変化する栄養ニーズに対応して、脂肪組織は、身体のニーズを満たすために脂質貯蔵または動員モードのいずれかを採用するためのエネルギーキャッシュとして機能します1。さらに、脂肪組織はまた、通性熱発生とも呼ばれる非震え熱発生と呼ばれるプロセスを介して、体温調節において重要な機能を果たしている。これは典型的には、褐色脂肪組織(BAT)によって達成され、豊富なレベルのミトコンドリア膜タンパク質脱共役タンパク質1(UCP1)を発現する。プロトンキャリアとして、UCP1はプロトン輸送とATP生成2を解離して発熱します。低温刺激を受けると、BATにおける熱発生は交感神経系の活性化(SNS)によって動き始め、続いてノルエピネフリン(NE)の放出が続く。NEはβ3アドレナリン作動性受容体に結合し、細胞内サイクリックAMP(cAMP)の上昇をもたらす。その結果、CREBのcAMP/PKA依存性の関与(cAMP応答エレメント結合タンパク質)は、CREB応答エレメント(CRE)2への直接結合を介してUcp1転写を刺激する。BATに加えて、褐色様脂肪細胞も白色脂肪組織内に見出されるため、ベージュまたはブライト(ブラウン・イン・ホワイト)細胞と命名される1,3。特定の刺激(寒さなど)に応答して、これらの静止ベージュ細胞は、多座性脂肪滴、密集したミトコンドリア、および増強されたUCP1発現を含む複数の茶色様の特徴を示すように改造される3、4、5。
動物実験では、褐色およびベージュ色の脂肪細胞が、インスリン感作、脂質低下、抗炎症、および抗アテローム性動脈硬化症を含む、脂肪減少効果を超えて複数の代謝利益を有することが実証されている6,7。ヒトでは、ベージュ色/褐色の脂肪の量は、年齢、インスリン抵抗性指数、および心臓代謝障害と反比例します8。さらに、低温馴化またはβ3アドレナリン受容体アゴニストのいずれかによるヒトにおけるベージュ色/褐色脂肪細胞の活性化は、一連の代謝障害に対する保護を与える4,9,10。これらの証拠は、褐色およびベージュ色の脂肪組織の誘導が肥満およびそれに関連する医学的合併症の管理のための潜在的な治療戦略であることを集合的に示している8。
興味深いことに、ベージュ色と古典褐色の脂肪細胞は同様の機能を共有していますが、異なる前駆体に由来し、重複しているが別個のメカニズムによって活性化されます1。したがって、褐色およびベージュ色の脂肪細胞のインビボイメージングおよび定量化は、これらの脂肪組織の分子制御のより良い理解を達成するために不可欠である。現在、18F-フルオロデオキシグルコース([18F]FDG)陽電子放射断層撮影(PET)スキャンとコンピュータ断層撮影法(CT)の組み合わせは、臨床研究における熱発生性褐色およびベージュ色の細胞の特性評価のゴールドスタンダードのままです8。磁気共鳴イメージング(MRI)は、強力な磁場と無線周波数パルスを使用して、詳細な解剖学的構造を生成します。CTスキャンと比較して、MRIはより高い解像度で臓器や軟部組織の画像を生成します。ここでは、低温曝露に順応した後のマウスモデルにおける機能性褐色およびベージュ脂肪の視覚化および定量化のためのプロトコルが提供され、脂肪の褐変を誘発する一般的で最も信頼性の高い方法である。この方法は、小動物モデルにおける熱発生性脂肪貯蔵所を高精度に特徴付けるために適用することができる。

PET/MRイメージングは、これらのいわゆる熱発生性脂肪細胞の活性を機能的に測定できるように、生きた動物におけるこれらの代謝活性な茶色およびベージュ色の脂肪細胞について定性的および定量的測定の両方を可能にする。この技術の主な利点は、同じ被験者に対するPETスキャンとMRIスキャンの取得と組み合わせを可能にし、小動物の異なるデポーにおける熱発生脂肪細胞の正確かつ同時取得を可能にすることです。この方法は、任意の前臨床システムに容易に転送したり、複数のマウスを同時に画像化することによってハイスループット形式に変更したりすることができ、それによって、画像化データの統計的検出力を低コストおよび時間で増加させることができる。肩甲骨間褐色脂肪組織を採取するには、麻酔をかけられた8週齢のC57 Black 6マウスを加熱パッドの上に置き、背側正中線に沿って2センチメートルの切開を行う。iBATパッドを肩甲骨間領域から取り外します。出血が止まったら、7ミリメートルのステンレス製の巻線クリップを使用して切開部を閉じます。すべてのiBATが収集されたら、マウスを集中治療室に14日間収容する。メロキシカム1kgあたり5ミリグラムをマウスに6日間投与し、創傷が治癒したらすぐにクリップを取り外す。イメージングセッションの前に自動シーケンシャルPET/MRスキャンを有効にするには、識別レベルを400~600キロ電子ボルト、信頼度モードを1~3、スキャン時間を20分、研究同位体をF-18に設定してPET取得のワークフローを設定します。