メタロプロテイナーゼM1ファミリーに属する月光酵素であるアミノペプチダーゼN(APN)は、酵素依存性および酵素非依存性の経路を介して腫瘍マーカー、受容体、およびシグナル伝達分子として機能します1,2。APNは、様々な生理活性ペプチドのN末端アミノ酸残基を切断してそれらの生物学的活性を調節することに加えて、様々な炎症性疾患の病因において重要な役割を果たす。APNは、主要組織適合遺伝子複合体クラスII分子に強固に結合するトリミングペプチドによる抗原プロセシングおよび提示に関与しています2,3。APNはまた、複数のシグナル伝達に関与するGタンパク質共役受容体と結合し、サイトカイン分泌を調節し、免疫応答におけるFcガンマ受容体媒介食作用に寄与することにより、抗炎症効果を発揮します4,5,6,7。
広く分布している膜結合型エキソペプチダーゼとして、APNはブタの小腸粘膜に豊富にあり、受容体媒介性エンドサイトーシスと密接に関連しています1,5,8。APNは、細胞侵入のために伝染性胃腸炎ウイルスのスパイクタンパク質を認識して結合し、毒素原性大腸菌F4線ブリアのFaeGサブユニットと直接相互作用して、宿主細胞との細菌接着に影響を与えます9,10,11。したがって、APNは、ウイルスおよび細菌感染症の治療における潜在的な治療標的である。
1975年にモノクローナル抗体(mAb)産生のためのハイブリドーマ技術およびその他の戦略が開発されて以来、mAbは免疫療法、薬物送達、および診断に広く使用されてきました12,13,14。現在、モノクローナル抗体は、癌、炎症性腸疾患、多発性硬化症などの疾患の治療に成功しています12,15。モノクローナル抗体は、その強力な親和性と特異性により、抗体薬物複合体(ADC)や新規ワクチンの開発において理想的な標的となり得る16,17。APNタンパク質は、抗原を特定の細胞に選択的に送達するために重要であり、低タンパク質発現、酵素不活性、または構造変化を含む干渉なしに、病原体に対する特異的で強力な粘膜免疫応答を誘発することができる5,8,18。したがって、APN特異的モノクローナル抗体に基づく治療用製品は、細菌およびウイルス感染に対して有望です。本研究では、ハイブリドーマ技術を用いたAPN特異的モノクローナル抗体の産生、および原核生物および真核生物ベクターを用いた抗APN組換え抗体(rAbs)の発現について述べる。この結果は、APNタンパク質がpIRES2-ZSGreen1-rAbs-APN-CHO細胞およびハイブリドーマ由来のモノクローナル抗体の両方で発現したrAbsによって認識されたことを示している。

小腸粘膜に豊富に存在する膜結合メタロペプチダーゼであるブタアミノペプチダーゼN(APN)は、低タンパク質発現、酵素不活性、構造変化などの干渉なしに粘膜免疫応答を開始できます。これにより、APNは粘膜上皮を選択的に標的とするワクチンの開発において魅力的な候補となります。以前の研究では、APNが毒素原性 大腸 菌(大腸菌)F4と伝染性胃腸炎ウイルスの両方の受容体タンパク質であることが示されています。したがって、APNは、抗体薬物複合体またはAPN特異的抗体に基づく新規ワクチンの開発において有望である。本研究では、従来のハイブリドーマ技術と遺伝子組換え抗体発現法を用いたAPN特異的モノクローナル抗体(mAb)の産生を比較した。また、pIRES2-ZSGreen1-rAbs-APNとpET28a(+)-rAbs-APNベクターを保持する 大腸菌 発現BL21(DE3)株を使用して、安定にトランスフェクトされたチャイニーズハムスター卵巣(CHO)細胞株を樹立しました。結果は、pIRES2-ZSGreen1-rAbs-APN-CHO細胞およびハイブリドーマを用いて産生されたmAbsで発現する抗体がAPNタンパク質を認識し、結合できることを示しています。これは、異なるAPN特異的エピトープを標的とする治療薬の開発のためのAPN受容体機能のさらなる解明の基礎を提供します。

Production of Monoclonal Antibodies Targeting Aminopeptidase N in the Porcine Intestinal Mucosal Epithelium

