このプロトコルは、細胞内のリボソームの活性に関する分子の詳細を明らかにするポリソームプロファイルを生成する方法を説明しています。この技術により、ユーザーは、自動グラジエント分画システムにアクセスできないポリソームプロファイルによって提供される貴重な情報を取得できます。勾配を準備するときは時間をかけて、線形勾配に落ち着くときにコンプレッサーやその他の機械的操作によって中断されない場所に勾配を保管してください。まず、スクロースグラジエントバッファーに7%および47%スクロースのストック溶液を調製します。フィルターは、0.22ミクロンのフィルターを通してショ糖ストック溶液を滅菌します。14%ストック溶液と27%ストック溶液を分注して混合することにより、17、27、および47%のスクロース溶液を47ミリリットル準備します。6本のポリプロピレン製遠心チューブをフルビューの試験管ラックに入れます。一方のチューブでのアクションが他のチューブの邪魔にならないように、チューブ間に十分なスペースがあることを確認してください。先端が鈍い9インチ、22ゲージの針を3ミリリットルのシリンジに取り付け、テスト充填と分注を実行して、シリンジが滴り落ちることなくショ糖溶液を保持できることを確認してから、勾配を設定します。各遠沈管の底に2ミリリットルの7%スクロースを加えます。次に、針先をチューブ底部のすぐ近くに配置することにより、7%溶液の下に17%スクロースを2ミリリットル加えます。慎重にゆっくりと溶液を分配します。27、37、および47%スクロース溶液のそれぞれ2ミリリットルで手順を繰り返し、各層が密度の分離を示す線によって互いに区別できることを確認します。勾配を摂氏4度で一晩保存して、スクロースの割合が連続的に増加するようにします。酵母の増殖および回収後、サッカロミセス・セレビシエ細胞を、冷却した700マイクロリットルのポリソーム抽出バッファーに細胞を再懸濁する。RNAse阻害剤を100単位添加します。次に、約425〜600ミクロンのサイズの400マイクロリットルのプレチルドガラスビーズを追加します。ガラスビーズを入れた1.5ミリリットルの遠沈管に混合物を移し、ビーズビーターで5分間激しく攪拌して酵母細胞を破壊します。細胞を破壊した後、遠心分離によってライセートを清澄化する。蛍光ベースのRNA検出システムを使用して、260ナノメートルの吸光度を測定することにより、清澄化されたライセート中のRNAの濃度を決定します。RNA濃度がマイクロリットルあたり0.5〜1マイクログラムであることを確認してください。RNA濃度が低すぎる場合は、細胞を再懸濁するために使用する抽出バッファーの量を減らします。ライセートをグラデーションの上部に慎重にロードします。ピペットチップをポリプロピレンチューブ上部の内壁に置きます。チューブを傾け、ライセートをグラデーションの上部にゆっくりと分注し、壁に滴り落ちます。チューブをスイングバケットローターの事前に冷却されたバケットにそっと置き、グラジエントを遠心分離します。遠心分離後、スイングバケットローターから遠心管を慎重に取り外し、チューブホルダーに入れます。96ウェルプレートにラベルを付けて画分を保存し、氷上でプレチルします。ピペットチップをグラジエントの上部に注意深く挿入し、グラジエント全体が分注されるまでフラクションを収集することにより、グラジエントの上部から100または200マイクロリットルのフラクションを収集します。ブランクとして7%および47%スクロース溶液に対して分光光度計を使用して254ナノメートルで各画分の吸光度を測定します。分数対吸光度をプロットしてポリソームプロファイルを作成します。酵母S.Cerevisiae由来の3つの代表的なポリソームプロファイルが、示されている。各リボソームサブユニットの頂上およびポリソームピークは、各プロファイル上で明らかである。自動密度分画システムの代表的なプロファイルをここに示します。このプロファイルは、スクロース勾配が追跡溶液によって下から上に置換され、検出器フローセルを介してフラクションに収集されたときの連続吸光度プロファイルから生成されました。200マイクロリットルと100マイクロリットルのサンプルを手で分画することによって生成されたポリソームプロファイルをここに示します。この手順で覚えておくべき最も重要なことは、グラデーションを乱さないことです。溶液の分注が速すぎて気泡が発生したり、勾配が乱れたりしないように注意してください。この手順に続いて、RNAをグラジエントから抽出してさらに分析し、活性リボソームと会合しているmRNAを決定することができます。
