インフルエンザ菌は、COPDや肺炎などのさまざまな慢性肺疾患の炎症の主な原因です。このプロトコルは、肺の炎症に対するヘモフィルスの影響を評価する方法を説明しています。この技術の利点は、自然免疫応答と適応免疫応答の包括的な評価を可能にすることです。450マイクロリットルの末梢血と50マイクロリットルの活性化ヨウ化プロピジウム標識Hインフルエンザを5ミリリットルのチューブで摂氏37度のウォーターバスで20分間インキュベートします。サンプルを水浴から取り出し、5マイクロリットルのDHRとボルテックスを10秒間加えます。その後、さらに10分間水浴に戻します。サンプルを水浴から取り出した後、赤血球を5ミリリットルの0.8%塩化アンモニウム溶液で溶解し、本文に記載されているようにフローサイトメーターでサンプルを分析します。末梢血サンプルをコントロールと抗原刺激のためにアリコートに分割します。両方のサンプルに共刺激抗体を追加します。次に、非定型インフルエンザ菌を抗原サンプルに加え、摂氏37度、二酸化炭素5%で1時間インキュベートします。次に、ゴルジ系ブロッキング剤ブレフェルジンAをサンプルに加え、さらに5時間インキュベートします。前述のように赤血球を溶解した後、500マイクロリットルの1〜2%パラホルムアルデヒドを1時間使用して白血球を固定します。血球計算盤を使用して細胞をカウントし、100マイクロリットルの0.1%サポニンで15分間100万個の細胞を透過処理します。次に、細胞を適切な蛍光標識抗体でインキュベートします。細胞を洗浄し、フローサイトメーターを使用して分析します。関連するリンパ球集団を取得することにより、抗原応答細胞の割合を決定します。分析するすべてのサイトカインについて、非刺激細胞のバックグラウンド染色を実行します。約20〜40グラムの葉切除サンプルを3〜5立方ミリメートルのセクションにスライスします。それらを滅菌50マイクロリットルのチャンバー内に置き、適切なディスアグリゲーターを使用して組織を機械的に断片化します。組織脱凝集後、実証されたように赤血球を溶解し、細胞を滅菌RPMIに再懸濁します。次に、100マイクロメートルの滅菌ナイロンメッシュで細胞をろ過し、トリパンブルー排除法を使用して生細胞をカウントします。感染アッセイでは、肺細胞をRPMIで、チューブあたりミリリットルあたり400万個の最終濃度まで再懸濁します。次に、細胞あたり100細菌のMOIで細胞を感染させます。キャップを半回転緩めて、チューブ内のガス移動を可能にします。細胞をチューブローテーターに入れ、12RPMで回転させながら摂氏37度でインキュベートします。刺激の1時間後、サイトカインの細胞外輸送を防ぐためにグレフェルジンAを添加し、細胞懸濁液を回転しながらさらに16〜22時間インキュベートします。翌日、1%ウシ血清アルブミンと0.01%アジ化ナトリウムを含む500マイクロリットルのPBSで細胞懸濁液を洗浄します。次に、特定のヒトリンパ球細胞表面マーカーの細胞懸濁液を1時間染色する。次に、細胞をPBSで洗浄し、前に示したように固定して透過処理します。次に、細胞内サイトカイン染色抗体を用いて細胞を1時間インキュベートする。その後、細胞を洗浄し、フローサイトメーターでデータを取得する前に100マイクロリットルのPBSに再懸濁します。メタロプロテイナーゼの作用によりタンパク質分解を測定し、フルオレセイン標識ゼラチン基質を肺組織切片スライドに直接加えます。スライドを水平に置き、摂氏37度の光で保護された加湿チャンバーで1時間インキュベートします。ネガティブコントロールを調製するには、蛍光ゼラチンを含まない1つの反応バッファーを切片に追加し、さらに分析します。末梢血単核球を、食細胞集団を定義するために前方散乱および側方散乱を用いてゲーティングした。食細胞集団をさらに、単球を標識するためにCD14発現によって定義した。ROS測定は、蛍光を生成するためにDHR 123の酸化を介して実行されました。刺激されたサンプルの蛍光の中央値をコントロールと比較した。リンパ球による細胞内サイトカイン産生は、フローサイトメトリーを用いて測定した。細胞は、最初に白血球マーカーCD45の発現について分析され、次にCD3およびCD4、CD8について分析された。CD3枚、CD陽性細胞4枚を、コントロールおよび刺激試料における細胞内サイトカイン産生について評価した。プロテアーゼ活性は、未固定肺組織切片におけるNCツーザイモグラフィにより測定した。蛍光染色はMMP活性の存在を示し、これもクロマチンの発現と共局在した。肺組織サンプルに抗体を添加するには濃縮が必要であり、細胞の染色には予備実験での最適化が必要です。肺組織サンプルの上清は、ELISAなどの技術を用いて他の炎症メディエーターについてさらに分析することができ、これらの技術は、肺炎症に対する潜在的な治療法の効果を評価するために使用することができる。
