偽ウイルスは、H5 HAPIウイルスに対する中和抗体を研究するための理想的なツールです。中和抗体効力を測定するために、疑似ウイルス中和(PN)アッセイ6が使用される。ヘマグルチニン(HA)とノイラミニダーゼ(NA)は、A型インフルエンザウイルスの表面にある糖タンパク質です7,8。HAは、受容体結合のための球状頭部ドメインと膜融合のためのステムドメインで構成されています。NAタンパク質は、ウイルス放出を促進するシアリダーゼ活性を有する7,8。PNアッセイは、HAタンパク質に対する中和抗体を測定することができます。HAの頭部および幹領域を標的とした中和抗体は、ウイルス付着および侵入アッセイによっても検出することができる。野生型ウイルスと比較して、疑似ウイルス中和実験はより感度の高い検出値を持ち、レベル2のバイオセーフティ研究所で安全に取り扱うことができ、一般的に実際に操作するのが簡単です。
このプロトコルは、H5 HPAI偽ウイルス調製およびPNアッセイの手順および重要なステップを詳細に提示する。また、これらのアッセイのトラブルシューティング、制限、および変更についても説明します。本研究では、H5N1 HPAIウイルス由来のA/Thailand/1(KAN)-1/2004(TH)株を例に挙げた。アッセイに用いた免疫血清を得るために、このプロトコールは、TH株由来のHAタンパク質をマウスを免疫するための免疫原として選択した。

このプロトコルは、H5高病原体鳥インフルエンザ疑似ウイルス調製物および疑似ウイルス中和アッセイの手順および重要なステップを記載する。また、これらのアッセイのトラブルシューティングの制限と変更についても説明します。高病原性病原体はレベル3のバイオセーフティラボで実施する必要がありますが、疑似ウイルスはレベル2のバイオセーフティラボで安全に処理できます。まず、HEK293 FT細胞の単一細胞懸濁液を完全DMEM培地で作製します。10ミリリットルのシングルセル懸濁液をT75フラスコに加え、続いて摂氏37度でインキュベートします。トランスフェクションの2時間前に、古い培地をクロロキンを含む10ミリリットルの新鮮な完全培地と交換します。原稿に記載されているように試薬とプラスミドDNAを混合し、この混合物を細胞の上の培地に移し、穏やかに揺らします。インキュベーション後、培地を15ミリリットルの新鮮な完全DMEM培地と交換します。65時間後、遠心分離により上清を回収する。上清を収集し、それを分注します。ヘマグルチニンタンパク質の発現を検出するには、50マイクロリットルのPBSを96ウェル丸底プレートのカラム2から12に添加します。100マイクロリットルの疑似ウイルスをカラム1のウェルに加えます。次に、50マイクロリットルの疑似ウイルスをカラム1からカラム2のウェルに移します。この2倍希釈をカラム11まで行い、その後穏やかに混合します。次に、カラム11から余分な50マイクロリットルを廃棄します。50マイクロリットルの0.5%赤血球を各ウェルに加え、続いて穏やかに混合します。室温で30〜60分間インキュベートした後、疑似ウイルスの赤血球凝集素力価を観察して記録する。疑似ウイルス滴定の場合は、完全DMEM培地中でMDCK細胞の1ミリリットル当たり4番目の細胞に10〜5倍の単一細胞懸濁液を作製する。1、250、2、500、5、000、10、000、20、000、および40, 000細胞を平底96ウェルプレートの異なるウェルに追加します。インキュベーション後、疑似ウイルスを解凍してボルテックスします。6 つの HAU の疑似ウイルスを 96 ウェル丸底プレートの各ウェルに添加し、各ウェルに最大 120 マイクロリットルの完全な DMEM を補充します。100マイクロリットルの混合物を96ウェルプレート内の細胞に移す。ウイルス感染のために培養プレートを48時間、60時間、および72時間インキュベートし、ルシフェラーゼアッセイを実行します。ルシフェラーゼアッセイを実行するには、培地を注意深く除去し、200マイクロリットルのPBSですすいで細胞を洗浄します。できるだけ多くのPBSを除去し、次に50ミリリットルの溶解バッファーを各ウェルに追加します。翌日、プレートを室温で2時間平衡化した後、培養プレートを数回揺り動かし、すべての細胞ライセートを不透明な96ウェルプレートに移します。各プレートに50マイクロリットルの基板を追加し、よく混ぜます。ルミノメーターを用いて疑似ウイルスのルシフェラーゼ力価を記録する。MDCK細胞の単一細胞懸濁液100マイクロリットルを平底96ウェルプレートの各ウェルに加え、摂氏37度で20時間インキュベートします。インキュベーション後、擬似ウイルスとマイナス80°Cで保存された血清を解凍してボルテックスします。血清を完全培地で20倍に希釈する。100マイクロリットルのDMEM培地をカラム3から10に追加します。200マイクロリットルの希釈血清を第2カラムのウェルに加え、段階希釈を行う。次に、100マイクロリットルのDMEM培地をネガティブコントロールとして11番目のカラムに追加します。各ウェルに100マイクロリットルの疑似ウイルスを加えて混合します。100マイクロリットルの混合物を96穴丸底プレートから96穴細胞培養プレートの対応するウェルに移す。摂氏37度で60時間インキュベートした後、ルシフェラーゼアッセイを行う。ヘマグルチニンアッセイは、インフルエンザ偽ウイルス1ミリリットルあたり643 HA単位がインフルエンザ偽ウイルスストックに存在することを示した。HAとGag P24の比率は正常範囲内であった。ウェスタンブロットアッセイでは、HA0、HA1、HA2、NAタンパク質を含む4種類のタンパク質の存在が示され、インフルエンザ疑似ウイルスと野生型ウイルスの類似性が示されました。疑似ウイルス滴定の結果は、最高の相対ルシフェラーゼ活性のための適切な条件は、96ウェルプレートのウェル中で5, 000細胞の給餌およびウイルス感染の60時間後の相対ルシフェラーゼ活性測定値の検出を含むことを示している。DDV群の免疫血清は、偽ウイルスA Thailand 1 Can104株に対して高い中和力価を示したが、対照群の血清は中和活性を示さなかった。免疫血清の中和活性は対照血清と比較して強力であり、免疫血清中のHAヘッドに多くの抗体が向けられたことを示している。ウイルス侵入の評価中の免疫血清と対照との間の有意差は、抗体が免疫血清のHAステム領域に向けられていることを示している。この手順を実行する際は、トランスフェクション溶液と培地のpHを安定に保つことを忘れないでください。
