Leishmaniasis is een verwaarloosde tropische ziekte veroorzaakt door protozoaire parasieten van het geslacht Leishmania, resulterend in een breed spectrum van klinische manifestaties in de gewervelde gastheer, waaronder cutane leishmaniasis, mucocutane leishmaniasis en viscerale leishmaniasis1. De Wereldgezondheidsorganisatie (WHO) schat dat meer dan een miljard mensen risico lopen, met meer dan een miljoen nieuwe gevallen gemeld per jaar2.
Leishmania spp. zijn obligate intracellulaire protozoën die overleven in gastheercellen, waaronder monocyten, macrofagen en dendritische cellen3. Leishmania-macrofaaginteractie is een complex proces waarbij meerdere gastheercelreceptoren en parasietliganden betrokken zijn, hetzij door directe interactie of door opsonisatie waarbij complementreceptoren betrokken zijn 4,5. Klassieke oppervlaktereceptoren, zoals CR1, CR3, mannose-fucose, fibronectine, tolachtige en aaseterreceptoren, bemiddelen parasitaire aanhechting aan macrofagen 6,7,8. Deze receptoren herkennen moleculen op het oppervlak van Leishmania, waaronder het 63 kDa glycoproteïne (gp63) en glycolipide lipofosfosfornaat (LPG)9. Dit zijn de meest voorkomende moleculen op het oppervlak van promastigoten en spelen een essentiële rol in de ondermijning van de immuunrespons van de gastheer, waardoor de vestiging van parasietinfectie in zoogdiercellenwordt bevorderd 10. Nadat parasiet oppervlakteliganden binden aan macrofaagreceptoren, hoopt F-actine zich op op zoogdierceloppervlakken, die parasieten omringen terwijl ze gefagocyteerd zijn. Vervolgens leidt dit tot de vorming van een parasiet-geïnduceerd compartiment genaamd parasitophorous vacuole (PV), dat fagolysosomale kenmerken vertoont11. Eenmaal binnen deze fagolysosomen ondergaan parasieten verschillende veranderingen die essentieel zijn voor overleving en vermenigvuldiging3.
De biogenese van PV's is een sterk gereguleerd membraanhandelproces dat cruciaal is voor de intracellulaire overleving van deze ziekteverwekker12. De vorming van dit compartiment is het gevolg van opeenvolgende fusiegebeurtenissen tussen fagosomen en compartimenten van de endocytische route van de gastheer. Klassieke celbiologische studies hebben aangetoond dat de rijping van PV's de verwerving van monomere G-eiwitten Rab 5 en Rab 7-eiwitten omvat, die voornamelijk geassocieerd zijn met vroege en late endosoomrijping, respectievelijk13. Bovendien verwerven deze compartimenten lysosoom-geassocieerde membraaneiwitten 1 en 2 (LAMP 1, LAMP 2), de belangrijkste eiwitbestanddelen van het lysosomale membraan en microtubule-geassocieerd eiwit 1A/1B-lichte keten 3 (LC3), een autofagosoommarker14. Ondanks schijnbare overeenkomsten variëren de kinetiek van PV-formatie15,16 en de morfologie van deze compartimenten afhankelijk van Leishmania-soorten. Infectie veroorzaakt door L. mexicana of L. amazonensis induceert bijvoorbeeld de vorming van grote compartimenten met een groot aantal parasieten17. Andere soorten, zoals L. braziliensis en L. infantum, vormen daarentegen kleinere vacuolen die normaal gesproken slechts één of twee parasieten bevatten in elke vacuole18.
Ondanks deze kennis rond gastheercel-Leishmania interactie, zijn de initiële gebeurtenissen veroorzaakt door contact tussen gastheerreceptoren en parasietliganden niet volledig opgehelderd. Van deze gebeurtenissen is bekend dat ze determinanten zijn van de uitkomst van parasietinfectie en zijn afhankelijk van parasietsoorten, het type gastheercelreceptoren dat wordt gerekruteerd om parasieten te herkennen en de activering van macrofaagsignaleringsroutes19,20. Daarom is het essentieel om de moleculen te identificeren die betrokken zijn bij de biogenese van door Leishmania geïnduceerde PV's en de rol (en) te bepalen die deze moleculen spelen bij het vaststellen en uitpakken van infecties. Hier beschrijven we een methode voor het monitoren van vroege gebeurtenissen die optreden tijdens de fagocytose van Leishmania, inclusief binding, internalisatie, fagosoomvorming en rijping. Dit werk zou kunnen helpen bij het verduidelijken van de deelname van PLC, Akt, Rab5, Rab7 en LC3 aan de vorming van PV's geïnduceerd door verschillende Leishmania-soorten. Belangrijk is dat dit protocol kan worden gebruikt om de deelname van andere eiwitten die betrokken zijn bij PV-rijping te onderzoeken. Toekomstige studies zullen de kennis uitbreiden rond mechanismen die betrokken zijn bij leishmania-gastheer celinteractie en bijdragen aan het ontwerp van nieuwe chemotherapeutische strategieën.

