这里提出的方案描述了一种基于标准组合物15的简单、时间和具有成本效益的制备多功能PURE表达系统20的方法。通过将方案与提供的每日时间表（表1）结合使用，所有组分都可以在1周内制备，并且产量足以进行多达500个10μL PURE反应。由于该方案中使用的蛋白质从高拷贝质粒中过表达并且对大肠杆菌具有低毒性，因此所有必需蛋白质都观察到良好的表达水平（图1）。这允许轻松调整菌株，因此也可以通过修改接种菌株的比例20来调节共培养中的蛋白质组成。除核糖体蛋白外，EF-Tu的浓度对表达产生6具有根本重要性。相反，其他蛋白质组分浓度的变化对PURE系统7，24的鲁棒性的影响相对较小。因此，通过调整EF-Tu相对于所有其他组分的接种比例，可以实现与标准PURE组合物相当的成分，并且可以获得具有相似产量20的PURE系统。在制备蛋白质溶液时，至关重要的是要确保所有菌株生长良好并在诱导后过度表达编码的蛋白质（图1）。
核糖体功能是PURE系统24整体性能的关键。在该协议中，演示了两种用于制备核糖体溶液的不同方法，即无标记和His标记核糖体纯化。无标记核糖体纯化基于疏水相互作用色谱，然后用蔗糖垫离心，这需要使用FPLC纯化系统和超速离心机15。相比之下，利用His标记的核糖体18 和重力流亲和色谱纯化的方法不需要专门的设备，并且可以在大多数实验室中进行。因此，后一种方法具有诸如简单性和可访问性等优点。然而，与无标记变体相比，我们观察到在OnePot PURE中使用His标记的核糖体时合成产量显着降低（图3）。根据应用类型，这种较低的产量可能是可以接受的。
能量解决方案提供为 体外 TX-TL反应提供燃料所需的低分子量组分和tRNA。该协议为典型的能源解决方案提供了配方，可以根据用户需求轻松调整。与tRNA，NTP和磷酸肌酸一起，Mg2 + 离子的丰度和浓度对于PURE系统8的整体性能至关重要，因为它们是转录和翻译的关键辅助因子。因此，在某些情况下，离子滴定可以大大提高整体PURE性能。DNA完整性对于PURE性能至关重要。因此，序列验证启动子区域，核糖体结合位点和靶基因并确保足够的DNA浓度（<2 nM）将有助于解决在建立PURE反应时可能出现的问题。
PURE系统是最小的TX-TL系统，因此特定应用可能需要额外的调整25。这些可能包括掺入不同的RNA聚合酶9、26、伴侣13和蛋白质因子，如EF-P或ArfA8。虽然这些蛋白质的表达菌株可以包含在共培养中，但将它们单独添加到制备的系统中可以更好地控制所需的蛋白质水平。此外，囊泡的内含物对于膜蛋白10，11的产生至关重要。氧化而不是还原环境和二硫键异构酶有利于适当的二硫键形成，例如，分泌蛋白12所需的二硫键。
必须确保任何其他组分不会干扰反应。下面列出了设置反应或添加其他组分时要注意的最重要因素。确保既不使用不相容的缓冲液，也不干扰离子浓度。尽可能避免使用含有甘油、高浓度钾、镁、钙离子、渗透液、焦磷酸盐、抗生素或EDTA的溶液。例如，在DNA纯化过程中用水代替洗脱缓冲液可能是有益的，因为EDTA是该缓冲液中的常见添加剂。为溶液提供额外的带负电荷的分子，如NTP或dNTP，需要调节镁浓度8，因为带负电荷的分子表现为螯合剂并结合带正电荷的分子。中性pH值是反应的理想选择。因此，所有组分都应缓冲到相应的pH值;这对于高酸性或碱性分子（如NTPs）尤其重要。最后，温度和体积是反应的关键参数。为了获得良好的产量，应实现37°C左右的温度，因为低于34°C的温度将显着降低产量27。
值得注意的是，在准备OnePot PURE之前，应考虑目标应用和相关要求，例如体积，纯度，易修改性以及组分的包含或遗漏。对于许多应用，该系统将是一个很好的选择，但其他应用可能需要产量、可调节性和其他因素，而OnePot系统无法提供这些因素。无论如何，引入的协议将有利于任何自制系统的制备，因为此处总结了此类制备的所有关键步骤。

