</ol>2. 通过突触前胆碱能信号传导评估DD树突棘的激活

选择Airyscan探测器来获取DD棘的快照，因为它提供了比传统共聚焦显微镜更高的信噪比和更好的分辨率19，20。AiryScan成像还允许使用传统的荧光蛋白（例如GFP，mCherry等），现在广泛用于 秀丽隐杆线虫。尽管可以使用其他超分辨率方法（例如，STORM，STED，PALM）获得更高分辨率的图像，但这些方法需要可光激活或可光切换的荧光蛋白21。作为Airyscan的替代方案，建议使用传统的共聚焦显微镜。例如，使用奈奎斯特采集成像（图3）使用123.9nm的40x/1.3物镜实现像素尺寸，足以区分脊柱形态类型（图2）。
为了确定脊柱密度，建议使用细胞溶质荧光蛋白，例如（1）mCherry或GFP，（2）LifeAct标记肌动蛋白细胞骨架，或（3）肉豆蔻酰化荧光蛋白（例如MYR：：mRuby）标记质膜（图1B）。相比之下，F-肌动蛋白结合蛋白促卵磷蛋白降低脊柱密度（图1C），表明当促卵核蛋白过度表达时对脊柱形态发生有负面影响。
目前的成像方法应该有助于识别控制脊柱形态的遗传变异1，16。DD脊柱形态（即细/蘑菇状，丝状，粗短，分枝，见 图2）可以从腹侧神经索侧图像的单个2D投影进行评估，因为大多数DD脊柱采用特征性的腹侧定向方向。在这些比较中，必须对每种情况使用相同的荧光标记物，因为明显的脊柱形态类型似乎受到标记方法的影响（例如，MYR：：mRuby 与生活法：：GFP）。此外，注意到脊柱形状是动态的，并且可能响应外部信号2，16而改变形状。因此，在相似的发育阶段和相似的条件下比较基因型之间的脊柱形状也很重要。
秀丽隐杆线虫腹侧索的方向对于准确的图像采集至关重要。动物相对两侧的腹侧和背带都应该在同一个Z平面上可见，表明蠕虫的方向是在其侧面（图1B）。最好不要收集蠕虫移动或与其他蠕虫或腹侧脊髓附近的气泡接触的图像，因为这会降低棘的图像。
对于 体内 钙成像，需要在每次采集前立即准备新鲜载玻片。最好对仅与细胶纤维接触的蠕虫进行成像， 而不是胶水的“球”，往往会使蠕虫干燥并降低图像质量（图4B）。在 图4所示的实验中，561 nm光的脉冲激活了整个视场。为了提高时间和空间分辨率以检测局部Ca++ 瞬变，例如，在单个DD棘内，可以使用为561 nm激光线设置的振镜迷你扫描仪来刺激较小的感兴趣区域17。

树突棘是特殊的细胞结构，接收来自邻近神经元的输入以进行突触传递。神经递质受体的激活提高了这些特征性神经元突起中的细胞内钙和下游信号通路1。由于树突棘对神经传递及其在神经发育疾病中的失调至关重要1，因此发现调节树突棘形态发生和功能的因子对神经科学领域具有高度兴趣。
最近，根据与哺乳动物棘共享的关键特征，在秀丽隐杆线虫神经系统中发现了树突棘2。这一决定至关重要，因为它为利用秀丽隐杆线虫的优势来研究脊柱生物学开辟了可能性。背侧D（DD）运动神经元上的树突棘接收腹侧神经索胆碱能神经元（VA和VB）的输入（图1A）2，3，4。本文介绍了成像方法，以探索DD树突棘的结构及其在完整神经系统中的体内功能，该系统易于实时成像和遗传分析。为了监测树突棘的形状，（1）细胞溶质荧光蛋白，其填充树突突和棘;（2）膜结合的荧光蛋白，装饰树突棘和树突的边界;或（3）使用富含树突棘的肌动蛋白标记LifeAct5或Utrophin6，从而揭示其形状。为了监测DD棘的功能，GCaMP荧光用于检测突触前胆碱能神经元中红移视蛋白Chrimson激活引起的Ca++瞬变7。这两种策略都有望促进野生型和突变动物中DD树突棘的研究。
