本研究では、hDPCRは、MSC表面マーカー検出のためのフローサイトメトリー、多系統分化電位、およびRT-qPCRにより、ステムネス特性および増殖性遺伝子マーカーの発現を決定することを特徴とする(データは示さない)8、9、10。hDDPCは、決定的な内胚葉、膵臓内胚葉、膵臓内分泌物、および膵臓β細胞またはIPC(図1)に誘導された(図1)、それぞれ7。細胞を誘導するために、3段階誘導アプローチをバックボーンプロトコルとして用いた。このプロトコルは非統合型プロトコルと呼ばれます。統合型プロトコルの場合、必須の膵転写因子PDX1は、hDDPCsで過剰発現し、続いて3段階分化プロトコルを用いてhDDPSCsで過発現PDX1誘導を行った。非統合型プロトコルと統合型プロトコルの違いは、非統合型プロトコルではなく、統合型プロトコルにおけるPDX1の過剰表現です。膵分化は、本研究における統合的プロトコルと非統合的プロトコルの間で比較された。