誘導されたモノクローナル抗体は、主に診断用治療用免疫試薬に使用され、限られた生産で安定で信頼性の高い抗体を産生する必要性を強調しています。ここで説明した方法は、組換え抗体の生産がモノクローナル抗体の生産効率を高め、人件費と時間に関連するコストを最小限に抑えることができることを証明しました。これらの方法は、アミノペプチダーゼおよび抗体ベースの抗体薬物複合体およびその他の治療製品の開発に使用され、細菌およびウイルス感染の予防および治療におけるAPNの役割を明らかにするのに役立ちます。手順を実演するのは、私の研究室の大学院生であるYan Liです。まず、選択した成体雌マウスに100マイクログラムのAPNタンパク質を腹腔内注射して、最終的な抗原ブーストを行います。3日間の注射後、マウスから脾臓を採取し、DMEMで2回洗浄して血液と脂肪細胞を除去します。200メッシュの銅グリッドを使用して脾臓細胞懸濁液をろ過して組織の破片を取り除き、遠心分離を使用して脾臓細胞を採取して脾臓膜を除去します。マウスSP20骨髄腫細胞を、6%ウシ胎児血清を添加した5ミリリットルのDMEMを含む25平方センチメートルのフラスコに播種し、細胞生存率を維持するために摂氏37度、二酸化炭素6%の雰囲気で細胞を培養します。5〜6日間の培養後、細胞は蘇生後に80〜90%のコンフルエンスに達し、顕微鏡下で丸く、明るく、透明に見えるはずです。ハイブリダイゼーションの前日に、マウスの腹腔からマクロファージを採取する。腹膜マクロファージを1ミリリットルあたり10〜5倍の0.1〜0.2倍の密度で、それぞれ100マイクロリットルのHAT培地を含む96ウェルプレートに播種し、一晩インキュベートします。ハイブリダイゼーションのために、SP20細胞を8〜10本のピペットで穏やかに吸引し、10ミリリットルの無血清DMEM培地に懸濁します。細胞を新鮮なDMEMで洗浄し、2回遠心分離してから、10ミリリットルのDMEMに再懸濁します。定量した脾臓細胞とSB20細胞を10:1の比率で混合し、50ミリリットルのチューブに移します。遠心分離後、上清を捨て、チューブの底にペレットを集めます。チューブをタップして、ハイブリダイゼーションの前にペレットを緩めます。スポイトを使用して、チューブの底を穏やかに回転させながら、緩んだセルペレットに摂氏37度に予熱したポリエチレングリコール1500を1ミリリットル加えます。次に、摂氏37度に予熱した1ミリリットルのDMEMを混合物に90秒間ゆっくりと加え、続いてさらに30ミリリットルの新しいDMEMを加え、融合管を摂氏37度の水浴に30分間入れます。インキュベーション後、細胞を収穫します。HAT培地を再懸濁し、腹膜マクロファージを接種した96ウェルプレートに培養します。5日後、100マイクロリットルの新鮮なHAT培地を各ウェルに加えます。そして再び5日間のインキュベーションの後、培地をHAT培地と交換する。0.05モルPBSで希釈した5マイクログラム/ミリリットルのAPNタンパク質でコーティングしたマイクロタイタープレートを使用して、ELISAアッセイを使用してハイブリドーマ上清中のモノクローナル抗体を分析します。pET28aと組換え抗体アミノペプチダーゼNBL21形質転換細菌を、0.4ミリモルのβ-d-1-チオガラクトピラノシドの存在下で、摂氏37度のオービタルシェーカーで10時間増殖させます。ウェルあたり100マイクロリットルの0.5倍10〜5番目のチャイニーズハムスター卵巣細胞を96ウェルプレートに播種し、摂氏37度、6%二酸化炭素雰囲気で18〜24時間インキュベートします。細胞が80〜90%コンフルエントに達したら、pIRES2-zs Green 1組換え抗体アミノペプチダーゼおよびプラスミドをOpti-MEMで最終濃度0.1マイクログラム/マイクロリットルに希釈します。5分後、50マイクロリットルの希釈pIRES2-zsグリーン1組換え抗体アミノペプチダーゼおよびプラスミドを1マイクロリットルのリポフェクタミン2000および49マイクロリットルのOpti-MEMと混合する。20分間のインキュベーション後、CHO細胞を含む96ウェルプレートの各ウェルに100マイクロリットルの混合物を加える。トランスフェクション後4〜6時間で、10%FBSを添加したDMEM F12で培地を交換します。48時間のインキュベーション後、各ウェルに400マイクログラム/マイクロリットルのG418を添加し、安定にトランスフェクトされた細胞を選択します。10%FBSおよび400マイクログラム当たりマイクロリットルG418を添加したDMEM F12培地を用いて10日間選択した後、蛍光活性化細胞選別で細胞を選別する。採取した陽性細胞を順次希釈する。96ウェルプレートにウェルあたり平均0.5〜2細胞で播種し、摂氏37度、6%二酸化炭素インキュベーターで培養します。代表的な顕微鏡分析では、すべてのハイブリドーマ細胞が丸く、明るく、透明に見えました。重い50キロダルトン鎖と軽い25キロダルトンの鎖を有する精製モノクローナル抗体をSDS-PAGEで確認し、精製腹水中に見出した。培養上清および腹水中におけるこれらの抗APNモノクローナル抗体の力価をここに示す。マウスモノクローナル抗体アイソタイピングにより、クローン5B31、5B36、3C48、5C51、および6C56に由来する抗体は免疫グロブリンG2Bサブクラスを有し、APN 2A20は免疫グロブリンG2Aカッパ型抗体であり、モノクローナル抗体APN 3FD9、3F10、および10F3は免疫グロブリンM型および加工されたカッパ軽鎖に属することが明らかになりました。これらのモノクローナル抗体のほとんどは、APNの異なるエピトープを標的としているのに対し、APN 5C51抗体はAPN 3C48、5B31、および6C56モノクローナル抗体によって認識されるような抗原エピトープを認識していることを示す50%を超えるAV値を示しました。増幅したAPN 5B36 VHVL遺伝子をpET28a陽性またはpIRES2-zsグリーン一ベクターにライゲーションし、pET28a陽性RABS APNおよびpIRES2-zsグリーン一RABS APNの組換え発現プラスミドをそれぞれ構築した。pET28aと組換え抗体アミノペプチダーゼNBL21およびpIRES2-zsグリーン一組換え抗体アミノペプチダーゼNCHO細胞の両方によって発現された抗体を精製し、ELISAおよびIFAアッセイを用いて分析した。しかし、pIRES2-zsグリーン一組換え抗体アミノペプチダーゼNCO細胞の上清で発現した抗体のみがAPNタンパク質を認識する。APN 5B36モノクローナル抗体のAPNタンパク質への結合は、組換え抗体よりも早く平衡に達した。プロテインAアガロースを用いた上清中のモノクローナル抗体の精製は、飽和硫酸アンモニア沈殿を用いるよりも優れている。
