Erratum: Tumor Transplantation for Assessing the Dynamics of Tumor-Infiltrating CD8+ T Cells in Mice

CD8+ T-cel-gemedieerde immuunrespons speelt een cruciale rol bij het beheersen van tumorgroei. Tijdens tumorigenese worden naïeve CD8+ T-cellen geactiveerd bij antigeenherkenning op een MHC klasse I-beperkte manier en differentiëren vervolgens in effectorcellen en infiltreren in tumormassa 1,2. Binnen de tumormicro-omgeving (TME) drijven langdurige blootstelling aan antigeen, evenals immunosuppressieve factoren, geïnfiltreerde tumorspecifieke CD8 + T-cellen echter in een hyporesponsieve toestand die bekend staat als "uitputting"3. Uitgeputte T-cellen (Tex) onderscheiden zich van effector- of geheugen-T-cellen die worden gegenereerd bij acute virale infectie, zowel transcriptioneel als epigenetisch. Deze Tex-cellen worden voornamelijk gekenmerkt door de aanhoudende en verhoogde expressie van een reeks remmende receptoren en het hiërarchische verlies van effectorfuncties. Verder resulteert de verminderde proliferatieve capaciteit van uitgeputte CD8+ T-cellen in afnemende aantallen tumorspecifieke T-cellen, zodanig dat de resterende CD8+ T-cellen in de TME nauwelijks voldoende beschermende immuniteit tegen tumorprogressie kunnen bieden3. Het behoud of de versterking van intratumorale antigeenspecifieke CD8+ T-cellen is dus onmisbaar voor tumorrepressie.
Bovendien wordt aangenomen dat immuun checkpoint blokkade (ICB) therapie Tex in tumoren nieuw leven inblaast door de infiltratie van T-cellen en dus T-celaantallen te verhogen en T-celfuncties te verjongen om tumorrepressie te stimuleren. De wijdverspreide toepassing van ICB-behandeling heeft het landschap van kankertherapie veranderd, met een aanzienlijke subgroep van patiënten die duurzame responsenervaren 4,5,6. Toch reageren de meeste patiënten en kankersoorten niet of slechts tijdelijk op ICB. Ontoereikende T-celinfiltratie in de TME is gepostuleerd als een van de onderliggende mechanismen die verantwoordelijk zijn voorICB-weerstand 7,8.
Verschillende studies hebben de heterogeniteit van tumor-infiltrerende CD8+ T-cellen (TILs) aangetoond bij zowel patiënten als muismodellen 9,10,11,12. Het is bevestigd dat een subset van CD8 + T-cellen die T-celfactor-1 (TCF1) in een tumormassa tot expressie brengen, stamcelachtige eigenschappen vertoont, die verder aanleiding kunnen geven tot terminaal uitgeputte T-cellen en verantwoordelijk is voor de proliferatie-uitbarsting na ICB-therapie 12,13,14,15,16,17,18,19,20, 21,22. Het is echter bewezen dat slechts een klein deel van de antigeenspecifieke TCF1+CD8+ T-cellen in de TME aanwezig is en een uitgebreide pool van gedifferentieerde nakomelingen genereert als reactie op ICB 23,24,25,26. Of de beperkte omvang van deze populatie voldoende is om de persistentie van cytotoxische T-lymfocyten (CTL's) te garanderen om tumorprogressie onder controle te houden, blijft onbekend en of er aanvulling is vanuit periferieweefsels vereist verder onderzoek. Bovendien suggereert recent onderzoek de onvoldoende revitaliseringscapaciteit van reeds bestaande tumorspecifieke T-cellen en het verschijnen van nieuwe, voorheen niet-bestaande clonotypen na anti-geprogrammeerde celdoodeiwit 1-behandeling. Dit geeft aan dat de T-celrespons op checkpointblokkade te wijten kan zijn aan de nieuwe instroom van een duidelijk repertoire van T-celklonen27. Samen met de aanwezigheid van omstanders niet-tumor-reactieve cytotoxische T-celfractie in de TME, leidden deze bevindingen tot de oprichting van een tumor allograftmodel om de rol van periferie-afgeleide CD8 + T-cellen11 te bestuderen.
Tot nu toe werden verschillende soorten tumorimplantatie, evenals immuuncel adoptieve overdracht, op grote schaal gebruikt op het gebied van tumorimmunologie28. TILs, perifere mononucleaire bloedcellen en tumorreactieve immuuncellen afkomstig uit andere weefsels kunnen goed worden gekarakteriseerd met behulp van deze methoden. Bij het bestuderen van de interacties tussen systemische en lokale antitumorimmuniteit lijken deze modellen echter ontoereikend om de interacties tussen immuuncellen afgeleid van de periferie en de TME te onderzoeken. Hier werden tumorweefsels getransplanteerd van donoren in tumor-gematchte ontvangermuizen om de instroom van van de ontvanger afgeleide immuuncellen nauwkeurig te traceren en de van de donor afgeleide cellen in de TME tegelijkertijd te observeren.
In deze studie werd een murine syngenetisch model van melanoom vastgesteld met de B16F10-OVA melanoom cellijn, die stabiel het surrogaat neoantigeen ovalbumine tot expressie brengt. TCR transgene OT-I muizen, waarbij meer dan 90% van de CD8+ T cellen specifiek het OVA-afgeleide peptide OVA257-264 (SIINFEKL) herkennen gebonden aan het klasse I MHC molecuul H2-Kb, maken de studie van antigeenspecifieke T-celresponsen mogelijk die zijn ontwikkeld in het B16F10-OVA tumormodel. Door dit model te combineren met tumortransplantatie, werden de immuunresponsen van tumor-inherente en periferie-afkomstige antigeen-specifieke CD8 + T-cellen vergeleken om een dynamische overgang tussen deze twee populaties te onthullen. Gezamenlijk heeft dit experimentele ontwerp een andere benadering geboden om de immuunresponsen van CD8 + T-cellen in de TME nauwkeurig te onderzoeken, wat nieuw licht werpt op de dynamiek van tumorspecifieke T-celimmuunresponsen in de TME.

