Leishmaniasis er en forsømt tropisk sygdom forårsaget af protozoiske parasitter af slægten Leishmania, hvilket resulterer i et bredt spektrum af kliniske manifestationer hos hvirveldyrværten, herunder kutan leishmaniasis, mucokutan leishmaniasis og visceral leishmaniasis1. Verdenssundhedsorganisationen (WHO) anslår, at over en milliard mennesker er i fare, med mere end en million nye tilfælde rapporteret om år2.
Leishmania spp. er obligatoriske intracellulære protozoer, der overlever inde i værtsceller, herunder monocytter, makrofager og dendritiske celler3. Leishmania-makrofaginteraktion er en kompleks proces, der involverer flere værtscellereceptorer og parasitligander enten gennem direkte interaktion eller ved opsonisering, der involverer komplementreceptorer 4,5. Klassiske overfladereceptorer, såsom CR1, CR3, mannose-fucose, fibronectin, toll-lignende og scavenger receptorer, medierer parasitbinding til makrofager 6,7,8. I modsætning hertil danner andre arter, såsom L. braziliensis og L. infantum, mindre vakuoler, der normalt kun indeholder en eller to parasitter i hver vakuol18.
På trods af denne viden omkring værtscelle-Leishmania-interaktion er de indledende begivenheder udløst af kontakt mellem værtsreceptorer og parasitligander ikke blevet fuldt ud belyst. Disse hændelser vides at være determinanter for resultatet af parasitinfektion og er afhængige af parasitarter, typen af værtscellereceptorer, der rekrutteres til at genkende parasitter og aktiveringen af makrofagsignaleringsveje19,20. Derfor er det vigtigt at identificere de molekyler, der er involveret i biogenesen af Leishmania-inducerede PV'er, og bestemme den eller de roller, som disse molekyler spiller i infektionsetablering og -resultat. Her beskriver vi en metode til overvågning af tidlige begivenheder, der forekommer under fagocytose af Leishmania, herunder binding, internalisering, fagosomdannelse og modning. Dette arbejde kunne hjælpe med at afklare PLC's, Akts, Rab5s, Rab7' og LC3's deltagelse i dannelsen af PC'er induceret af forskellige Leishmania-arter. Fagocytose er en orkestreret proces, der involverer forskellige trin: anerkendelse, binding og internalisering. Professionelle fagocytter optager Leishmania-parasitter ved fagocytose, der består i at genkende ligander på parasitoverflader af flere værtscellereceptorer. Binding af Leishmania til makrofagmembraner sker gennem komplementreceptor type 1 (CR1) og komplementreceptortype 3 (CR3) og mønstergenkendelsesreceptorer. Lipophosphoglycan (LPG) og 63 kDa glycoprotein (gp63) er de vigtigste ligander involveret i makrofag-Leishmania interaktioner. Efter den første anerkendelse af parasitligander af værtscellereceptorer bliver parasitter internaliseret, overlever og formerer sig inden for parasitoforiske vakuoler. Modningsprocessen af Leishmania-inducerede vakuoler involverer erhvervelse af molekyler fra intracellulære vesikler, herunder monomert G-protein Rab 5 og Rab 7, lysosomalt associeret membranprotein 1 (LAMP-1), lysosomalt associeret membranprotein 2 (LAMP-2) og mikrotubuli-associeret protein 1A / 1B-lyskæde 3 (LC3).
Her beskriver vi metoder til at evaluere de tidlige begivenheder, der forekommer under Leishmania-interaktion med værtscellerne ved hjælp af konfokal mikroskopi, herunder (i) binding (ii) internalisering og (iii) fagosommodning. Ved at tilføje til mængden af viden omkring disse determinanter for infektionsresultat håber vi at forbedre forståelsen af patogenesen af Leishmania-infektion og støtte den eventuelle søgning efter nye kemoterapeutiske mål.