Fagocytose is een georkestreerd proces dat verschillende stappen omvat: herkenning, binding en internalisatie. Professionele fagocyten nemen Leishmania-parasieten op door fagocytose, bestaande uit het herkennen van liganden op parasietoppervlakken door meerdere gastheercelreceptoren. Binding van Leishmania aan macrofaagmembranen vindt plaats via complementreceptor type 1 (CR1) en complementreceptor type 3 (CR3) en patroonherkenningsreceptoren. Lipofosfosfaliek (LPG) en 63 kDa glycoproteïne (gp63) zijn de belangrijkste liganden die betrokken zijn bij macrofaag-Leishmania interacties. Na de eerste herkenning van parasietliganden door gastheercelreceptoren, worden parasieten geïnternaliseerd, overleven en vermenigvuldigen zich in parasitofore vacuolen. Het rijpingsproces van leishmania-geïnduceerde vacuolen omvat de verwerving van moleculen uit intracellulaire blaasjes, waaronder monomeer G-eiwit Rab 5 en Rab 7, lysosomaal geassocieerd membraaneiwit 1 (LAMP-1), lysosomaal geassocieerd membraaneiwit 2 (LAMP-2) en microtubule-geassocieerd eiwit 1A / 1B-lichtketen 3 (LC3).
Hier beschrijven we methoden om de vroege gebeurtenissen te evalueren die optreden tijdens leishmania-interactie met de gastheercellen met behulp van confocale microscopie, waaronder (i) binding (ii) internalisatie en (iii) fagosoomrijping. Door toe te voegen aan de hoeveelheid kennis rond deze determinanten van infectie-uitkomst, hopen we het begrip van de pathogenese van Leishmania-infectie te verbeteren en de uiteindelijke zoektocht naar nieuwe chemotherapeutische doelen te ondersteunen.

Het mechanisme geassocieerd met fagocytose bij Leishmania-infectie blijft slecht begrepen. Hier beschrijven we methoden om de vroege genezingstoestanden tijdens Leishmania-interactie met de gastheercellen te evalueren. Deze techniek presenteert als het belangrijkste voordeel, waardoor de studie van verschillende toestanden van Leishmania fagocytose en de deelname van verschillende eiwitten mogelijk is.Het is vermeldenswaard dat deze techniek ook kan worden uitgebreid naar andere soorten studies, waaronder infectie door bacteriën, gisten of overspoeling door andere soorten cellen. Mensen die deze techniek proberen, moeten zorgen voor de blootstellingstijd van de cellen aan de Nucleofector-oplossing. Daarnaast stellen we voor om transfectie te testen met maximaal twee plasmiden.De procedure wordt gedemonstreerd door Amanda Reboucas, een promovenda uit ons laboratorium. Om te beginnen zaai je 0,2 miljoen THP-1-cellen, of van menselijke monocyten afgeleide macrofagen in 500 microliter RPMI per put op een 24-putplaat met 13 millimeter glazen coverslips. Incubeer gedurende 24 uur bij 37 graden Celsius onder 5% koolstofdioxide, was cellen twee keer met 0,9% zoutoplossing en incubeer de cellen in een compleet RPMI-medium.Voeg stationaire fase Leishmania-soorten toe aan de cellen bij een 10-parasiet is tot één gastheercelverhouding en centrifugeer vervolgens op 720 keer g gedurende 10 minuten bij vier graden Celsius. Na het incuberen van de cellen op vier graden Celsius gedurende vijf minuten, wast u de cellen tweemaal met 0,9% zoutoplossing om niet-geïnternaliseerde promastigoten te verwijderen. Incubeer de cellen in aangevuld RPMI Medium bij 37 graden Celsius gedurende een uur en fixeer de cellen vervolgens met 4% PFA gedurende 15 minuten.Monteer vervolgens coverslips met behulp van de gewenste montagemedia. Tel vervolgens ten minste 400 cellen in willekeurige velden onder een fluorescentiemicroscoop. Om de door Leishmania geïnduceerde PV-biogenese te onderzoeken, begint u met het transfecteren van THP-1-cellen met plasmide door 15 miljoen