このプロトコルは、昆虫のさまざまな動き、昆虫タンパク質の機能、およびウイルスとベクター昆虫のin vivoでの相互作用を研究するために使用できます。この方法は効率的で有益です。昆虫の腸と補助細胞の構造は、レーザー走査型共焦点顕微鏡で見るとはっきりと視覚化されます。手順をデモンストレーションするのは、私たちの研究室のLu Zhang博士です。まず、ガラスビーカーから非ウイルス性昆虫を、2日間のウイルス獲得アクセス期間のために防虫ネットで覆われた新鮮な南部米黒縞矮性ウイルス(SRBSDV)に移植します。次に、新鮮な米の苗を含むガラスビーカーに昆虫を集めます。2日後、腸の解剖と切除のために手動吸引器を使用してガラスビーカーから昆虫を集めます。テキスト原稿に記載されているように、0.01モルのPBS、4%パラホルムアルデヒド、および2%Triton X-100を準備します。ピペッターを使用して100マイクロリットルのPBSをスライドガラス上に置き、スライドを光学顕微鏡のステージに置きます。手動吸引器を使用して、ガラスビーカーからSRBSDVに感染した成人を収集し、氷上の1.5ミリリットルのチューブに入れて昆虫を麻痺させます。麻痺した成人を腹部を上にしてスライド上の100マイクロリットルのPBSに移します。ピンセットを使用して本体を固定し、別のピンセットで頭を取り外します。1組のピンセットで、腹部の側面を固定し、産卵管または尾の交尾器官をもう一方のセットで固定し、次に腹部セグメントのセグメント間膜を慎重に引っ張って、腹部の腸をゆっくりと露出させます。膜を引き裂き続け、腹部から完全な腸を徐々に引き出します。腸の端に接続されている尾をそっと引き抜いて、腸全体を取り除きます。切除した腸を200マイクロリットルの遠沈管に入れます。チューブに200マイクロリットルのPBSを加え、ピペットを使用して腸を完全に洗浄します。ピペッターを使用して100マイクロリットルのPBSをスライドガラス上に置き、スライドを光学顕微鏡のステージに置きます。手動吸引器を使用して、ガラスビーカーからSRBSDVに感染したニンフを収集し、昆虫を麻痺させるために氷の上に置かれた1.5ミリリットルのチューブに入れます。麻痺したニンフを、腹部を上に向けてスライド上の100マイクロリットルのバッファーに移します。ニンフの尾を外し、昆虫の体をクランプして穏やかに固定し、もう一方のペアを使用して頭をクランプします。頭が体から離れるように、腸への付着を維持したまま頭を体からそっと引き離しますが、腸はまだ胸部と腹部に付着しています。ピンセットがまだ体を固定している状態で、もう一方のペアを使用して頭を慎重に動かし、徐々に腸を引き抜きます。腸を頭から切り離して、体に損傷を与えることなく無傷の腸を取得します。切除した腸をチューブに入れます。チューブにPBSを追加し、ピペットで溶液を静かに吸い込み、腸を完全に洗います。抗体および封入剤を本文原稿に記載されているように調製する。新たに切除しPBS洗浄したWBPH腸を、室温で200マイクロリットルの遠沈管内の100マイクロリットルの4%パラホルムアルデヒドに入れます。2時間後、パラホルムアルデヒド溶液を200マイクロリットルのPBSと交換します。10分後、PBSを除去してパラホルムアルデヒドを除去し、PBS洗浄ステップを2回繰り返します。PBSを2回洗浄した後、200マイクロリットルの非イオン性界面活性剤Triton X-100を加えて、室温でサンプルを透過処理します。30分後、Triton X-100を取り外し、PBSで10分間3回洗浄して、残っている洗剤を洗い流します。標識抗体を50マイクロリットルの雄牛血清アルブミンで1〜50に希釈する。希釈した抗体をチューブに加え、サンプルを摂氏4度で一晩インキュベートします。抗体を除去した翌朝、残った抗体希釈液をPBSで10分間3回洗浄して洗い流します。1マイクロリットルのDyLight 633-ファロイジンを50マイクロリットルのPBSで希釈し、50マイクロリットルの希釈ファロイジンをチューブに加え、室温で2時間インキュベートします。ファロイジンを除去した後、PBSで10分間3回洗浄して残りのファロイジンを完全に洗い流します。DAPIを含む封入剤を一滴顕微鏡スライドに置きます。内臓を培地に移し、気泡を発生させずにカバーガラスをサンプルの上にそっと置きます。ファロイジンで標識した後の成人から摘出したWBPH腸の代表的なレーザー走査型共焦点顕微鏡写真は、前腸、中腸、および後腸の3つのセクションを示しました。これらのうち、中腸はSRBSDVの最初の感染部位です。腸の単層上皮細胞構造は、昆虫タンパク質の細胞局在およびウイルスおよび昆虫タンパク質の共局在の研究を容易にする。DyLight 488標識抗VAMP-7で切除したWBPH腸と、DyLight 549標識抗SRBSDV抗体で切除したSRBSDVビリオンをここに示します。VAMP-7とSRBSDVのビリオンは細胞質内で共局在することが示され、VAMP-7がin vivoでのウイルス感染に関与している可能性があることが示唆されました。透過処理された腸は、ルシフェリン結合抗体とのインキュベーション後に徹底的に洗浄して、バックグラウンドと非特異的結合を減らす必要があります。これは、昆虫のウイルス、他の病原体、および昆虫タンパク質の局在を表示するための信頼できる方法です。それは病原体と昆虫の関係のさらなる研究のための基礎を提供します。
