筋萎縮性側索硬化症(ALS)、アルツハイマー病、パーキンソン病、ハンチントン病などの神経変性疾患を含むタンパク質凝集は毒性 が知られており、細胞や臓器のタンパク質恒常性(プロテオスタシス)を乱し、その後老化につながる可能性がある。タンパク質凝集のクリアランスは、治療戦略として期待されています;しかし、タンパク質の凝集を防ぎ、タンパク質凝集体(例えば、小分子や薬物)を分解する化学物質はまだ確立されていません。さらに、タンパク質凝集が毒性を発揮する方法は、依然として不可解です。したがって、タンパク質凝集に関する研究プロジェクトを推進するためには、単にタンパク質凝集を検出する高スループット手順を導入することが重要です。タンパク質凝集および凝集特異的蛍光色素の立体構造を認識する抗体を用いたタンパク質凝集検出は広く用いられている3。しかし、このような古典的な手順を用いた生細胞において特に、凝集を検出することは困難である。 
フェルスター共鳴エネルギー移動(FRET)は、タンパク質凝集および構造変化を検出する手順である。しかし、FRETはタンパク質のダイナミクス(例えば、生細胞におけるタンパク質の拡散およびオリゴマー化)を分析することができない3。そこで、ここでは、単一分子感度4で蛍光分子の拡散特性と明るさを測定する蛍光相関分光法(FCS)を用いて、溶液中のタンパク質凝集(例えば、細胞溶解物)および生細胞を検出する簡単なプロトコルを紹介する。FCSは、レーザースキャン共焦点顕微鏡(LSM)を用いた光子計法である。高感度光子検出器と光子到着時間の自己相関関数(ACF)の計算を用いて、検出体積中の蛍光分子の通過時間と明るさが測定される。拡散は分子量の増加と共に遅くなる。従って、FCSを用いて分子間相互作用を推定することができる。さらに強力に、蛍光分子の明るさの増加は、分子のホモオリゴマー化を示す。したがって、FCSは、このようなタンパク質凝集を検出するための強力なツールである。

タンパク質凝集は、筋萎縮性側索硬化症(ALS)、アルツハイマー病(AD)、パーキンソン病(PD)、ハンチントン病(HD)などの神経変性疾患の特徴である。可溶性または拡散性タンパク質オリゴマーまたは凝集体を検出および分析するために、単一分子感度で単一粒子の拡散速度および明るさを検出できる蛍光相関分光法(FCS)が使用されている。しかし、タンパク質凝集検出のための適切な手順とノウハウは広く共有されていません。ここでは、細胞ライセートおよび生細胞における凝集しやすいタンパク質の拡散特性に関するFCS測定の標準的な手順を示す:ALS関連25 kDaカルボキシル末端断片のTAR DNA/RNA結合タンパク質43kDa(TDP25)およびスーパーオキシドジスムターゼ1(SOD1)を示す。代表的な結果は、緑色蛍光タンパク質(GFP)タグ付きTDP25の凝集物の一部が、マウス神経芽細胞腫Neuro2a細胞溶解物の可溶性画分にわずかに含まれていたことを示している。さらに、ALS関連変異を運ぶGFPタグ付きSOD1は、生細胞における拡散が遅い。そこで、FCSを用いた拡散特性を介してタンパク質凝集を検出する手順を紹介します。

細胞ライセートとSOD1-G85R-GFPのFCS測定を生細胞で行いました。どちらの場合も、正の振幅と滑らかなADFを獲得することができました。Neuro2a細胞で発現したGFP-TDP25の一部が、示された条件6の下で可溶性分率で回収されたことを示した。細胞溶解液の可溶性分率では、FCSを用いた光子カウント率記録において極めて明るい蛍光分子が検出された(図2A、上、?...
