growth, purification, and titration of oncolytic herpes simplex virus

単純ヘルペスの腫瘍は、がん免疫療法の創発的な形態である。ウイルスの伝播、精製、およびタイテリック定量の効率的なシステムは、実験研究での使用に不可欠です。この方法は非常に簡単で、前臨床試験のための高品質のウイルスストックを受け取るために任意のバイオセーフティレベル2の実験室の設定で容易に採用することができる。それは抗癌免疫応答を研究し、ウイルスベースの癌免疫療法の新しい形態を開発するために使用されるため、高品質、純粋なウイルスストックの生産は非常に重要です。このプロトコルは、野生型または遺伝子操作されたヘルペス単純ヘルペスウイルスを浄化するために使用することができます。このプロトコルは、これまでこのテクニックを実行したことがない個人のために読みやすく、従いやすいものでなければなりません。この技術を初めて試す前に、リストされた材料と試薬を設置する必要があります。T-150平方センチメートルのフラスコを1%IFCSを補った2倍の高グルコースDPBSで洗浄し始めます。DPBSを吸引し、フラスコあたり7ミリリットルのウイルス接種を加える。フラスコを5分間軽く揺らし、摂氏37度で1.5~2時間インキュベートします。次に、接種を取り除き、各フラスコに1%IFCSを加えたDMEMの25ミリリットルを加えます。摂氏37度と5%の二酸化炭素で2〜4日間インキュベートした後、各フラスコから20ミリリットルの培養上清を50ミリリットルの遠心管に集める。細胞スクレーパーを使用して、フラスコの底部から細胞を削ります。各フラスコに、以前に採取した培養上清の約15ミリリットルを加え、体積を20ミリリットルまで引き上げ、10ミリリットルの滅菌血清学的ピペットを使用してフラスコの底部を数回静かに洗います。氷の上に置かれた50ミリリットル円錐形遠心分離管に培地で細胞を収集します。チューブを摂氏4度で10分間300倍Gで遠心分離する。各ペレットを1.25ミリリットルのウイルスバッファー、VB、および1.25ミリリットルの以前に採取した培養上清で完全に再懸濁させる。再懸濁されたペレットをすべて50ミリリットル円錐形遠心分離チューブ1個に混ぜます。ドライアイスと100%エタノールを使用して再懸濁セルを凍結し、マイナス80°Cで保存します。以前にスナップした冷凍細胞を摂氏37度の温水浴で解凍し、水浴で3サイクル1分間超音波処理します。ベンゾナーゼヌクレアーゼ175単位と、細胞のライセートと渦のミリリットル当たり2ミリモル塩化マグネシウム2マイクロリットルを加えます。30分のインキュベーションと37°Cの温水浴の後、氷の上にチューブを置きます。細胞のライセートをGの300倍に10分間回転させます。新しい50ミリリットル円錐型遠心分離管に上澄み物を集め、VBの500マイクロリットルで細胞ペレットを再懸濁します。ペレット1として指定します。収集した上清をG500倍に10分間再び回転させ、後で使用するために新鮮な50ミリリットル円錐形遠心分離管に上澄み剤を別々に保管します。細胞ペレットをVBの500マイクロリットルに再懸濁し、ペレット2と指定する。再懸濁されたペレット1個とペレット2を新しい1.75ミリリットル遠心管と渦に組み合わせ、水浴で2回超音波処理してから、400倍Gでチューブを10分間回転させます。1.7ミリリットル遠心管から上清を収集し、以前に収集した上清と組み合わせます 50 ミリリットル円錐形遠心分離チューブ.無菌5マイクロメートルのPVDF膜フィルターを通して上清をチューブ1と呼ばれる新しい50ミリリットル遠心管にフィルターします。VBを50ミリリットルの遠心管に1ミリリットル加え、前工程から上清を濾過した後に空にし、チューブ1に配置された同じPVDF膜フィルターを通過します。チューブ1からチューブ1から濾過液を、チューブ2と呼ばれる新しい50ミリリットル遠心管に置かれた無菌0.8マイクロメートルのMCE膜フィルターを通して渡します。空にしたチューブ1、渦にVBの1ミリリットルを追加し、チューブ2に配置された同じMCEフィルタを介してそれを渡します。チューブ2から濾液を、チューブ3とラベル付けされた新しい50ミリリットル遠心管に配置された無菌0.45マイクロメートルPVDFフィルターを通して渡します。前に述べたように、チューブ2の洗浄を1ミリリットルのVBで繰り返し、チューブ3に濾液を加える。この代表的な分析では、細胞性効果は、影響を受けた細胞の丸めによるoHSV感染後36、48、および72時間のベロ細胞で観察することができた。時間の経過に伴うCPEの増加レベルは、mCherry発現によって視覚化されるように明らかである。感染後72時間で、ウイルスプラークをX-galで染色し、lacZ発現でoHSVを可視化した。ウイルス感染細胞を穏やかにこすり落とし、フラスコの底を静かに洗うことは、HSV感染細胞をすべてそのまま維持し、高いっきりしたウイルスストックを得るために重要です。
