Haemophilus influenzae (Hi) is een normale commensale bacterie die aanwezig is in de keelholte van de meeste gezonde volwassenen. Hi kan een polysaccharidecapsule hebben (types A-F, bijvoorbeeld type B of HiB) of geen capsule hebben en niet-typebaar zijn (NTHi)1. Kolonisatie van het slijmvlies met deze bacterie begint in de vroege kindertijd en er is een omzet van verschillende koloniserende stammen2. Deze bacterie is ook in staat tot invasie van zowel de bovenste als de onderste luchtwegen; in deze context kan het activering van de immuunrespons en ontsteking induceren 3,4. Deze ontstekingsreactie kan klinische ziekten veroorzaken en bijdragen aan een verscheidenheid aan belangrijke ademhalingsaandoeningen, waaronder sinusitis, otitis media, bronchitis, cystische fibrose, longontsteking en chronische obstructieve longziekte (COPD). De meeste van deze aandoeningen zijn te wijten aan NTHi-stammen2. Dit artikel beschrijft methoden om respiratoire immuunresponsen op Hi te beoordelen met behulp van flowcytometrie en confocale microscopie.
De hieronder beschreven methoden zijn aangepast aan gevestigde technieken die zijn aangepast om de ontstekingsreactie op Hi te beoordelen. De selectie van een geschikte antigene vorm van Hi is een belangrijk onderdeel van deze beoordeling. Antigene preparaten variëren van celwandcomponenten tot levende bacteriën. Om assays vast te stellen en te standaardiseren, kan het gebruik van perifere bloedmonsters in eerste instantie zeer nuttig zijn.
Flowcytometrie maakt het mogelijk om een verscheidenheid aan parameters en functionele testen van één monster op cellulair niveau te meten. Deze techniek heeft het voordeel dat specifieke cellulaire responsen (bijv. productie van reactieve zuurstofsoorten (ROS) of intracellulaire cytokineproductie) kunnen worden beoordeeld in vergelijking met andere meer algemene methoden zoals enzyme-linked immunosorbent assay (ELISA) of ELISspot.
Extracellulaire vallen worden tot expressie gebracht door neutrofielen (NET's)5,6,7 en door andere cellen zoals macrofagen (METs)8. Ze worden steeds meer erkend als een belangrijke ontstekingsreactie, met name bij infectie in de long9. Ze kunnen worden beoordeeld met confocale fluorescentiemicroscopie. Deze techniek maakt definitieve identificatie van NET's/METs mogelijk en onderscheidt hun expressie van andere vormen van celdood6.
Zowel flowcytometrie als confocale microscopie zijn op fluorescentie gebaseerde assays. Hun succes is afhankelijk van optimale overbelastingsprotocollen van biologische monsters. Deze methoden hebben enige tijd nodig om te leren en vereisen de juiste toezichthoudende expertise. De betrokken instrumenten zijn ook duur, zowel in aanschaf als in gebruik. De optimale setting voor hun gebruik omvat grote universiteiten en tertiaire verwijzingsziekenhuizen.
De methoden die in dit protocol worden gebruikt, zijn overdraagbaar voor de studie van andere vergelijkbare organismen die betrokken zijn bij luchtwegaandoeningen (bijv. Moxarella catarrhalis en Streptococcus pneumoniae). NTHi interageert ook met andere veel voorkomende respiratoire bacteriën10.

