Leishmaniasis er en forsømt tropisk sykdom forårsaket av protozoa parasitter av slekten Leishmania, noe som resulterer i et bredt spekter av kliniske manifestasjoner i vertebratverten, inkludert kutan leishmaniasis, mukokutan leishmaniasis og visceral leishmaniasis1. Verdens helseorganisasjon (WHO) anslår at over en milliard mennesker er i faresonen, med mer enn en million nye tilfeller rapportert per år2.
Leishmania spp. er obligatoriske intracellulære protozoer som overlever inne i vertsceller, inkludert monocytter, makrofager og dendrittiske celler3. Leishmania-makrofaginteraksjon er en kompleks prosess som involverer flere vertscellereseptorer og parasittligander enten gjennom direkte interaksjon eller ved opsonisering som involverer komplementreseptorer 4,5. Klassiske overflatereseptorer, som CR1, CR3, mannose-fucose, fibronektin, tolllignende og scavenger reseptorer, formidler parasittfeste til makrofager 6,7,8. Disse reseptorene gjenkjenner molekyler på overflaten av Leishmania, inkludert 63 kDa glykoprotein (gp63) og glykolipid lipofosfoglykan (LPG)9. Dette er de mest tallrike molekylene på overflaten av promastigoter og spiller en viktig rolle i undergravingen av vertsimmunrespons, og favoriserer etableringen av parasittinfeksjon i pattedyrceller10. Etter at parasittoverflateligander binder seg til makrofagreseptorer, akkumuleres F-aktin på pattedyrcelleoverflater, som omgir parasitter når de fagocytoseres. Biogenesen av PVs er en sterkt regulert membranhandelsprosess som er kritisk for den intracellulære overlevelsen av dette patogenet12. Dannelsen av dette rommet skyldes sekvensielle fusjonshendelser mellom fagosomer og rom i vertens endocytiske vei. Klassiske cellebiologiske studier har vist at modning av PVer innebærer oppkjøp av monomere G-protein Rab 5 og Rab 7 proteiner, som hovedsakelig er assosiert med henholdsvis tidlig og sen endosommodning, henholdsvis13. I tillegg får disse rommene lysosomassosierte membranproteiner 1 og 2 (LAMP 1, LAMP 2), de viktigste proteinbestanddelene i lysosomalmembranen og mikrotubuliassosiert protein 1A / 1B-lyskjede 3 (LC3), en autofagosommarkør14. Til tross for tilsynelatende likheter, varierer kinetikken til PV-dannelse15,16 og morfologien til disse rommene avhengig av Leishmania-arter. For eksempel induserer infeksjon forårsaket av L. mexicana eller L. amazonensis dannelsen av store rom som inneholder et stort antall parasitter17. Derimot danner andre arter, som L. braziliensis og L. infantum, mindre vakuoler som normalt bare inneholder en eller to parasitter i hver vacuole18.
Til tross for denne kunnskapen rundt vertscelle-Leishmania-interaksjon, har de første hendelsene utløst av kontakt mellom vertsreseptorer og parasittligander ikke blitt fullstendig belyst. Disse hendelsene er kjent for å være determinanter for utfallet av parasittinfeksjon og er avhengige av parasittarter, typen vertscellereseptorer rekruttert for å gjenkjenne parasitter og aktiveringen av makrofagsignalveier19,20. Derfor er det viktig å identifisere molekylene som er involvert i biogenesen av Leishmania-induserte PV-er og bestemme rollen (e) som disse molekylene spiller i infeksjonsetablering og utfall. Her beskriver vi en metode for å overvåke tidlige hendelser under fagocytose av Leishmania, inkludert binding, internalisering, fagosomdannelse og modning. Dette arbeidet kan bidra til å avklare deltakelsen av PLC, Akt, Rab5, Rab7 og LC3 i dannelsen av PVer indusert av forskjellige Leishmania-arter. Fagocytose er en orkestrert prosess som involverer forskjellige trinn: anerkjennelse, binding og internalisering. Profesjonelle fagocytter tar opp Leishmania-parasitter ved fagocytose, bestående av å gjenkjenne ligander på parasittoverflater av flere vertscellereseptorer. Binding av Leishmania til makrofagmembraner skjer gjennom komplementreseptortype 1 (CR1) og komplementreseptor type 3 (CR3) og mønstergjenkjenningsreseptorer. Lipofosfoglykan (LPG) og 63 kDa glykoprotein (gp63) er de viktigste ligandene som er involvert i makrofag-Leishmania-interaksjoner . Etter den første anerkjennelsen av parasittligander av vertscellereseptorer, blir parasitter internalisert, overlever og multipliserer innenfor parasitoforøse vakuoler. Etter å ha klikket på live-knappen, sett Z-stack og trykk på startalternativet. Folk som prøver denne protokollen må være oppmerksom på temperaturinkubasjon og beskytte mikroskopprøvene fra lyset, alle typer.
