onepot pure cell-free system

대장균은 50개 이상의 개별 단백질 하위유닛(23)으로구성된 복합 분자 기체이다. 태그없는 리보솜 정화를 위한 260 nm의 대표적인 흡수 스펙트럼이 도 4에도시되어 있다. 세 번째 피크는 성공적인 리보솜 용출의 특징입니다. 두 리보솜 정제 방법에 대해, SDS-PAGE젤(도 3A)18에서 예상되는 실행 패턴을 관찰하였다. 소량(<10%)이었기는 하지만 두 정제모두에 대한 오염을 관찰했습니다. 특히, 타구가 없는(차선 5, 6) 및 그의 태그가 지정된(차선 11, 12) 리보솜에 상이한 오염물질이 존재했다. 사용자 참조를 위해 결합된 시스템에 대한 SDS-PAGE 젤도 포함됩니다(차선 8, 9 및 14, 15).
마지막으로, 상이한 리보솜 변종을 이용한 준비된시스템(도 3)의성능이 비교된다. 시험관 내 eGFP 발현의 시간 과정은 PURE 시스템이 모두 기능적이고 형광 eGFP를 생성한다는 것을 보여줍니다. 그러나, 원포트 단백질 용액은 적정에 의해 최적화된 리보솜 농도를 이용하여, 태그가 없는 리보솜버전(그림 3B)의발현 레벨의 3분의 1만 산출하였다. 유사한 결과는 다른 크기의 3개의 단백질이 체외 라벨링 시스템(그림3C)에서녹색 Lys tRNA를 사용하여 발현및 표지되었을 때 관찰되었다. 형광 젤에서 볼 수 있듯이, 전체 길이 제품은 성공적으로 두 시스템에서 발현되었다; 그러나, 표현 수준의 절반 정도만이 그의 태그 리보솜 시스템으로 달성되었다. 형광 라벨링 외에도, 3가지 단백질 모두에 대한 예상 밴드는 쿠마시 스테인드젤(도 3D)에서구별할 수 있다. 결과는 표준 장비가 있는 실험실에서 1주일 이내에 제조될 수 있는 도입된 발현 시스템이 선형 템플릿으로부터 T7 프로모터의 하류로 인코딩된 단백질의 체외 발현에 사용될 수 있음을 보여준다.
