このプロトコルは、新しい製剤のスクリーニングを容易にして、最適な薬物送達システムを設計する上で重要な組織滞留時間に対する修飾がどのような影響を与えるかをよりよく理解する。この技術は非侵襲的であり、リアルタイムのイメージングを可能にし、サンプリングのための動物の過度の使用を減らす。薬物送達システムは、単に治療薬とイメージング部分を1つのナノキャリアに含めることによって、画像化および治療用途向けに再設計することができる。これは、がんと感染症の治療に有用な用途を有する。この手順を実証することは、私の研究室の上級研究員であるラシャ・ムサラムです。まず、結膜下注射の2時間前に、エバンスブルー、またはEBの1キログラムあたり2.5ミリグラムで尾を介してマウスを静脈内に注入します。5%イOFLURANeでマウスを鎮静した後、マウスを鼻コーンに移し、加熱パッドで鎮静マウスを維持します。次に、注射する目の近くでヒゲをトリミングします。0.5%のプロキシメタカイン塩酸塩溶液を直接目に植え付けます。次に、蛍光リポソームを備えた32ゲージの針を備えた10マイクロリットルのガラス注射器をロードし、気泡がシリンジに存在しないことを確認します。ピンビーザーを使用して、結膜を持ち上げ、ゆっくりと結膜下腔にリポソームを注入し、針をゆっくりと引き出し、後部の流れを防ぎます。蛍光リポソームで満たされたブレブの形成を確認します。目に1%のフジチ酸の低下を投与し、意識を取り戻すまでマウスを監視します。CLMシステムのスイッチを入れます。プローブをCLMシステムに接続します。この時点での取得ファイルのフィールドと場所を選択します。蛍光DHPEの蛍光検出が直線範囲にあることを確認し、異なる時点で撮影した画像間の比較のために強度を一貫して保つようにレーザー強度を調整します。システムが15分間温まったら、各レーザーのクレンジング、すすめ、フルオロ4のソリューションを含むキャリブレーションキットを使用して、メーカーの指示に従ってシステムを較正します。簡単に言えば、クレンジングバイアルにそれぞれ5秒間チップを浸し、次にリンスバイアルに入れて校正を開始します。プローブのさまざまなファイバーからの背景値を正規化するために、バックグラウンド記録のために先端を空中に残し、画像の均一性を確保します。再度、クレンジングバイアルにチップを5秒間浸し、次にリンスバイアルに浸します。次に、fluoro-4 488ナノメートルを含むバイアルに5秒間チップを浸し、信号値を正規化します。クレンジングバイアルでチップ浸漬を5秒間繰り返し、続いてリンスバイアルを洗い流します。前のステップで記録された蛍光シグナルが消えるまで、リンスバイアルに先端を置いてください。次に、フルオロ4 660ナノメートルを含むバイアル内の先端を転写し、プローブ内の異なる繊維からのシグナル値を正規化する。キャリブレーション後、プローブの背景の値が 100 未満であることを確認します。それ以外の場合は、綿先端アプリケータでプローブの洗浄を繰り返し行います。加熱パッドを取り付けた動物温度コントローラのスイッチを入れ、温度を摂氏37度に調整します。外科用ドレープで加熱パッドを覆った後、加熱パッドのノーズコーンを固定します。誘導室で5%のイOFLURANeを使用してマウスを鎮静した後、マウスを鼻コーンに移し、鎮静したマウスを加熱パッドに維持します。次に、麻酔薬0.5%のプロキシメタカイン塩酸塩溶液を眼に植え付ける。眼表面を洗い、レンズの結晶化を避けるために、目を潤滑して、生理食塩水を少し落とします。次に、顕微鏡の接眼レンズを回転させて調整し、マウスの眼を0.67倍の倍率で直接焦点で目を見るようにします。レーザーをオンにし、画像を作成する目の領域に直接ペンのようにプローブを配置します。次に、獲得の記録を開始し、対象領域で眼の蛍光を観察する。すべての領域にフラグが立てられ、正確なフレームでのプローブ位置を知るために、アイマップに従ってラベルが付いている場合は、記録を停止します。取得ファイルは、以前に選択した場所にビデオファイルとして自動的に保存されます。蛍光イメージングは、スクレラ領域と角膜領域の間に明確な分化を明らかにした。エピスクレラと結膜の高い血管化により、強膜領域はEBで赤く染色された。血管系を欠く角膜は黒く見えた。7日間の研究で、リンブとスクレラのコントラスト蛍光が観察された。蛍光色素注入対照マウスは、リポソーム由来蛍光の特異性を確認した。蛍光強度は1日目と3日目に高かった。緑色蛍光は、時間の経過とともに四肢およびスクレラ領域のリポソーム減少をプロキシした。一時的な四肢および優れた四肢領域における注射後1日目から3日目までの蛍光シグナルの違いは有意ではなく、特に角膜周辺に対する中性リポソームの好みを示す。1日目の経時的および優れた領域では、クリンフラの蛍光は四肢の6倍であった。3日目と7日目までに、リポソームは眼クリアランス機構に起因する側頭および優れた領域の側頭および上位領域のスクレラで有意に減少した。蛍光の最も低い量は、注射部位からの距離のために、鼻および劣った領域で検出された。記録が停止する前に、眼球のフレームを眼球に従ってラベル付けすることが重要です。レーザー強度を一貫して保持し、異なる実験間で適切な比較を行うための最大背景値を定義します。鉛製剤の同定に続いて、動物における包括的な薬物動態および生体分布試験を行い、薬物の生物学的利用可能性を研究し、それらの製剤の治療効果を検証することができる。
