ligeret til en pET28a-positiv eller pIRES2-zs grøn en vektor for at konstruere henholdsvis de rekombinante ekspressionsplasmider pET28a-positive RABS APN og pIRES2-zs Green en RABS APN.Antistofferne udtrykt af både pET28a plus rekombinante antistoffer aminopeptidase NBL21 og pIRES2-zs Green one rekombinante antistoffer aminopeptidase NCHO celler blev oprenset og analyseret ved hjælp af ELISA og IFA assays. Imidlertid genkender kun antistoffet udtrykt i supernatanten af pIRES2-zs Green one rekombinante antistoffer aminopeptidase NCHO-celler APN-proteinet.
