Het protocol begint met de groei van oHSV in low-passage Vero cellen. De samenvloeiing van de Vero-celmonolaag moet ~ 80% zijn op het moment van virusinenting, omdat overwoekerde cellen strakke vezelige structuren kunnen ontwikkelen die de oHSV-invoer in Vero-cellen kunnen verminderen20. Oncolytische virussen (OV's) zijn een opkomende en unieke vorm van kankerimmunotherapie. OP's repliceren selectief tumorcellen in en lyse (sparende normale/gezonde cellen)1 terwijl ze antitumorimmuniteit induceren2. Oncolytisch herpes simplex-virus (oHSV) is een van de meest uitgebreid bestudeerde virussen onder alle OV's. Het is het verst in de kliniek, met Talimogene laherparepvec (T-VEC) als eerste en enige OV die FDA-goedkeuring in de VS heeft ontvangen voor de behandeling van gevorderd melanoom3. Naast T-VEC worden veel andere genetisch gemanipuleerde oHSV's preklinisch en klinisch getest bij verschillende kankertypen3,4,5,6,7,8. De huidige geavanceerde recombinante DNA-biotechnologie heeft de haalbaarheid van de engineering van nieuwe oHSV's voor therapeutische transgene(n)3,5verder vergroot . Een efficiënt systeem van oHSV propagatie, zuivering en titer bepaling is van cruciaal belang voordat een (nieuw ontwikkelde) oHSV kan worden getest voor in vitro en in vivo studies. Dit artikel beschrijft een eenvoudige stapsgewijze methode van oHSV-groei (in Vero-cellen), zuivering (volgens de sucrose-gradiëntmethode) en titratie (door een oHSV-plaquetest in Vero-cellen) (Figuur 1). Het kan gemakkelijk worden aangenomen in elke bioveiligheid niveau 2 (BSL2) laboratoriuminstelling om een hoogwaardige virale voorraad voor preklinische studies te bereiken.
Vero, een Afrikaanse groene apenniercellijn, is de meest gebruikte cellijn voor oHSV-propagatie9,10,11,12,13 omdat Vero-cellen een defecte antivirale interferonsignaleringsroute hebben14. Andere cellijnen met inactivated stimulator of interferon genes (STING) signalering kunnen ook worden gebruikt voor oHSV-groei12,13. Dit protocol maakt gebruik van Vero-cellen voor oHSV-groei en plaquetest. Na vermeerdering worden met oHSV geïnfecteerde cellen geoogst, gelyseerd en onderworpen aan zuivering, waarbij gelyseerde cellen eerst worden behandeld met benzonasenuclease om gastheercel-DNA af te doen, nucleïnezuur-eiwitaggregatie te voorkomen en de viscositeit van het cellysaat te verminderen. Aangezien een goede activering van benzonase vaak Mg2+vereist , wordt in dit protocol mgCl2 1-2 mM MgCl2gebruikt . Het gastheercelresten van het met benzonase behandelde cellysaat worden verder geëlimineerd door seriële filtratie vóór snelle sucrose-gradiëntcentrifugatie. Een stroperig 25% sucrose-oplossingskussen helpt om een langzamere virusmigratie door de sacharoselaag te garanderen, waardoor gastheercelgerelateerde componenten in het supernatant achterblijven, waardoor de zuivering wordt verbeterd en virusverlies in de pellet wordt beperkt16. De gezuiverde oHSV wordt vervolgens getitreerd op Vero-cellen en virale plaques worden gevisualiseerd door Giemsa-kleuring17 of X-gal-kleuring (voor LacZ-codering van oHSV's)18.

growth, purification, and titration of oncolytic herpes simplex virus

Oncolytische virussen (OV's), zoals het oncolytische herpes simplex-virus (oHSV), zijn een snel groeiende behandelingsstrategie op het gebied van kankerimmunotherapie. OP's, waaronder oHSV, repliceren selectief in en doden kankercellen (sparen gezonde/normale cellen) terwijl ze antitumorimmuniteit induceren. Vanwege deze unieke eigenschappen worden oHSV-gebaseerde behandelingsstrategieën steeds vaker gebruikt voor de behandeling van kanker, preklinisch en klinisch, waaronder fda-goedgekeurde talimogene laherparevec (T-Vec). Groei, zuivering en titratie zijn drie essentiële laboratoriumtechnieken voor alle OVs, inclusief oHSV's, voordat ze kunnen worden gebruikt voor experimentele studies. Dit artikel beschrijft een eenvoudige stapsgewijze methode om oHSV in Vero-cellen te versterken. Naarmate oHSV's zich vermenigvuldigen, produceren ze een cytopathisch effect (CPE) in Vero-cellen. Zodra 90-100% van de geïnfecteerde cellen een CPE vertonen, worden ze voorzichtig geoogst, behandeld met benzonase en magnesiumchloride (MgCl2),gefilterd en onderworpen aan zuivering met behulp van de sucrose-gradiëntmethode. Na zuivering wordt het aantal infectieuze oHSV (aangeduid als plaquevormende eenheden of PFUs) bepaald door een "plaquetest" in Vero-cellen. Het hierin beschreven protocol kan worden gebruikt om hoog-titer oHSV-voorraad voor te bereiden op in vitro studies in celkweek en in vivo dierproeven.

