デカフルオロブタンは、気体状態と液体状態5、6、7の間で相転移を行うのに理想的なパーフルオロカーボン化合物である。デカフルオロブタンは、マイクロバブルを温度低下だけで液滴に凝縮することを可能にするが、一方、より密度の低いパーフルオロカーボンは追加の加圧5を必要とする。この穏やかな方法は結露7、8、9の間に気泡の破壊を最小にする。そのコアは液体であるとして、液滴は非エコー原性であり、超音波には見えない。しかし、十分な音響エネルギーまたは熱エネルギーの適用により、液体コアは気体状態に戻って気化し、エコー原性マイクロバブル8を発生させることができる。この気化により、マイクロバブルを生成するタイミングと場所を制御できます。

synthesis and characterization of multi-modal phase-change porphyrin droplets

位相変化液滴は、十分な音響エネルギーを適用してその 時に エコー原性マイクロバブルに変換することができる超音波造影剤のクラスである。液滴は、マイクロバブルの対応よりも小さく、より安定しています。しかし、従来の超音波造影剤は音響フィードバック測定を超えて追跡可能ではなく、造影剤の生体分布または蓄積 ex vivo の定量化が困難になる。研究者は、生体分布を推測するために蛍光または光学吸収性コンパニオン診断粒子に依存する必要があります。このプロトコルの目的は、凝縮法を用いてマルチモーダル位相変化ポルフィリン液滴を作成するためのステップを詳述することにある。ポルフィリンは、脂質に結合し、液滴に組み込んで液滴に組み込むことができる明確な吸光度バンドを持つ蛍光分子であり、音響特性を保持しながらより堅牢な生体分布を可能にします。ポルフィリン-脂質および塩基脂質含量の異なる7つの製剤を、マイクロバブルおよび液滴サイズ分布を調べるために作られた。ポルフィリン含有構造に適した特性評価は、その解析的汎用性をインソリューションで実証するプロトコルにも記載されている。サイジングは、凝縮後の平均直径が前駆体集団よりも1.72〜2.38倍小さいことを示した。吸光度特性は、無傷のアセンブリが700nmのQバンドピークを有し、破壊されたサンプルは671nmで吸光度ピークを有したことを示した。蛍光特性評価は、30%のポルフィリン-脂質集合体が蛍光的に消光(>97%)であり、中断時に蛍光回復が達成されたことを示した。音響気化により、ポルフィリン液滴は低圧で非エコー原性であり、十分な圧力でエコー原性マイクロバブルに変換できることが示された。これらの特性評価は、 インビボ またはex vivoでの送達または治療用途のための超音波造影剤の生体分布を定量化するための吸光度または蛍光ベースのコンパニオン診断戦略の必要性を排除するポルフィリン滴滴の可能性を示す。

あらかじめ凝縮されたサイズ選択されたマイクロバブルサンプル(n=3)とポスト凝縮されたドロップレットサンプル(n= 3)を、10 μmの開口部を有するコールターカウンター(CC)でサイズ設定しました。10 μmの絞りの制限は、200 nmより小さい粒子を測定できないことであり、平均サイズと濃度を偏らさせる可能性があります。図4は、パイロ-脂質含有量製剤の各々のサイジングデータを示す。表 1 は、サイズ変更データに基づく統計を示しています。前および縮合後平均直径の比率を用いて、結果は0%パイロ-脂質製剤が0.02±1.72で最小の平均直径シフトを有することを示した。50%のパイロ-脂質製剤は、0.08±2.38で最大の平均直径を持っていました。1%のパイロ-脂質液滴サンプルは、10 10 /mL×1010 /mLで最も高い観測濃度(2.71±0.13)を持ち、40%のパイロ-リドロップレットサンプルは10 9/mL×最も低い(7.36±0.81)で最も低い観察濃度を持っていました。サイジングデータは、10%のパイロ-リピッド液滴サンプルが261±13nmで最小のピークダイメーターを有し、50%のパイロリピッド液滴サンプルが390±55nmで最大であったことを示した。一般的に、パイロ-脂質含量が増加するにつれて、濃度が低下し、平均直径が増加した。凝縮後のサンプルは前駆体マイクロバブルサンプルに基づいているため、両方のタイプの超音波造影剤に対して傾向が生じた。パイロ-脂質含量が増加するにつれて、微小気泡亜集団(ピークサイズは約2000μm)が形成され始めた。この二次ピークは、0%のパイロ-脂質マイクロバブルサンプルには存在せず、40%および50%のパイロ脂質集団において最も明らかであった。
