IVDで空間細胞組織を分析しようとする場合、関節軟骨を調査する際に存在しないいくつかの技術的な困難を克服する必要があります。

第二に、組織は人間のドナーから来るのが難しい。膝の全置換を行う場合、関節の全面を得て手術中にさらなる分析を行うことができる。ジ関節関節の変形性関節症は関節全体の疾患でもあるが、関節の一部の領域がそのまま残っている軟骨の質には依然として強い焦点差があるが、例えばその領域の負荷が減少したためである。この状況は、通常、ディスクがグローバルに破壊されたときにのみ手術が行われるIVDでは異なります。手術室からヒトドナーから組織を得るとき、組織も高度に断片化されており、さらなる分析を行う前に、IVDの3つの軟骨タイプのいずれかに組織を正しく割り当てる必要があります。また、より大きな組織切片のより詳細な分析を可能にし、IVDの胚発生を調べるためには、動物モデル生物の選択が必要である。
このようなモデル生物を選択する場合、その解剖学的および寸法、その機械的負荷、現在の細胞集団ならびに組織組成に関して、ヒトディスクと同等のシステムを有することが重要である。ここで提示された技術の目的のために、我々は牛腰椎椎椎組織の使用を示唆する:その低い再生電位をもたらすヒトディスクの重要な特性は、核内の成熟中の脊索細胞の喪失である。しかし、多数のモデル生物において、脊索細胞は生涯を通じて検出することができる。羊、ヤギ、軟骨栄養症犬などの脊索細胞を失う少数の動物のほとんどは、人間のディスクよりもはるかに小さいIVDを持っています。ヒトIVDの直径に匹敵する矢状円盤径を有する腰部ウシディスク15.
初期のディスク変性につながる重要な要因は、過度の機械的負荷です。腰椎の立っている牛の円盤内圧は、背骨が水平に整列した約0.8 MPaです。驚くべきことに、これらの圧力は、勃起したヒト脊椎(0.5MPa)15,16について報告された腰部の椎間板内圧に匹敵する。また、ウシディスク中の水およびプロテオグリカンの量は、若いヒト17からのIVDのそれと同等である。したがって、運動セグメントの実際の移動パターンは、二足歩行動物と二足歩行動物では異なるかもしれないが、総負荷およびディスク特性に関しては、牛は羊や犬などのIVDの他の確立された動物モデルよりもはるかに人間の生物学に近い。
このプロトコルでは、初期胚発生から末期変性まで空間軟骨細胞組織の観点からIVDの変化を解析する方法を提示する。

椎間板(IVD)変性は腰痛の主な原因であり、罹患した個人に対して高い障害を伴う。ディスクの変性を解読し、再生アプローチを開発するためには、IVDの細胞生物学を完全に理解することが不可欠です。この生物学の1つの側面は、細胞が生理学的状態と変性の間にどのように空間的に配置されているかという問題です。IVDの生物学的性質とその可用性により、この組織を分析することは困難です。本研究では、初期胚発生から末期変性まで、アヌル線維症における空間的軟骨細胞組織を調査する。脊椎手術を受けた患者から得られた動物モデルやヒト円盤組織として、牛胚組織を用いた高分解能染色解析を行うため、光学断面化法(Apotome)を適用します。初期胚性牛円盤中の非常に高い軟骨細胞密度から、細胞の数は妊娠、成長、および成熟の間に減少する。ヒトのディスクでは、細胞密度の増加が組織変性の進行を伴った。関節軟骨で既に実証されているように、クラスター形成は高度なディスク変性の特徴を表す。