Haemophilus influenzae (Hi) is een veel voorkomende bacterie die voorkomt in een reeks ademhalingsaandoeningen. Een verscheidenheid aan verschillende testen / technieken kan worden gebruikt om de respiratoire immuun / ontstekingsreactie op deze bacterie te beoordelen. Flowcytometrie en confocale microscopie zijn op fluorescentie gebaseerde technologieën die gedetailleerde karakterisering van biologische reacties mogelijk maken. Verschillende vormen van Hi-antigeen kunnen worden gebruikt, waaronder celwandcomponenten, gedode / geïnactiveerde preparaten en levende bacteriën. Hi is een kieskeurige bacterie die verrijkte media vereist, maar over het algemeen gemakkelijk te kweken is in standaard laboratoriumomgevingen. Weefselmonsters voor stimulatie met Hi kunnen worden verkregen uit perifeer bloed, bronchoscopie of gereseceerde long (bijvoorbeeld bij patiënten die een operatie ondergaan voor de behandeling van longkanker). Macrofaag en neutrofiele functie kunnen uitgebreid worden beoordeeld met behulp van flowcytometrie met een verscheidenheid aan gemeten parameters, waaronder fagocytose, reactieve zuurstofsoorten en intracellulaire cytokineproductie. De lymfocytenfunctie (bijv. T-cel- en NK-celfunctie) kan specifiek worden beoordeeld met behulp van flowcytometrie, voornamelijk voor intracellulaire cytokineproductie. Hi-infectie is een krachtige inductor van extracellulaire valproductie, zowel door neutrofielen (NET's) als macrofagen (METs). Confocale microscopie is misschien wel de meest optimale manier om NET- en MET-expressie te beoordelen, die ook kan worden gebruikt om protease-activiteit te beoordelen. De longimmuniteit tegen Haemophilus influenzae kan worden beoordeeld met behulp van flowcytometrie en confocale microscopie.

De representatieve resultaten laten zien hoe inflammatoire immuunresponsen op NTHi kunnen worden beoordeeld/gekwantificeerd door flowcytometrie en confocale microscopie. Een belangrijk onderdeel van de interpretatie van de resultaten is de vergelijking in fluorescentie tussen controle- en gestimuleerde monsters. Een aantal voorbereidende experimenten zijn meestal nodig om de kleuring van monsters te optimaliseren. Assessing Respiratory Immune Responses to Haemophilus Influenzae

Haemophilus influenza is een belangrijke oorzaak van ontsteking bij verschillende chronische longziekten, waaronder COPD en longontsteking. Dit protocol beschrijft methoden voor het beoordelen van het effect van haemophilus op longontsteking. De voordelen van deze techniek is dat het een uitgebreide beoordeling van aangeboren en adaptieve immuunresponsen mogelijk maakt.Incubeer 450 microliter perifeer bloed met 50 microliter van een geactiveerd propidiumjodide met het label H influenzae in een buis van vijf milliliter gedurende 20 minuten in een waterbad bij 37 graden Celsius. Verwijder het monster uit het waterbad, voeg vijf microliter DHR en vortex toe gedurende 10 seconden. Plaats het vervolgens nog eens 10 minuten terug in het waterbad.Nadat u het monster uit het waterbad hebt verwijderd, lyseert u de erytrocyten met vijf milliliter ammoniumchlorideoplossing en analyseert u de monsters op een flowcytometer zoals beschreven in de tekst. Verdeel het perifere bloedmonster in aliquots voor controle en antigeenstimulatie. Voeg costimulatoire antilichamen toe aan beide monsters.Voeg vervolgens de niet-typbare H-influenzae toe aan het antigeenmonster en incubeer gedurende een uur bij 37 graden Celsius en 5% koolstofdioxide. Voeg vervolgens het Golgi-blokkerende middel brefeldine A toe aan de monsters en incubeer ze nog eens vijf uur. Na het lyseren van de erytrocyten zoals eerder aangetoond, fixeren de leukocyten met behulp van 500 microliter van één tot 2% para-formaldehyde gedurende één uur.Tel de cellen met behulp van een hemocytometer en permeabiliseer vervolgens 1 miljoen