Hier presenteren we een stapsgewijze methode die HCC's uit hESC's in drie fasen induceert. In de eerste fase werden Activin A en CHIR99021 gebruikt om hESC's te differentiëren in DE. In de tweede fase werden KO-DMEM en DMSO gebruikt om DE te differentiëren in hepatische voorlopercellen. In de derde fase werden HZM plus HGF, OSM en hydrocortison 21-hemisuccinaatnatriumzout gebruikt om leverfappende voorlopercellen te blijven differentiëren in HCC's.

De lever is een zeer metabolisch orgaan dat verschillende rollen speelt, waaronder deoxidatie, opslag van glycogeen en afscheiding en synthese van eiwitten1. Verschillende pathogenen, geneesmiddelen en ertstrouw kunnen pathologische veranderingen in de lever veroorzaken en de functies ervan beïnvloeden2,3. Hepatocyten, als de belangrijkste functionele eenheid van de lever, spelen een belangrijke rol in kunstmatige leverondersteunende systemen en eliminatie van geneesmiddelentoxiciteit. De bron van primaire menselijke hepatocyten is echter beperkt in celgebaseerde therapie, evenals in onderzoek naar leverziekten. Daarom is het ontwikkelen van nieuwe bronnen van functionele menselijke hepatocyten een belangrijke onderzoeksrichting op het gebied van regeneratieve geneeskunde. Sinds 1998, toen hESC's4werden vastgesteld, zijn hESC's door onderzoekers op grote schaal overwogen vanwege hun superieure differentiatiepotentieel (ze kunnen differentiëren in verschillende weefsels in een geschikte omgeving) en hoge mate van zelfvernieuwbaarheid, en bieden zo ideale broncellen voor bioartificiële levers, hepatocyttransplantatie en zelfs leverweefseltechnologie5.
Momenteel kan de efficiëntie van de leverdifferentiatie sterk worden verhoogd door het endoderm te verrijken6. In de afstammingsdifferentiatie van stamcellen in endoderm zijn niveaus van de transformerende groeifactor β (TGF-β) signalerings- en WNT-signaleringsroutes de belangrijkste factoren in de knoop van de endodermvormingsfase. Activering van een hoog niveau van TGF-β en WNT-signalering kan de ontwikkeling van endoderm7,8bevorderen . Activine A is een cytokine behorend tot de TGF-β superfamilie. Daarom wordt Activine A veel gebruikt bij endoderm inductie van door de mens geïnduceerde pluripotente stamcellen (hiPSC's) en hESC's9,10. GSK3 is een serine-threonine eiwitkinase. Onderzoekers hebben ontdekt dat CHIR99021, een specifieke remmer van GSK3β, typische WNT-signalen kan stimuleren en stamceldifferentiatie onder bepaalde omstandigheden kan bevorderen, wat suggereert dat CHIR99021 potentieel heeft voor het induceren van stamceldifferentiatie in endoderm 11,12,13.
Hier rapporteren we een efficiënte en reproduceerbare methode om de differentiatie van hESC's in functionele HCC's effectief te induceren. De sequentiële toevoeging van Activin A en CHIR99021 produceerde ongeveer 89,7±0,8% SOX17 (DE marker)-positieve cellen. Na verder gerijpt te zijn in vitro, drukten deze cellen leverspecifieke markers uit en oefenden hepatocyt-achtige morfologie uit (gebaseerd op hematoxyline-eosinekleuring (H & E)) en functies, zoals opname van indocyaninegroen (ICG), glycogeenopslag en CYP3-activiteit. De resultaten tonen aan dat hESC's met succes kunnen worden gedifferentieerd in volwassen functionele HLC's door deze methode en een basis kunnen vormen voor leverziektegerelateerd onderzoek en in vitro geneesmiddelenscreening.

an efficient method for directed hepatocyte-like cell induction from human embryonic stem cells

De potentiële functies van hepatocyt-achtige cellen (HLC's) afkomstig van menselijke embryonale stamcellen (hESC's) zijn veelbelovend voor ziektemodellering en geneesmiddelenscreeningtoepassingen. Hier wordt een efficiënte en reproduceerbare methode voor differentiatie van hESC's in functionele HCC's gegeven. De oprichting van een endoderm-afstamming is een belangrijke stap in de differentiatie naar HCC's. Met onze methode reguleren we de belangrijkste signaleringsroutes door continu Activin A en CHIR99021 aan te vullen tijdens hESC-differentiatie tot definitief endoderm (DE), gevolgd door het genereren van hepatische voorlopercellen, en ten slotte HCC's met typische hepatocytmorfologie in een stapsgewijze methode met volledig gedefinieerde reagentia. De hESC-afgeleide HCC's die door deze methode worden geproduceerd, drukken stadiumspecifieke markers uit (waaronder albumine, HNF4α nucleaire receptor en natrium taurocholaat cotransporterend polypeptide (NTCP)), en vertonen speciale kenmerken met betrekking tot volwassen en functionele hepatocyten (waaronder indocyaninegroene kleuring, glycogeenopslag, hematoxyline-eosinekleuring en CYP3-activiteit), en kunnen een platform bieden voor de ontwikkeling van HLC-gebaseerde toepassingen in de studie van leverziekten.

