肝臓は、脱酸化、グリコーゲンの貯蔵、タンパク質の分泌と合成など、いくつかの役割を果たす高度に代謝性の高い器官である1。様々な病原体、薬物および遺伝は肝臓の病理学的変化を引き起こし、その機能に影響を及ぼす2,3.肝細胞は、肝臓の主要な機能単位として、人工肝支援システムおよび薬物毒性除去において重要な役割を果たす。しかし、原発性ヒト肝細胞の資源は、細胞ベースの治療だけでなく、肝臓病の研究において限られている。したがって、ヒト肝細胞の機能性を新たに開発することは、再生医療分野における重要な研究の方向性である。hESCが確立された1998年から、hESCは優れた分化能力(適切な環境で様々な組織に分化することができる)と高度な自己更新性のために研究者によって広く検討され、バイオ人工肝、肝細胞移植、さらには肝組織工学5に理想的なソース細胞を提供する。
現在、肝分化効率は、内胚葉6を充実させることにより大きく高めることができる。幹細胞の内胚葉への系統分化において、変容成長因子β(TGF-β)シグナル伝達およびWNTシグナル伝達経路のレベルは、内胚形成段階のノードにおける重要な因子である。高レベルのTGF-βおよびWNTシグナル伝達の活性化は、endoderm7,8の開発を促進することができる。アクチビンAは、TGF βスーパーファミリーに属するサイトカインである。従って、アクチビンAは、ヒト誘導多能性幹細胞(hiPSC)及びhESC9,10の内胚葉誘導に広く用いられている。GSK3はセリンスレオンニンプロテインキナーゼです。研究者は、GSK3βの特定の阻害剤であるCHIR99021が典型的なWNTシグナルを刺激し、特定の条件下で幹細胞分化を促進できることを発見し、CHIR99021がエンドドーム11、12、13に幹細胞分化を誘導する可能性があることを示唆している。
ここでは、hESCの機能的なHCへの分化を効果的に誘導するための効率的で再現可能な方法を報告します。アクチビンAおよびCHIR99021の順次添加により、約89.7±0.8%SOX17(DEマーカー)陽性細胞が産生された。さらに 体質化された細胞は、肝特異的マーカーを発現し、肝細胞様形態(ヘマトキシリン-エオシン染色(H&E)に基づく)およびインドシアニングリーン(ICG)の取り込み、グリコーゲン貯蔵およびCYP3活性の取り込みなどの機能を発揮した。この結果は、hESCがこの方法によって成熟した機能的なHlCに正常に分化され、肝疾患関連の研究と インビトロ 薬物スクリーニングの基礎を提供できることを示している。

ヒト胚性幹細胞由来肝細胞様細胞は、疾患のモデル化および薬物スクリーニングに有用である。これは、ヒト胚性幹細胞の機能的肝細胞様細胞への分化を効果的に誘導するための効率的で再現性の高い方法です。ヒト胚性幹細胞の未分化状態および適切な合流は、肝細胞様細胞への分化を成功させるために重要である。幹細胞の維持のために、1Xヒト胚性幹細胞修飾マトリゲル1個のミリリットルを持つ無菌6ウェル組織培養処理プレートをウェルあたりコーティングし、一晩摂氏4度で保存する。プレートは室温で30分間放置してから使用してください。クライオ保存HESEsを37度の水浴で3分間揺らさずに解凍し、すぐに4ミリリットルの事前温められた摂氏37度のmTESR培地を含む15ミリリットルの遠心管にピペットして幹細胞を移管する。HESEsを遠心分離し、上清を吸引し、mTESR培地の1ミリリットルで細胞を穏やかに再懸濁する。プレートからDMEM F-12培地を吸引する。mTESRの2ミリリットルで5番目の細胞に1 x 10の密度で6ウェルプレートに細胞を播種し、5%の二酸化炭素インキュベーターでインキュベートします。培地を毎日予熱mTESR培地に置き換えて細胞を維持する。約70〜80%合流で細胞を通過させる、または細胞コロニーが接触し始めるとき。通過する細胞の場合は、培地を吸引し、37°Cで5分間酵素溶液1ウェルあたり1ミリリットルで細胞をインキュベートします。セルを摂氏37度に予め温めたDMEM F-12の4ミリリットルを含む15ミリリットルの遠心分離管にピペット化して移管する。24ウェルプレートを250マイクロリットルの1x hESC修飾マトリゲル培地でコーティングして準備します。500マイクロリットルのmTESR培地で1〜10倍の密度で5番目の細胞を500マイクロリットルで播種する。両方のアクチビンAを1ミリリットル当たり100ナノグラムの最終濃度に加え、CHIR99021を適切な体積の予熱ステージ1分化基本培地で3マイクロモルの最終濃度に加える。分化の3日後、分化細胞は、FOXA2、SOX17、GATA4、CXCR4、およびFOXA1のような決定的な内胚細胞のマーカーを発現すべきである。3日目の分化時に、培地を吸引し、ステージ2培地に交換し、24時間インキュベートする。4日目から8日目に、1日おきにメディアを変更します。8日間の分化後、分化した細胞は、HNF4アルファ、AFP、TBX3、TTR、ALB、NTCP、CEBPAなどの肝前駆細胞の適切なマーカーを発現するべきである。8日目に、ステージ2培地を細胞から吸引し、ステージ3培地に置き換える。細胞が二酸化炭素インキュベーターで摂氏37度で10日間インキュベートし、1日おきに新たに添加された分化因子で培地を交換することを可能にする。分化の18日後、分化細胞は、例えばAAT、ALB、TTR、HNF4アルファ、NTCP、ASGR1、CYP3A4などの肝細胞の特徴的なマーカーを発現すべきである。hESCからの肝細胞様細胞の概略図と各分化段階の明視野画像を示す。ステージ1では、殺菌剤AおよびCHIR99021を3日間添加し、幹細胞を誘導して内胚葉細胞を形成した。ステージ2では、内胚葉細胞は5日間分化培地で処理された後、肝前駆細胞に分化した。ステージ3では、10日後、初期肝細胞は成熟し、肝細胞増殖因子およびオンコスタチンにおいて肝細胞様細胞に分化した。分化の最終段階では、細胞は典型的な肝細胞表現型を示した。RT-PCR、免疫蛍光染色、およびウェスタンブロッティングを使用して、内胚葉細胞、肝前駆細胞、および成熟肝細胞のマーカーを検出した。分化した細胞は、3日目の内胚葉マーカーSOX17、8日目の肝前駆体マーカーHNF4アルファ、14日目のAFP、18日目の成熟肝細胞マーカーALBなど、各段階で分化関連遺伝子およびタンパク質の高い発現レベルを示した。HLCは緑色染色と広範な細胞質酸シフト染色を示した。肝細胞は典型的な二核形態を示した。CYP3A4の酵素活性は、HLCsにおける肝臓における必須の代謝CYPであり、酵素アッセイで確認された。ヒト胚性幹細胞を適切な密度で座り、次の日に分化を開始することが重要である。ステージ1では、細胞が浮かび上がりやすいので、メディアに優しく触れます。アクチビンAと他の小分子の組み合わせは、分化効率をさらに向上させ、この手順でより成熟した肝細胞様細胞を産生する可能性がある。この技術は、肝細胞移植、肝臓組織工学、生物人工肝臓、および疾患モデリングを探求するためのプラットフォームを提供することができる。