cellen met 100 microliter 0,1% saponine gedurende 15 minuten. Incubeer vervolgens de cellen met geschikte fluorescerende gelabelde antilichamen. Was de cellen en analyseer ze vervolgens met behulp van een flowcytometer.Bepaal het aandeel antigeen reagerende cellen door de relevante lymfocytenpopulaties te krijgen. Voer achtergrondkleuring uit op niet-gestimuleerde cellen voor alle cytokines die moeten worden geanalyseerd. Snijd ongeveer 20 tot 40 gram van het lobectomiemonster in secties van drie tot vijf kubieke millimeter.Plaats ze in een steriele kamer van 50 microliter en fragmenteer het weefsel mechanisch met behulp van een geschikte desaggregator. Na weefseldisaggregatie, lyseer de rode bloedcellen zoals aangetoond en resuspensie de cellen in steriele RPMI. Filter vervolgens de cellen door een steriel nylon gaas van 100 micrometer en tel de levensvatbare cellen met behulp van de trypan blue exclusion-methode.Voor de infectietest resuspensie van de longcellen in RPMI tot een uiteindelijke concentratie van 4 miljoen cellen per milliliter per buis. Infecteer vervolgens de cellen bij een MOI van 100 bacteriën per cel. Maak de dop een halve rotatie los om gasoverdracht in de buizen mogelijk te maken.Plaats de cellen in de buisrotator en incubeer ze bij 37 graden Celsius terwijl ze draaien bij 12 RPM. Voeg een uur na stimulatie grefeldine A toe om de extracellulaire export van cytokines te voorkomen en incubeer de celsuspensies gedurende nog eens 16 tot 22 uur met rotatie. Was de volgende dag de celsuspensie met 500 microliter PBS met 1% runderserumalbumine en 0,01% natriumazide.Kleur vervolgens de celsuspensie voor specifieke menselijke lymfocyten celoppervlakmarkers gedurende één uur. Was vervolgens de cellen met PBS en fixeer en permeabiliseer ze zoals eerder aangetoond. Incubeer vervolgens de cellen met intracellulaire cytokinekleuringsantistoffen gedurende een uur.Was vervolgens de cellen en resuspenteer ze in 100 microliter PBS voordat gegevens worden verzameld op een flowcytometer. Meet proteolyse via de werking van metalloproteinasen, voeg fluoresceïne gelabeld gelatinesubstraat direct toe aan de longweefselsectieglaasjes. Plaats de dia's horizontaal en incubeer ze gedurende een uur in een licht beschermde bevochtigde kamer bij 37 graden Celsius.Om negatieve controles voor te bereiden, voegt u één x-reactiebuffer zonder fluorescerende gelatine toe aan de secties voor verdere analyse. Perifere mononucleaire bloedcellen werden afgesloten met behulp van voorwaartse en zijwaarts verspreide om de fagocytenpopulatie te definiëren. De fagocytenpopulatie werd verder gedefinieerd door CD14-expressie om de monocyten te labelen.ROS-meting werd uitgevoerd via oxidatie van DHR 123 om fluorescentie te produceren. De mediane fluorescentie van het gestimuleerde monster werd vergeleken met de controle. Intracellulaire cytokineproductie door lymfocyten werd gemeten met behulp van flowcytometrie.Cellen werden eerst geanalyseerd op hun expressie van de leukocytenmarker CD45 en vervolgens op CD drie en CD vier, CD acht. De CD drie, CD vier positieve cellen werden beoordeeld op intracellulaire cytokine productie in controle en gestimuleerde monsters. Protease-activiteit werd gemeten door NC twee zymografie in niet-gefixeerde longweefselsecties.Fluorescerende kleuring duidde op de aanwezigheid van MMP-activiteit, die ook werd gecolokaliseerd met de expressie van chromatine. Het toevoegen van antilichamen aan de longweefselmonsters vereist concentratie en kleuring van de cellen vereist optimalisatie in voorbereidende experimenten. Het supernatant van de longweefselmonsters kan verder worden geanalyseerd voor andere ontstekingsmediatoren met behulp van technieken zoals ELISA en Deze technieken kunnen worden gebruikt om het effect van potentiële therapieën op longontsteking te beoordelen.