Her beskriver vi metoder til at evaluere de tidlige hærdningstilstande under Leishmania-interaktion med værtscellerne. Denne teknik præsenterer som den største fordel, der tillader undersøgelsen af forskellige tilstande af Leishmania fagocytose og deltagelse af flere proteiner.Det er værd at bemærke, at denne teknik også kan udvides til andre typer undersøgelser, herunder infektion med bakterier, gær eller det være sig opslugning af andre typer celler. Til at begynde med frø 0,2 millioner THP-1-celler eller humane monocytafledte makrofager i 500 mikroliter RPMI pr. Brønd på en 24 brøndplade med 13 millimeter glasdæksler. Inkuberes i 24 timer ved 37 grader Celsius under 5% kuldioxid, vask celler to gange med 0,9% saltvand og inkuber cellerne i et komplet RPMI-medium.Tilsæt stationær fase Leishmania-arter til cellerne ved en 10 parasit er til et værtscelleforhold, og centrifugeres derefter ved 720 gange g i 10 minutter ved fire grader Celsius. Efter inkubation af cellerne ved fire grader Celsius i fem minutter, vask cellerne to gange med 0,9% saltopløsning for at fjerne eventuelle ikke-internaliserede promastigotes. Inkuber cellerne i suppleret RPMI Medium ved 37 grader Celsius i en time, og fastgør derefter cellerne med 4% PFA i 15 minutter.Monter derefter dæksler ved hjælp af foretrukne monteringsmedier. Derefter centrifugeres ved 250 gange g i fem minutter ved stuetemperatur. Derefter resuspenderes THP-1-cellerne i en milliliter RPMI Medium og udfører tællinger i et Neubauer-kammer.Centrifuger cellerne igen ved 250 gange g i 10 minutter ved stuetemperatur og kassér supernatanten. Resuspender 2 millioner celler i 100 mikroliter Nucleofector-opløsning og inkuberes med 0,5 mikrogram plasmidbelægning til proteinet af interesse, mærket med et fluorescerende protein. Suspensionen indeholdende THP-1-celler og nukleinsyre overføres til nukleofektorkuvetten.Derefter transficeres cellerne ved hjælp af Nucleofector-program Y one. Rengør dækslerne med absorberende væv. Tilsæt derefter en dråbe nedsænkningsolie til målet, og tilsæt derefter diaset.Flyt målet op, indtil olien rører rutsjebanen. Overhold og juster fokus på mikroskopet og vælg mulighed 63x mål med olie. Åbn LIQA-programmet, og juster laserne i bølgelængderne 488, 552 og 405.Vælg billedopløsningen som 1024 x 1024. Vent på billedoptagelsen, og vælg derefter indstillingen, maksimal projektion, i værktøjerne.Gem eksperimentet, og eksporter billeder i TIFF-format til en computer. Resultaterne viste, at både Leishmania Braziliensis LCL og Leishmania Braziliensis DL ligeledes binder sig til makrofager. Der blev heller ikke observeret nogen forskelle med hensyn til Leishmania Braziliensis LCL og Leishmania Braziliensis DL fagocytose af værtsceller.Rekrutteringen af LC3 til parasitophorous vakuol eller PV'er, induceret af Leishmania Braziliensis LCL eller Leishmania Braziliensis DL, blev sammenlignet i inficerede celler. Efter fire timers infektion blev der observeret lignende procentdele af LC3-dekorerede PV'er i Leishmania Braziliensis LCL og Leishmania Braziliensis DL-inficerede makrofager. Resultaterne viste således, at Leishmania Braziliensis LCL og Leishmania Braziliensis DL på samme måde interagerer med makrofager under binding, fagocytose og biogenese af PV'er vedrørende LC3-rekrutteringen.Repræsentative mikroskopiske billeder demonstrerede effektiv transfektion af THP-1-celler med PLC-GFP, Rab5-GFP, Rab7-GFP-plasmider.