図5は、30%パイロ-脂質液滴サンプルの代表的な吸光度測定を示す。PBSの無傷のサンプルのピークは700nmであったが、トリトンでの破壊されたサンプルはピークを671nmにシフトした。これは、無傷の組立体が、個々の未組立脂質成分と比較して異なる光学特性を有することを示した。
図6A は、予縮されたマイクロバブルサンプルの代表的な蛍光測定を示し、 図6B は30%パイロ脂質を有するポスト凝縮液滴サンプルを示す。PBSの無傷のサンプルは704 nmで蛍光ピークを有し、破壊された形態は674nmでピークを有した。カーブの下の破壊領域を曲線の下の無傷の領域で差を引き、その差を曲線の下の破壊された領域で除算すると、クエンチ効率が得られ、30%のパイロ脂質マイクロバブルサンプルと液滴サンプルでそれぞれ98.61%と98.07%になります。
マイクロバブルに変換する液滴を実証するために、希釈された液滴を、超音波システムを用いた37°Cフローファントムで画像化および気化した。 図7 は、異なる圧力で画像化された30%パイロ-リドロップレットサンプルの代表的な超音波画像を示す。低圧(図7A)では、寒天合成から立ち往生した気泡からのバックグラウンド信号のみ、信号がほとんどなかった。これは、液滴はエコー原性ではなく、超音波を散乱しないためです。わずかに高い電力で、明るい斑点の出現によって示されるように、いくつかのマイクロバブル(図7B)が発生した。圧力が高くなるにつれて、より多くのマイクロバブルが発生した(図7C および 7D)。また、液滴が37°Cで自然気化しないことも実証した。

このプロトコルは、脂質膜からマイクロバブルへの脂質溶液への液滴を作成し、次に液滴に液滴を作成するための完全な手順を提供するので、重要です。この技術の主な利点は、マイクロバブルを液滴に凝縮するためのシンプルさです。冷却と旋回だけで、加圧を必要とせずに液滴を作ることができます。マイクロバブルが正常に凝縮されているかどうかを確認する簡単なテストは、半透明の変化を視覚的にチェックすることであるため、視覚的なデモンストレーションが重要です。この手順のデモンストレーションは、ギャング・ジェン研究所の修士課程の学生、キムン・ユーです。デキャッパーを使用して、血清バイアルのアルミニウムシールを取り外し、脂質溶液の1ミリリットルをフェノールスクリューキャップ付きの1.85ミリリットルのホウケイ酸ガラスサンプルバイアルに移し、脂質溶液を気泡を作らずに内壁に流し込みます。1.85ミリリットルのサンプルバイアルを使用すると、ガス交換器を使用してサンプルバイアルヘッドスペースにデカフルオロブタンガスで流れる準備をします。ガス交換器のすべてのバルブが適切に閉じられており、ポンプの電源が切れていることを確認します。マニホールドバルブを開き、マニホールドから対応する針を慎重に外します。圧力バルブAと圧力バルブBを開き、ガスボンベバルブを約1/16~1/8反時計回りに回して部分的に開きます。Tハンドルバルブを開き、脂質溶液でサンプルバイアルをキャップしません。マニホールド針をバイアルの液体エアインターフェースの上に置き、空気調整器ゲージの針が静止位置からわずかに動くまで空気調整バルブを時計回りにゆっくりと回し、パーフルオロカーボンガスを30秒間介機用ヘッドスペースに静かに流し込み、気泡を作らないよう注意します。必要に応じて、エアレギュレータバルブを調整します。30秒後、慎重かつ迅速にバイアルをあまり動かさずにサンプルバイアルを保管しました。ガスボンベバルブ、Tハンドルバルブ、エアレギュレータバルブ、圧力バルブA、圧力バルブB、マニホールドバルブの両方。慎重に針をシースし、サンプルバイアルにラベルを付け、時計回りに行くワックスフィルムで首を密封します。サンプルバイアルは暗闇の中、摂氏4度で少なくとも10分または24時間保存します。別の断熱容器の所定の場所に、約100グラムのドライアイスを断熱容器に入れます。