Menselijke embryonale stamcel-afgeleide hepatocyt-achtige cellen zijn nuttig bij ziektemodellering en geneesmiddelenscreening. Dit is een efficiënte en reproduceerbare methode om de differentiatie van menselijke embryonale stamcellen in functionele hepatocyt-achtige cellen effectief te induceren. Een ongedifferentieerde status en een geschikte samenvloeiing van menselijke embryonale stamcellen zijn van cruciaal belang voor een succesvolle differentiatie van hepatocytenachtige cellen.Voor stamcelonderhoud, bedek steriele zes-put weefsel cultuur behandelde platen met een milliliter van 1X menselijke embryonale stamcel-gekwalificeerde Matrigel per put, en bewaar ze op vier graden Celsius 's nachts. Laat de platen voor gebruik 30 minuten op kamertemperatuur staan. Ontdooi de cryo-geconserveerde HESE's gedurende drie minuten in een waterbad van 37 graden Celsius zonder te schudden en breng de stamcellen onmiddellijk over door ze in een centrifugebuis van 15 milliliter te gieten met vier milliliter voorverwarmd, 37 graden Celsius mTESR-medium.Centrifugeer de HESE's en aspireer het supernatant en strek de cellen vervolgens voorzichtig opnieuw op in een milliliter mTESR-medium. Aspireer het DMEM F-12 medium van de plaat. Zaai de cellen in een plaat met zes putten met een dichtheid van 1 x 10 tot de 5e cellen in twee milliliter mTESR en incubeer in een incubator van 5% kooldioxide.Onderhoud de cellen door het medium dagelijks te vervangen door voorverwarmd mTESR-medium. Passage de cellen op ongeveer 70 tot 80% samenvloeiing, of wanneer de celkolonies contact beginnen te maken. Voor cellen die passeren, aspireer het medium en incubeer de cellen met één milliliter per put enzymoplossing gedurende vijf minuten bij 37 graden Celsius.Breng de cellen over door ze in een centrifugebuis van 15 milliliter te gieten met vier milliliter DMEM F-12 voorverwarmd tot 37 graden Celsius. Bereid 24-put platen voor door ze te coaten met 250 microliters van 1X hESC-gekwalificeerd Matrigel medium. Zaad hESC's met een dichtheid van 1 tot 1,5 keer 10 tot de 5e cel per put in 500 microliters mTESR medium.Voeg beide activine A toe aan een eindconcentratie van 100 nanogram per milliliter en CHIR99021 aan een eindconcentratie van drie micromolaren in een geschikt volume voorverwarmd basismedium voor fase één differentiatie. Na drie dagen differentiatie moeten gedifferentieerde cellen markers van definitieve endodermcellen uitdrukken, zoals FOXA2, SOX17, GATA4, CXCR4 en FOXA1. Op dag drie van differentiatie, aspireer het medium, vervang het door fase twee medium en incubeer gedurende 24 uur.Verander het medium om de andere dag op dag vier tot en met acht. Na acht dagen differentiatie moeten gedifferentieerde cellen geschikte markers van hepatische voorlopercellen uitdrukken, zoals HNF4-alfa, AFP, TBX3, TTR, ALB, NTCP, CEBPA. Op dag acht, aspireer het stadium twee medium uit de cellen, en vervang het door fase drie medium.Laat cellen 10 dagen incuberen bij 37 graden Celsius in een koolstofdioxide-incubator en verander de media om de andere dag met vers toegevoegde differentiatiefactoren. Na 18 dagen differentiatie moeten gedifferentieerde cellen karakteristieke markers van hepatocyten uitdrukken, zoals AAT, ALB, TTR, HNF4 alpha, NTCP, ASGR1, CYP3A4. In fase twee differentieerden de endodermcellen zich in leverfingsfogenen nadat ze gedurende vijf dagen met een differentiatiemedium waren behandeld. In fase drie, na 10 dagen, waren vroege hepatocyten gerijpt en gedifferentieerd in hepatocytenachtige cellen in hepatocytgroeifactor en oncostatine.In de laatste fase van differentiatie vertoonden cellen een typisch hepatocytfenotype. RT-PCR, immunofluorescentiekleuring en westelijke blotting werden gebruikt om markers van endodermcellen, leverprecursorcellen en volwassen hepatocyten te detecteren. De gedifferentieerde cellen vertoonden hoge expressieniveaus van differentiatiegerelateerde genen en eiwitten in elke fase, zoals endodermmarkers SOX17 op dag drie, leverprecursormarker HNF4-alfa op dag acht, AFP op dag 14 en volwassen hepatocytmarker ALB op dag 18.De HLC's vertoonden groene kleuring en uitgebreide cytoplasmatische zuurverschuivingskleuring. De hepatocyten vertoonden typische bi-nucleated morfologie. De enzymatische activiteit van CYP3A4, de essentiële metabolische CYP in de lever in HCC's, werd bevestigd met een enzymtest.Plaatsing van menselijke embryonale stamcellen met een geschikte dichtheid en begindifferentiatie op de volgende datum zijn van cruciaal belang. Raak in fase één de media voorzichtig aan omdat de cellen gevoelig zijn om omhoog te zweven. De combinatie van activine A en andere kleine moleculen kan de differentiatie-efficiëntie verder verbeteren en in deze procedure meer volwassen hepatocytenachtige cellen produceren.Deze techniek kan een platform bieden om hepatocyttransplantatie, leverweefseltechnologie, bioartificiële levers en ziektemodellering te verkennen.