THP-1-cellen te zaaien in weefselkweekkolven van 75 vierkante centimeter met 10 milliliter Complete RPMI Medium aangevuld met 100 nanogram per milliliter PMA en 50 micromolaire tot mercaptoethanol gedurende 48 uur.Was vervolgens de cellen eenmaal in een zoutoplossing van 0,9% en maak de cellen los met behulp van een niet-enzymatische celdissociatieoplossing. Centrifugeer vervolgens op 250 maal g gedurende vijf minuten bij kamertemperatuur. Resuspendeer vervolgens de THP-1-cellen in één milliliter RPMI Medium en voer tellingen uit in een Neubauer-kamer.Centrifugeer de cellen opnieuw op 250 maal g gedurende 10 minuten bij kamertemperatuur en gooi het supernatant weg. Resuspendeer 2 miljoen cellen in 100 microliter Nucleofector-oplossing en incubeer met 0,5 microgram van de plasmidecoating voor het eiwit van belang, gelabeld met een fluorescerend eiwit. Breng de suspensie met THP-1-cellen en nucleïnezuur over naar het Nucleofector-cuvet.Vervolgens transfecteer je de cellen met behulp van Nucleofector-programma Y one. Herstel de getransfecteerde cellen en zaden in 500 microliter RPMI Medium op 24 putplaten met 13 millimeter glazen afdekplaten. Incubeer de cellen en complete RPMI Medium op 37 graden Celsius gedurende 0,5, 2, 4, 6, 12 en 24 uur.Laat de 13 millimeter glazen afdekplaten op kamertemperatuur staan en bescherm ze minstens 30 minuten voor de verwerving tegen het licht. Reinig de coverslips met absorberend weefsel. Voeg vervolgens een druppel dompelolie toe aan het doel en voeg vervolgens de dia toe.Verplaats het objectief omhoog totdat de olie de dia raakt. Observeer en pas de scherpstelling aan op de microscoop en kies optie 63x objectief met olie. Open het LIQA-programma en pas de lasers aan in de golflengten 488, 552 en 405.Selecteer de afbeeldingsresolutie als 1024 x 1024. Nadat u op de live-knop hebt geklikt, stelt u de Z-stack in en drukt u op de startoptie. Wacht op de afbeeldingsacquisitie en selecteer vervolgens de optie, maximale projectie, in de gereedschappen.Sla het experiment op en exporteer de TIFF-indelingsafbeeldingen naar een computer. De resultaten toonden aan dat zowel Leishmania Braziliensis LCL als Leishmania Braziliensis DL op dezelfde manier binden aan macrofagen. Ook werden geen verschillen waargenomen met betrekking tot Leishmania Braziliensis LCL en Leishmania Braziliensis DL fagocytose door gastheercellen.De rekrutering van LC3 naar de parasitofore vacuole, of PV's, geïnduceerd door Leishmania Braziliensis LCL of Leishmania Braziliensis DL, werd vergeleken in geïnfecteerde cellen. Na vier uur infectie werden vergelijkbare percentages LC3-versierde PV's in Leishmania Braziliensis LCL en Leishmania Braziliensis DL geïnfecteerde macrofagen waargenomen. De resultaten toonden dus aan dat Leishmania Braziliensis LCL en Leishmania Braziliensis DL op dezelfde manier interageren met macrofagen tijdens binding, fagocytose en biogenese van PV's met betrekking tot de LC3-rekrutering.Representatieve microscopische beelden toonden een efficiënte transfectie van THP-1 cellen met PLC-GFP, Rab5-GFP, Rab7-GFP plasmiden. Mensen die dit protocol proberen, moeten aandacht besteden aan temperatuurincubatie en de microscoopmonsters beschermen tegen het licht, alle soorten. Het moduleren van de expressie van de geïdentificeerde eiwitten die Leishmania fagocytose neerslaan, zal het belang van die eiwitten in de uitkomst van Leishmania-infecties aantonen.We kunnen deze techniek uitbreiden naar verschillende soorten pathologiestudies en mechanismen die verband houden met het opzetten van parasieten en doelidentificatie voor de ontwikkeling van therapeutische strategieën.