1.5インチ20ゲージ針、1ミリリットルのプラスチック注射器、200ミリリットルの容器、金属トング、および温度計に以前に準備されたデカフルオロブタン血清バイアルを取り出します。サンプルバイアルを脂質溶液と一緒に機械的攪拌機に入れ、攪拌します。色や半透明に変化があるはずです。機械的な攪拌の後、光からシールドされたサンプルバイアル右側を立ち上げ、15分間のカウントダウンを開始してバイアルを冷却し、サイズはマイクロバブルを選択します。カウントダウンが10分に達したら、約200ミリリットルのイソプロパノールで容器を満たし、金属トングを使用してドライアイスでマイナス20度まで冷却します。マイクロバブルのサイズを 15 分間選択した後、サンプルバイアル内で選択したパーティションのサイズを探します。サンプルバイアルの右側を上に保ち、サンプルバイアルを慎重にキャップを外し、1ミリリットルのプラスチックシリンジに取り付けられた1.5インチ20ゲージ針で底の仕切りの約0.7ミリリットルを引き出します。トップパーティションのどれも引き抜かないことを確認し、エアポケットを取り外すために注射器をフリックしないでください。ゴム栓の中央円を目指して、別の20ゲージ針をデカフルオロブタン血清バイアルに挿入し、血清バイアルの上部付近に針を置いてベントし、選択したマイクロバブルのサイズで針を挿入します。選択したマイクロバブルのサイズをゆっくりと転送します。デカフルオロブタン血清バイアルの内壁を静かに滑り落とします。すべてのサイズが選択されたマイクロバブル溶液が移された後、注射器で針を取り除くが、負圧を緩和するために通気針を保つ。イソプロパノール浴に少量のドライアイスまたは室温イソプロパノールを加えて、浴温がマイナス15〜マイナス17°Cの間であることを確認します。20ゲージの通気針をセラムバイアルの上部付近に挿入し、血清バイアルとイソプロパノール浴を置き、マイクロバブルレベルをイソプロパノールのレベル以下に保ちますが、バイアルネックを上にして2分間断続的に旋回してマイクロバブルを凝縮します。イソプロパノールで血清バイアルを連続的に旋回させず、溶液を凍結させないでください。約5秒間旋回し、イソプロパノールから血清バイアルを持ち上げます。氷の核形成を確認し、イソプロパノールで旋回を再開します。氷の形成がある場合は、空気中の血清バイアルを放散するまで旋回する。2分間の結露の後、イソプロパノール浴から血清バイアルを取り除き、通気針を取り除きます。マイクロバブルは、半透明の変化によって示されるように液滴に凝縮しているはずです。血清バイアルを拭き、ラベルを付け、使用できるまで暗い断熱容器の中の通常の氷の上に置きます。無傷のアルミニウムシールを持つ未開封の液滴は、溶けた氷が必要に応じて交換される限り、最大6時間安定する必要があります。使用する準備ができたら、デキャッパーでアルミニウムシールを取り外します。このプロトコルを使用した後、あらかじめ凝縮されたサイズは、選択されたマイクロバブルおよびポスト凝縮された液滴を、10ミクロンの開口を有するコールターカウンター上でサイズ設定した。30%のパイロ-脂質製剤のサイジングデータが示されている。サイジングデータに基づく統計を以下に示します。前と後凝縮平均直径の比率を使用して、結果は、火光脂質含量が増加するにつれて、濃度が低下し、平均直径が増加することを示した。代表吸収剤は、30%のパイロ-脂質液滴試料の測定が示されている。これは、無傷のアセンブリが個々の未組み立て脂質成分と比較して異なるピークによって反射される異なる光学特性を有することを示した。前凝縮されたマイクロバブルの代表的な蛍光測定は、30%のパイロ脂質を有する縮合後液滴サンプルをここに示し、破壊された形態の無傷のサンプルに対して異なる蛍光ピークを示す。異なる圧力で画像化された30%の熱量脂質液滴サンプルの代表的な超音波画像が示されている。低圧では、寒天合成から貼り付けた気泡からの背景信号のみが観察された。わずかに高い電力で、いくつかのマイクロバブルが生成され、これは明るい斑点の出現によって示される。圧力が高まるにつれて、より多くのマイクロバブルが発生した。ここでの凝縮温度は、この特定の液滴シェル製剤に最適化されていることを覚えておくことが重要です。異なるシェル製剤は、異なる温度を必要としてもよい。