tumor transplantation for assessing the dynamics of tumor-infiltrating cd8+ t cells in mice

T-cel-gemedieerde immuniteit speelt een cruciale rol in immuunresponsen tegen tumoren, waarbij cytotoxische T-lymfocyten (CTL's) de hoofdrol spelen bij het uitroeien van kankercellen. De oorsprong en aanvulling van tumorantigeenspecifieke CD8+ T-cellen in de tumormicro-omgeving (TME) blijven echter onduidelijk. Dit protocol maakt gebruik van de B16F10-OVA melanoom cellijn, die stabiel het surrogaat neoantigeen, ovalbumine (OVA) en TCR transgene OT-I muizen tot expressie brengt, waarin meer dan 90% van de CD8 + T-cellen specifiek het OVA-afgeleide peptide OVA257-264 (SIINFEKL) herkennen dat is gebonden aan het klasse I major histocompatibiliteitscomplex (MHC) molecuul H2-Kb. Deze kenmerken maken de studie van antigeenspecifieke T-celresponsen tijdens tumorigenese mogelijk.
Door dit model te combineren met tumortransplantatiechirurgie, werden tumorweefsels van donoren getransplanteerd in tumor-gematchte syngenetische ontvangermuizen om de instroom van van de ontvanger afgeleide immuuncellen in getransplanteerde donorweefsels nauwkeurig te traceren, waardoor de immuunresponsen van tumor-inherente en periferie-afkomstige antigeen-specifieke CD8 + T-cellen. Er bleek een dynamische overgang plaats te vinden tussen deze twee populaties. Gezamenlijk heeft dit experimentele ontwerp een andere benadering geboden om de immuunresponsen van CD8 + T-cellen in TME nauwkeurig te onderzoeken, wat nieuw licht zal werpen op de tumorimmunologie.