Een kort overzicht van het hele protocol is weergegeven in figuur 1, die de kritieke stappen vertegenwoordigt die betrokken zijn bij de groei, zuivering en titratie van oHSV. Oncolytisch herpes simplex-virus is een opkomende vorm van kankerimmunotherapie. Een efficiënt systeem van virusvoortplanting, zuivering en titerische bepaling is van cruciaal belang voor het gebruik in experimentele studies. Deze methode is zeer eenvoudig en kan gemakkelijk worden aangenomen in elke bioveiligheid niveau twee laboratoriuminstelling om een hoge kwaliteit virale voorraad voor preklinische studies te ontvangen.De productie van een hoogwaardige, zuivere virale voorraad is cruciaal omdat het wordt gebruikt om de immuunrespons tegen kanker te bestuderen en een nieuwe vorm van op virussen gebaseerde kankerimmunotherapie te ontwikkelen. Dit protocol kan worden gebruikt om elk wild-type of genetisch gemanipuleerd herpes simplex-virus te zuiveren. Dit protocol moet gemakkelijk te lezen en gemakkelijk te volgen zijn voor een persoon die deze techniek nog nooit eerder heeft uitgevoerd.De vermelde materialen en reagentia moeten aanwezig zijn voordat u deze techniek voor het eerst probeert. Begin met het wassen van de T-150 vierkante centimeter kolven met twee keer hoge glucose DPBS aangevuld met 1%IFCS. Aspireer de DPBS en voeg zeven milliliter virusenoculum per kolf toe.Laat de kolven 5 minuten zachtjes schommelen en incubeer gedurende 1,5 tot 2 uur op 37 graden Celsius. Verwijder vervolgens het entmateriaal en voeg 25 milliliter DMEM aangevuld met 1% IFCS toe aan elke kolf. Verzamel na incubatie bij 37 graden Celsius en 5% kooldioxide gedurende twee tot vier dagen 20 milliliter van het kweeksupernatant uit elke kolf in centrifugebuizen van 50 milliliter.Schraap met behulp van een celschraper de cellen van de bodem van de kolven. Voeg ongeveer 15 milliliter van het eerder verzamelde kweeksupernatant toe aan elke kolf om het volume tot 20 milliliter te brengen en gebruik vervolgens een steriele serologische pipet van 10 milliliter om de bodem van de kolven een paar keer voorzichtig te wassen. Verzamel de cellen met medium in 50 milliliter conische centrifugebuizen die op ijs zijn geplaatst.Centrifugeer de buizen 10 minuten op 300 maal G bij 4 graden Celsius. Resuspendeer elke pellet grondig in 1,25 milliliter virusbuffer, of VB, en 1,25 milliliter eerder verzameld kweeksupernatant. Meng alle geresuspendeerde pellets tot één conische centrifugebuis van 50 milliliter.Vries de geresuspendeerde cellen in met droogijs en 100% ethanol en bewaar ze bij min 80 graden Celsius. Ontdooi de eerder gebroken bevroren cellen in een warmwaterbad bij 37 graden Celsius en soniceer gedurende één minuut gedurende drie cycli in een waterbad. Voeg 175 eenheden Benzonase Nuclease en twee microliters van twee millimol magnesiumchloride per milliliter cellysaat en vortex toe.Na een incubatie van 30 minuten en warmwaterbad bij 37 graden Celsius, plaats de buis op ijs. Draai de cel lysaat op 300 maal G gedurende 10 minuten. Verzamel het supernatant in een nieuwe conische centrifugebuis van 50 milliliter en resuspendeer de celkorrel in 500 microliter VB. Wijs het aan als pellet één.Draai het verzamelde supernatant opnieuw op 500 maal G gedurende 10 minuten en bewaar het supernatant afzonderlijk in een verse 50 milliliter conische centrifugebuis om later te gebruiken. Resuspendeer de celkorrel in 500 microliters VB en benoel het als pellet twee. Combineer de geresuspendeerde pellet één en pellet twee in een nieuwe centrifugebuis van 1,75 milliliter en vortex en soniceer tweemaal in een waterbad voordat u de buizen gedurende 10 minuten op 400 keer G draait.Verzamel het supernatant uit 1,7 milliliter centrifugebuis en combineer het met de eerder verzamelde 50 milliliter conische centrifugebuis. Filtreer het supernatant door een steriel PVDF-membraanfilter van vijf micrometer in een nieuwe centrifugebuis van 50 milliliter, buis één genaamd. Voeg een milliliter VB toe aan de centrifugebuis van 50 milliliter, geleegd na het filteren van het supernatant uit de vorige stap, vortex, en passeer het door hetzelfde PVDF-membraanfilter dat op buis één is geplaatst.Geef het filtraat van buis één door een steriel MCE-membraanfilter van 0,8 micrometer dat op een nieuwe centrifugebuis van 50 milliliter, buis twee, is geplaatst. Voeg een milliliter VB toe aan de geleegde buis één, vortex, en passeer deze door hetzelfde MCE-filter dat op buis twee is geplaatst. Geef het filtraat van buis twee door een steriel PVDF-filter van 0,45 micrometer dat op een nieuwe centrifugebuis van 50 milliliter met het label buis drie is geplaatst.Zoals eerder beschreven, herhaal het wassen van buis twee met één milliliter VB om filtraat toe te voegen aan buis drie. In deze representatieve analyse kon het cytopathische effect worden waargenomen in Vero-cellen op 36, 48 en 72 uur na oHSV-infectie met de afronding van de aangetaste cellen. Het toenemende niveau van CPE in de loop van de tijd is duidelijk zoals gevisualiseerd door mCherry expressie.Na 72 uur na infectie werden virale plaques bevlekt met X-gal om oHSV's met lacZ-expressie te visualiseren. Het voorzichtig afwassen van met virussen geïnfecteerde cellen en het voorzichtig wassen van de bodem op de kolf is van cruciaal belang om alle met HSV geïnfecteerde cellen intact te houden, zodat een hoge titer virale stam kan worden verkregen.