无细胞转录翻译（TX-TL）系统构成了研究和工程生物系统的有前途的平台。它们提供了简化和可调的反应条件，因为它们不再依赖于维持生命的过程，包括生长，体内平衡或调节机制1。因此，预计无细胞系统将有助于生物分子系统的研究，提供测试合理生物设计策略2的框架，并为未来的合成细胞3，4提供底盘。完全重组的PURE系统由于其定义和最小的组成以及可调节性和可调性而提供了特别吸引人的底盘5。
自2001年第一个功能性、全重组的PURE系统建立以来5，已努力扩大系统的限度并优化系统的组成，以提高系统的产量6、7、8，允许转录调节9、膜10、11和分泌蛋白合成12，并促进蛋白质折叠13、14.如今，有三种商用系统：PUREfrex（GeneFrontier），PURExpress（NEB）和Magic PURE（Creative Biolabs）。然而，这些系统成本高昂，其确切组成是专有的，因此未知，适应性有限。

onepot pure cell-free system

上述方案旨在促进在任何实验室中建立纯裸细胞无细胞TX-TL系统。该协议包括对PURE系统的三个不同部分的制备的详细说明：OnePot蛋白，核糖体和能量溶液。优化工作流程的详细每日计划如 表 1所示。工作流程针对他标记核糖体的纯化进行了优化，如果进行无标记核糖体纯化，时间范围可能会略有不同。一种制剂为至少500个10μL反应提供足够量的PURE。此外，制备的溶液在-80°C下稳定超过一年，并且可以承受多次冻融循环。
所有菌株的足够过表达水平对于最终蛋白质溶液的功能至关重要。 图1 显示了随后用于OnePot蛋白制备的所有36种菌株的成功过表达。过度表达的蛋白质条带强度的变化很可能是由于SDS-PAGE凝胶上样体积的偏倚。预期的蛋白质大小总结在 表2中。GlyRS和PheRS由两个不同分子量的亚基组成;其余34种蛋白质由单个亚基组成。该协议简单性和时间有效性的关键是共培养和共纯化步骤（图2）。通过增加EF-Tu菌株相对于所有其他表达菌株的比例来制备OnePot蛋白溶液。通过SDS-PAGE分析最终蛋白质的整体组成（图3A）。从凝胶（泳道2，3）中可以明显看出，与其他蛋白质相比，EF-Tu（43.3 kDa）的浓度更高，正如预期的那样。虽然凝胶提供了蛋白质表达比率的良好初步指示，但很难确定每种蛋白质是否以及在哪个水平上表达。因此，强烈建议在共培养之前确认每种菌株中的过表达，如上所示。
大肠杆菌核糖体是一种复杂的分子机器，由50多个单独的蛋白质亚基23组成。用于无标记核糖体纯化的260nm处的代表性吸收光谱如图4所示;第三个峰是核糖体洗脱成功的特征。对于两种核糖体纯化方法，在SDS-PAGE凝胶上观察到预期的运行模式（图3A）18。我们确实观察到两种净化的污染，尽管数量很少（<10%）。值得注意的是，由于方法的变化，无标记（泳道5，6）和His标记（泳道11，12）核糖体中存在不同的污染物。作为用户参考，还包括用于组合系统的SDS-PAGE凝胶（泳道8、9和14、15）。
最后，比较了使用不同核糖体变体的制备系统的性能（图3）。体外eGFP表达的时间过程表明，两种PURE系统都是功能性的，并产生荧光eGFP。然而，OnePot蛋白溶液与His标记的核糖体结合，使用通过滴定优化的核糖体浓度，仅产生非标记核糖体版本的表达水平的三分之一（图3B）。当使用Green Lys tRNA体外标记系统表达和标记三种不同大小的蛋白质时，观察到类似的结果（图3C）。如荧光凝胶所示，全长产物在两种体系中都成功表达;然而，只有大约一半的表达水平是通过His-tag核糖体系统实现的。除了荧光标记外，所有三种蛋白质的预期条带在考马斯染色的凝胶上也是可区分的（图3D）。结果表明，引入的表达系统可以在一周内在实验室中使用标准设备制备，可用于线性模板中编码的T7启动子下游的蛋白质的体外表达。
