메탈로프로테이나제 M1 계열에 속하는 달빛 효소인 아미노펩티다아제 N(APN)은 효소 의존성 및 효소 비의존성 경로를 통해 종양 표지자, 수용체 및 신호 분자로 작용합니다 1,2. APN은 생물학적 활성을 조절하기 위해 다양한 생리활성 펩타이드의 N-말단 아미노산 잔기를 절단하는 것 외에도 다양한 염증성 질환의 발병기전에 중요한 역할을 합니다. APN은 주요 조직 적합성 복합체 클래스 II 분자 2,3에 밀접하게 결합하는 절제된 펩타이드에 의한 항원 처리 및 제시에 참여합니다. APN은 또한 다중 신호 전달에 참여하는 G 단백질 결합 수용체와 결합하여 사이토카인 분비를 조절하고 면역 반응에서 Fc 감마 수용체 매개 식균 작용에 기여함으로써 항염증 효과를 발휘합니다 4,5,6,7.
널리 분포된 막 결합 엑소펩티다아제로서, APN은 돼지 소장 점막에 풍부하며 수용체 매개 엔도사이토시스(endocytosis)와 밀접한 관련이 있다 1,5,8. APN은 세포 진입을 위해 전염성 위장염 바이러스의 스파이크 단백질을 인식하고 결합하며, 장독소성 대장균 F4 섬유질의 FaeG 서브유닛과 직접 상호작용하여 숙주 세포에 대한 박테리아 부착에 영향을 미칩니다 9,10,11. 따라서, APN은 바이러스 및 세균성 감염성 질환의 치료에서 잠재적인 치료 표적이다.
1975년 하이브리도마 기술 및 단일클론항체(mAb) 생산을 위한 다른 전략의 개발 이후, mAbs는 면역요법, 약물 전달 및 진단에 널리 사용되어 왔다12,13,14. 현재, mAbs는 암, 염증성 장 질환 및 다발성 경화증과 같은 질병을 치료하는데 성공적으로 사용되고 있다12,15. 강한 친화력과 특이성으로 인해, mAbs는 항체-약물 접합체(ADC) 또는 새로운 백신의 개발에서 이상적인 표적이 될 수 있다16,17.