減衰補正のためにT1強調磁気共鳴を取得するには、グラジエントエコー-3Dパラメータを設定します。解剖学的基準としてT2加重磁気共鳴を取得するには、ファストスピンエコー2Dパラメータを設定します。PET画像の再構成を可能にするには、Tera-Tomo 3Dアルゴリズムを使用して、一致モードを1〜3に設定し、減衰、デッドタイム、ランダム、減衰、および散乱補正を設定して、ボクセルサイズが0.3立方ミリメートルの全体的な画像を作成します。PET定量の精度をチェックするためにイメージング研究の開始の1日前に、適切なPPEを着用し、製造業者の推奨に従って5ミリリットルのシリンジにF18-FDGを充填する。用量校正器を使用してシリンジの活性を記録し、測定の時間をメモします。ソフトウェアでは、補間楕円ROIを使用して、シリンジの再構成画像上に関心のある体積を描画し、回復した活性を線量キャリブレータを使用して測定された値と比較します。イメージング実験当日、鉗子を使用して18F-FDGストックバイアルをLブロック卓上シールドの後ろに慎重に置き、放射性同位元素のアリコートを100〜150マイクロリットルの滅菌生理食塩水で200〜250マイクロキュリーの総活性濃度に希釈する。18F-FDG溶液を針を備えた1ミリリットルのシリンジに引き込み、実証されているように線量校正器で放射能を測定する。18F-FDG溶液を尾静脈に注入する前に、画像化するマウスの体重を記録する。シリンジの注入時間と残留放射能に注意して、崩壊補正を有効にします。その後、マウスをケージに60分間戻します。注射された18F-FDG活性を計算するには、注射後に測定された活性から注射前のシリンジ内の活性を差し引く。放射性同位元素の取り込み期間が終了したら、マイクロPET/MRスキャナーのマウスベッド用のエアヒーターをオンにします。麻酔をかけたFDG注射マウスをスキャナーベッドの呼吸パッドの上に置き、偵察ビューを実行してマウスの位置を決定した。全身が観察できるようにマウスベッドの位置を調整し、MRI視野の中心が体の中心と一致するようにします。[PET 取得] で、[前回の取得でスキャン範囲] を選択してスカウト表示の位置を使用し、[準備] をクリックして動物用ベッドを MR から PET に移動します。[OK] を選択して PET スキャンを開始します。18F-FDG投与前後で測定した注射用量と時間を放射性医薬品エディタに記録し、被験者情報メニューにマウスの体重を入力します。PETスキャンが完了したら、MRイメージングに移動し、試験リストウィンドウですべてのMR取得を完了するために準備を選択します。[OK] を選択して MR スキャンを開始します。ワークフロー全体が完了したら、後処理ソフトウェアで取得したMR画像の品質を簡単に評価し、[ホーム]をクリックしてマウスベッドをMRから元の位置に移動します。すべてのマウスの画像化が完了したら、[生のスキャン]メニューでデータを再構築するには、[PET集録]を選択して完了したPETスキャンをロードし、[T1加重MR取得]を選択して材料マップを作成します。次に、Tera-Tomo 3Dアルゴリズムを使用して、デモンストレーションに従ってデータを再構築します。低温処理前のiBATの除去は、マイクロPET/MRイメージングによってiWATで観察された18F-FDG取り込みの顕著な増加によって示されるように、iWATの活性を変化させる。さらに、ベージュ色の脂肪組織で特徴的に観察される多座性脂肪細胞は、偽手術動物からのiWATで観察された脂肪細胞形態と比較して、iBAT除去後のマウスからのiWATにおいてより顕著である。低温治療された偽手術マウスは、iBATにおける18F-FDGの取り込みが著しく上昇したことを実証する。低温治療前にiBATを除去したマウスは、iWATにおいて最も高い18F-FDG取り込みを示した。実際、寒冷暴露は偽手術マウスのiBATにおける18F-FDG取り込みの7倍の増加を引き起こすが、寒冷誘発マウスにおけるiBATの除去は、わずかな2倍の増加しか観察されないサーモニュートラルマウスと比較して、18F-FDG取り込みの8倍の増加をもたらす。この手順を試みるとき、各PET/MRI取得のために各マウスを同じ位置に配置することは、異なる領域における褐色およびベージュ脂肪細胞の可視化および定量を改善するために重要である。この方法を使用し、画像化や温度測定による拡散などの他のMRI法とともに使用すると、マウスにおける褐色脂肪細胞およびベージュ脂肪細胞の空間分布および有病率に関するより多くの洞察が開始され得、これは潜在的にヒトに翻訳され得る。この方法により、研究者はこれらの熱起源の茶色およびベージュ色の脂肪細胞の機能を研究することができます。したがって、これらの古典的なマーカーが改変またはアップレギュレートされない非臨床熱発生経路を研究する際に特に強力である。