Het schema van dit protocol is weergegeven in figuur 1. Acht dagen na tumorinenting werden CD45.1+ en CD45.1+CD45.2+ OT-I cellen geïnjecteerd in B16F10-OVA tumordragende C57BL/6 muizen. Tumor Transplantation for Assessing the Dynamics of Tumor-Infiltrating CD8+ T Cells in Mice

Hier presenteren we een tumortransplantatieprotocol voor de karakterisering van tumor-inherente en periferie-afgeleide tumor-geïnfiltreerde lymfocyten in een muistumormodel. Specifieke tracering van de instroom van van de ontvanger afgeleide immuuncellen met flowcytometrie onthult de dynamiek van de fenotypische en functionele veranderingen van deze cellen tijdens antitumorimmuunresponsen.

Tumortransplantatie voor het beoordelen van de dynamiek van tumorinfiltrerende CD8 + T-cellen bij muizen

Deze methode kan helpen bij het beantwoorden van de belangrijkste vragen in de tumorimmunologie, waaronder of er een aanvulling is van tumorspecifieke immuuncellen uit perifere weefsels, de overgangen tussen nieuw geïnfiltreerde en reeds bestaande immuuncellen in de tumormicro-omgeving en hun reacties op immunotherapie. Dit is een handige en gemakkelijk te volgen techniek om de van de periferie afgeleide cellen te onderscheiden van tumor-geïnfiltreerde immuuncellen en de dynamische, fenotypische en functionele veranderingen van deze cellen in longitudinale assays te volgen. Begin met het opzuigen van 100 microliter van de bereide B16F10-OVA-celsuspensie in een tuberculinespuit van 1 ml.Tik op de loop om eventuele bubbels naar boven te verplaatsen en druk zachtjes op de zuiger om luchtbellen te verwijderen. Houd de muis in bedwang om zijn buik bloot te leggen. Druk met de pink op het linkerachterbeen om de huid van het linker liesgebied strakker te maken.Verwijder het haar van de muis uit de linker onderbuik met een elektrisch scheerapparaat. Gebruik katoen gedrenkt in 75% ethanol om het achterste kwadrant van de linkerbuik schoon te maken. Houd de spuit in een ondiepe hoek tussen 0 en 15 graden en breng deze, met de schuine kant van de naald naar boven gericht, in aan de zijkant van de linker bovenbeen.Breng de naald 0,5 tot 1 centimeter door het onderhuidse weefsel naar het liesgebied. Trek de zuiger terug voordat u injecteert. Als er negatieve druk is, drukt u de zuiger volledig in en observeert u de vorming van een kleine vloeistofzak of een bolus in de subcutis.Verwijder de naald nadat de injectie is uitgevoerd. Laat de muis los en plaats hem terug in de kooi. Meet op dag zes tot acht na B16F10-OVA-implantatie de tumorgrootte met een vernierschaal.Selecteer de muizen met een diameter van ongeveer drie millimeter of een tumor ter grootte van een mungboon en verdeel ze gelijk en willekeurig in twee groepen. Zuig 200 microliter OT-1-celsuspensie op in een 29-gauge insulinespuit met 100 eenheden en verwijder bubbels. Plaats de muis apart in een kooi, met een infraroodlamp over de kooi gedurende 5 tot 10 minuten om de staartader te verwijden.Immobiliseer de muis met een fixatieapparaat van de juiste grootte. Houd de staart vast om hem recht te trekken en spuit 75% ethanol om de ader zichtbaar te maken. Houd de spuit parallel aan de ader en steek deze in de ader onder een hoek van 0 tot 15 graden.Trek de zuiger iets terug en als er bloed in de loop komt, injecteer dan langzaam en gestaag de suspensie met een snelheid van niet meer dan één milliliter per minuut. Nadat de injectie is voltooid, verwijdert u de spuit en drukt u gedurende drie tot vijf seconden zachtjes op het injectiegebied om het bloeden te stoppen. Breng de muis terug naar de kooi en observeer deze een paar minuten nauwlettend op bijwerkingen.Als het normale mobiliteit en nasale afscheiding heeft, plaats het dan terug in het gezelschap van de andere