국소 림프절 (센티넬 림프절)의 생검은 일반적으로 흑색종 세포의 존재를 확인하기 위해 수행됩니다. 그러나 초음파 영상은 임상 평가를 능가하고 불필요한 생검을 피하는 데 도움이 될 수 있기 때문에 전이성 개입을 탐지하는 비 침습적 방법으로 자리 잡고 있습니다2,3,4. 또한, 초음파 영상은 림프절 감시, 특히 고령 및/또는 합병증의 경우5,6에 적합한 것으로 보인다. 초음파 분석에 의해 검출되고 정상 림프절과 전이성 림프절 사이의 분화를 허용하는 특징은 크기 (부피) 증가, 타원형에서 둥글게의 모양 변화, 불규칙한 여백, 변경된 echogenic 패턴 및 변경된 (증가된) 혈관화7를 포함한다.

Braf/Pten 마우스는 흑색종 발생9,10에 관여하는 신호전달 경로의 해부, 기원의 흑색종 세포의 확인11,12,13, 표적 요법과 면역요법 모두에서 새로운 치료 옵션의 시험을 포함하여 여러 목적으로 광범위하게 사용되어 왔습니다.8,14,15,16 . 특히, 우리는 Braf / Pten 마우스를 사용하여 약독화 된 리스테리아 단세포 유전자 (Lmat)가 항 흑색종 백신으로 작용한다는 것을 입증했습니다. 치료 환경에서 전신적으로 투여될 때, Lmat은 종양 부위에 선택적으로 축적되기 때문에 전반적인 독성과 관련되지 않는다. 또한, 원발성 흑색종 질량의 현저한 감소와 림프절과 폐의 전이성 부담의 감소를 일으킨다. 분자 수준에서, Lmat은 흑색종 세포의 사멸을 일으키는데, 이는 적어도 부분적으로는 비세포-자율적 활동(CD4+ 및 CD8+ T 림프구의 현장 모집)16에 기인한다.

Tyr::CreER+, BrafCA/+,Ptenlox/lox 유전자 조작 마우스(Braf/Pten 마우스)는 전이성 흑색종의 생체내 모델로 널리 사용됩니다. 일단 원발성 종양이 타목시펜 처리에 의해 유도되면, 유도 후 4-6 주 이내에 전이성 부담의 증가가 관찰됩니다. 이 논문은 UHFUS(Ultra-High-Frequency UltraSound) 이미징을 활용하여 부피의 증가를 측정하여 사타구니 림프절의 전이성 관여 증가를 모니터링하는 방법을 보여줍니다.
UHFUS 시스템은 UHFUS 선형 프로브 (22-55 MHz, 축 분해능 40 μm)로 마취 된 마우스를 스캔하는 데 사용됩니다. 사타구니 림프절 (왼쪽과 오른쪽 모두)의 B 모드 이미지는 짧은 축보기로 수집되어 동물을 등쪽 난해부에 배치합니다. 초음파 기록은 전동 기계식 암에 44 μm 스텝 크기를 사용하여 수집됩니다. 그 후, 2차원(2D) B 모드 획득은 초음파 이미지 후처리를 위한 소프트웨어 플랫폼으로 가져오고, 획득한 횡단면 2D 이미지에서 사타구니 림프절을 반자동으로 식별하고 분할합니다. 마지막으로, 3차원(3D) 부피의 전체 재구성은 림프절 부피의 렌더링과 함께 자동으로 얻어지며, 이는 또한 절대 측정으로 표현된다.