椎間板変性は、高度な障害と痛みにつながります。我々のプロトコルは、形態学的にディスクの発達と変性の変化を調査することを可能にする。緻密なコラーゲンタイプ1ネットワークは、通常、顕微鏡による椎間板の分析を困難にします。光学的断面化により、異なる組織内の軟骨細胞の3D配置を調べることができます。術中に得られた椎間板またはIVDサンプルをDMEMに2%体積ペニシリンストレプトマイシンで補い、容量で1.2%容量アンホテリシンBを配置し、すぐにコレクションをポストし、IVDサンプルを摂氏4度で保存し、さらなる処理まで摂氏4度で保存します。凍結した場合、ヒトIVDサンプルを室温で解凍し、大学の密度および配向などの顕微鏡特性に基づいてヒトIVD組織の起源を同定する。ウシ円盤組織を採取するには、その2つの隣接する椎骨を有するIVDからなる運動セグメントを取り、外科用ブレード番号15を使用して、軟骨下骨から牛盤全体を解剖し、ディスクを反転させて中心領域に到達する。軟骨の異なる領域を特定した後、外科用ブレード番号20または22を使用して、ディスク全体から対象領域を切り取ります。20センチメートル未満のクラウンランプ長を持つウシ胚からのIVDは、解剖なしで処理する必要があります。原稿に記載された組織の脱灰を行った。20ゲージ針がディスク組織に侵入した場合、または顕著な抵抗なしに椎骨に該当する場合は、正常な脱灰を確認してください。PBSの10倍の体積の組織サンプルを、摂氏4度で一晩PBSで正確に行う。翌日、水溶性埋め込み培地に組織をクライオトームノブに置き、通常はコラーゲンタイプを垂直に切断する軸平面または平面のいずれかが生成される。標準のCryotomeを使用して、組み込み組織を人間のサンプルで70マイクロメートル、牛サンプルで40マイクロメートルの厚さで切り離します。PBSで3回セクションをすすくう前にガラススライドのセクションを収集し、適切な蛍光色素を加え、その後に取り付け媒体を追加し、カバースリップでセクションを覆います。構造照明装置を開始することにより、蛍光顕微鏡のサンプルホルダーに染色組織部を有するスライドを置き、蛍光フィルターおよび適切な照明による単一視野イメージングを行う。蛍光顕微鏡と互換性のある画像最適化ソフトウェアを使用して、強度と明るさを最適化することによって画像を処理します。セクション全体を視覚化するには、ツールバーパネルから60件の取得設定を開き、モザイクレジスタでモザイク設定を調整することにより、モザイクイメージング技術を使用します。これを行うには、後で 1 つの概要画像にマージするビューイメージのフィールドの列と行の数を定義するには、設定を押して、個々のタイルのフォーカス補正を調整します。後処理強度と明るさを最適化することによって画像を処理します。空間コンドロシテ組織を解析するには、スタート/ストップボタンでZスタック設定を調整してソフトウェアに組み込まれた3D機能を使用して、Z軸の開始位置と停止位置やスライス距離などのスキャンパラメータを定義します。個々の細胞パターンを識別し、細胞パターンの定量的分析のために細胞数プラグインを使用して、選択した対象領域のサイズでカウントされたセルを割って細胞密度を計算する。アヌラス内の緻密なコラーゲン繊維ネットワークと柔らかい核を持つIVDのアーキテクチャは、軸および矢状面で撮影されたモザイク画像で認識することができます。モザイク画像の拡大領域では、単一細胞、対、文字列などの軟骨細胞の異なる空間的組織パターンが注目された。牛環状線維症の異なる発達および成熟段階の研究は、胚性円盤の発達中に細胞密度の継続的な減少を示した。一方、変性の間に成人ヒトディスクでは細胞密度とクラスター形成が増加し、観察された。牛の環状線維症およびウシ核パルポスのIVD発症初期の細胞密度は、出生まで急速に減少した。エピトームを用いたIVDのチャネルイメージングにより、細胞質や核などの空間パターンの3Dアーキテクチャを可視化することができ、無傷のIVD単発性ならびに軟骨細胞のペアが発見された。一方、退化したアヌラス細胞クラスターが見つかった。最も困難な部分は、その起源に組織を割り当て、切断のために埋め込まれたを修正することです。これは、信頼できる結果を得るために不可欠です。異なる段階および起源から正しく解剖し、選択されたIVD組織を有することで、ELISAや原子間力顕微鏡などのさらなる生化学的および生力分析によって分析を深めることができる。
