このプロトコルは、大型哺乳類におけるリンパ系の調査を可能にし、ヒトのリンパ系の理解に近づくので重要です。この手法には 2 つの利点があります。システルナマグナへの痕跡の導入は、脳内注射とは対照的に、脳への直接的な損傷を回避する。第二に、これは、cannulationを即座に視覚化し、それが成功したかどうかを知ることができる直接的なアプローチです。豚を起こしやすい位置に置くことから始めます。頭と首の後ろを触診して、後頭部の紋章、最初の胸椎の背骨、各耳の基部を見つけてマークします。長手方向の軸に沿って、紋章と椎骨の間に直線を描きます。次に、頭蓋骨の基部に続いて、各耳の基部に紋章から2本の線を引きます。動物の尻尾を注意深く締め付け、反射を見て深い眠り状態にあるかどうかを確認します。21本の刃が付いたメスを使用して縦線に沿って筋肉に下る真皮切開を行い、肩に沿ってさらに2つの垂直切開を延長し、それぞれ10〜15センチメートルの長さになります。後頭紋から始めて、各耳の基部までラインに沿って真皮切開を行います。解剖学的鉗子を使用して後頭部の甲骨で形成された皮膚の角をつかみ、その後、下の筋肉から皮膚を分離するために筋膜の上に軽くメスの刃を実行します。5つの切開の各々に沿って皮膚を切除して、トラペジウス筋の一部を視覚化する。メスを使用して、トラペジウスが正中線で一緒になる深さ約1センチメートルの縦切開を行い、まっすぐな外科的鉗子と湾曲した外科鉗子の組み合わせを使用して筋肉の縦切れに沿って鈍い解剖を行い、トラペジウスと基礎となる半脊髄管炎ビビンテンの腹を分離する。メスで持続する筋線維を切断し、半脊髄炎複合体が見えるまで鈍い解剖を行い続ける。頭蓋骨の後部の側面に沿って移動し、トラペジウスと半脊髄炎のビエンター筋肉の起源を切断します。2つの筋肉を縦方向に分離するには、外科的鉗子を使用して、半脊髄炎の複素体が完全に見えるまで鈍い解剖を行う。その後、自己保持リトラクターを使用して、トラペジウスおよび半脊髄炎胆管筋を引き込みます。メスを使用して、半脊髄炎の腹が正中線で一緒に来る1センチメートルの深い縦方向の切開を行います。アトラスと軸の両方が触知可能になるまで、筋肉の腹の間の縦方向の切り取りに沿って働く外科的鉗子を使用して鈍い解剖を行う。頭蓋骨の後部の側面に沿って移動し、半脊髄炎の複雑な筋肉の起源を切断する。メスと鈍い解剖を使用して、筋肉を下の椎骨から縦方向に分離し、別の自己保持リトラクターを使用して半脊髄炎の複雑性筋肉を引き込みます。メスを使用して、アトラスが頭蓋骨の基部と出会う領域の上にある残りの組織を慎重に取り除きます。動物の首に片方の腕を置き、アトラスと頭蓋骨の分岐点に片方の指を置き、同時に頭を高め、指で触手しながら首を曲げて水槽のマグナを明らかにします。一人の人が動物の頭と首を高めて曲げ、もう一人が水槽マグナを触診し、解剖学的位置をメモします。22ゲージのカニューレを長手方向の軸に斜めに斜めに硬膜を通してシステルナマグナにゆっくりと慎重に導入し、カニューレから針を引っ込み、ロックにキャップを置きます。スーパー接着剤とカニューレが組織に入るアクセルを塗布し、歯科用セメントを塗布します。セメントが固まるまで5分待ちます。カニューレから慎重にキャップを取り外します。10センチメートルの延長を使用してトレーサーとIVラインタップの男性の端にカニューレを取り付けます。トレーサーを1分あたり100マイクロリットルの速度でゆっくりと、手で、またはマイクロ注入ポンプを使用して注入します。IV ラインタップを外し、キャップに交換します。トレーサーがカニューレの基部で脈動して見えるかどうかを確認します。その後、いくつかの屈曲を維持するために、動物の首の下に土嚢を配置します。頭部を離し、動物を安静にする傾向がある状態にしておく。自己保持リトラクターを解放し、筋肉を交換します。外科用タオルクランプを使用して、筋肉の上に皮膚を一緒に持って来ます。まずガーゼを使用し、次に毛布を使用してタオルクランプと切開を覆い、熱損失を制限します。トレーサーが所望の時間だけ循環するようにします。脳の後部表面のステッチされた巨視画像は、スルチと裂け目全体のトレーサーの分布パターンに関する詳細な洞察を提供することができます。脳の腹側および側側面からの同様の画像は、側頭葉および側裂けにおけるトレーサー分布に関する情報を提供することができる。ステレオスコープを用いて生成された高倍率の脳表面の画像は、動脈に沿ってPVS内のトレーサーを視覚化するのに役立ちます。マクロスコピーのコロナ脳部は、海馬や線条体などの半球裂目および皮質下トレーサー分布および構造におけるトレーサー浸透の深さに関する洞察を提供する。AQP4、グリア線維性酸性タンパク質、平滑筋アクチンに対する免疫ヒストリカル染色は、トレーサーがPVS内に局在し、また脳の華脈に移動することを示した。PVSの外表面を形成するアストロサイトの足のプロセスは、AQP4およびグリア性フィブリリン酸性タンパク質染色を使用して同定される。PVSの内面を形成する内皮細胞は、レクチンとGLUT-1染色を用いて染色される。SMA染色は動脈および動脈を識別し、正常なリンパ管機能の基礎生理学を構成する静脈の代わりに動脈に沿ってPVS流入が起こることを示すために使用することができる。今後は、脳卒中や外傷性脳損傷などの神経病理学の文脈で、大型哺乳類の高分解能でリンパ系を探求したいと考えています。