muizen. Selecteer 8 tot 10 dagen na de adoptieve overdracht donormuizen met een vergelijkbare tumormassa van ongeveer vijf millimeter diameter voor transplantatiechirurgie. Plaats een schaal van 100 millimeter bij 20 milliliter in een bioveiligheidskast en voeg 10 milliliter steriele, ijskoude PBS toe.Na het euthanaseren van de muis, dompel het onder in 75% ethanol gedurende drie tot vijf minuten. Plaats de muis in rugligging op een dissectieplank bedekt met schoon absorberend papier in de bioveiligheidskast. Houd de muisledematen in bedwang met dissectienaalden.Knip de huid langs de middellijn van boven de urethrale opening naar de xiphoid met een schaar. Rek de huid naar de linkerkant van het muizenlichaam met een pincet en houd de huid in bedwang met dissectienaalden. Snijd de tumor weg en houd de capsule zo intact mogelijk.Verwijder voorzichtig en voorzichtig het bindweefsel in de buurt van de tumor met een chirurgische schaar en plaats het tumorweefsel vervolgens in de schaal met steriele, ijskoude PBS voor daaropvolgende transplantatie. Gebruik veterinaire zalf op de ogen om te voorkomen dat ze uitdrogen. Scheer de linkerflank van de muis met een elektrisch scheerapparaat en breng vervolgens een ontharingscrème aan om het resterende haar te verwijderen.Plaats de muis in de bioveiligheidskast in buikligging op een dissectiebord bedekt met schoon absorberend papier, met de verticale as van de muis evenwijdig aan en de kop aan de rechterkant van het experimenteermiddel. Wrijf de huid van het geschoren gebied met katoen gedrenkt in povidon-jodium. Til de huid op het middelpunt tussen de heupgewrichten van de muis op met een chirurgisch pincet.Gebruik de schaar om een vijf millimeter lange verticale excisie te maken en verleng de snede rostrally langs de dorsale middellijn tot ongeveer 10 tot 15 millimeter. Voer een scherpe dissectie uit door de gesloten uiteinden van de schaar in de incisie in te brengen en vervolgens te openen om het peritoneum van de linkerflank van de huid en het zachte weefsel te scheiden. Voer meerdere keren een scherpe dissectie uit om een huidzak op de linkerflank te genereren.Deponeer de ingekapselde, intacte, van donor afgeleide tumormassa in de zak. Sluit de incisie door een onderbroken hechting, met behulp van twee of drie hechtingen voor elke incisie. Desinfecteer de huid rond de snee met katoen gedrenkt in povidon-jodium.Plaats de muis in de zijwaartse positie in een schone en warme kooi. Houd het continu in de gaten totdat het voldoende bij bewustzijn is gekomen om de sternale lighouding te behouden. Dien buprenorfine subcutaan toe in een dosis van 0,1 milligram per kilogram lichaamsgewicht om de acht uur, voor een totaal van drie doses.Breng de ontvanger van de transplantatie pas terug naar het gezelschap van andere dieren nadat deze volledig is hersteld. Met behulp van flowcytometrie werden twee populaties van CD44-negatieve en CD8-positieve tumorantigeenspecifieke T-cellen gemakkelijk geïdentificeerd in de tumormicro-omgeving, waaronder CD45.1-positieve donor-afgeleide en CD45.1-negatieve en CD45.2-positieve ontvanger-afgeleide tumor-infiltrerende CD8-positieve T-cellen. Op dag twee na de transplantatie waren er ongeveer 83% van de donor-afgeleide antigeen-specifieke CD8-positieve T-cellen in de getransplanteerde tumor, meer overheersend dan hun van de ontvanger afgeleide tegenhangers.Het aandeel van de ontvanger-afgeleide OT-1-cellen was verhoogd in het late stadium van tumorigenese, meer dan tumor-inherente OT-1-cellen afgeleid van de donor. Het belangrijkste om te onthouden is dat het uitvoeren van een zachte dissectie tijdens subcutane tumortransplantatie om verwondingen aan omliggende weefsels te voorkomen, vooral in korrelige lymfeklieren.
