Kritische stappen van het protocol voor de groei van cellen in microzwaartekracht zijn de bioreactor, de steiger, de cellen die worden gebruikt voor 3D-cultuur en de steigercoating als middel om celdifferentiatie te induceren. Zwaartekracht beïnvloedt alle aspecten van het leven op aarde, inclusief de biologie van individuele cellen en hun functie binnen organismen. Cellen voelen zwaartekracht door middel van mechanoreceptoren en reageren op veranderingen in de zwaartekracht door cytoskeletale architecturen opnieuw te configureren en door de celdelingte veranderen 1,2,3. Andere effecten van microzwaartekracht zijn de hydrostatische druk in met vloeistof gevulde blaasjes, sedimentatie van organellen en door drijfvermogen aangedreven convectie van stroming en warmte4. Studies naar het effect van verlies van zwaartekracht op menselijke cellen en organen werden oorspronkelijk uitgevoerd om de gewichtloze omgeving van de ruimte op astronauten tijdens ruimtevluchtmissies te simuleren5. In de afgelopen jaren worden deze 3D-bioreactortechnologieën die oorspronkelijk door NASA zijn ontwikkeld om microzwaartekracht te simuleren, echter steeds relevanter als nieuwe benaderingen voor de cultuur van celpopulaties die anders niet vatbaar zijn voor 2D-cultuursystemen.
3D-bioreactoren simuleren microzwaartekracht door cellen in suspensie te laten groeien en zo een constant "vrije val" -effect te creëren. Andere voordelen van de roterende bioreactoren zijn het gebrek aan blootstelling aan lucht in orgaankweeksystemen, een vermindering van schuifspanning en turbulentie en een continue blootstelling aan een veranderende toevoer van voedingsstoffen. Deze dynamische omstandigheden die worden geboden door een rotary cell culture system (RCCS) bioreactor bevorderen ruimtelijke co-lokalisatie en driedimensionale assemblage van enkele cellen tot aggregaten 6,7.
Eerdere studies hebben de voordelen aangetoond van een roterende bioreactor voor botregeneratie8, tandkiemcultuur9 en voor de kweek van menselijke tandheelkundige follikelcellen10. Er is ook een rapport dat suggereert dat RCCS de proliferatie en differentiatie van EOE-cellen in ameloblastenverbetert 11. Gedifferentieerde cellen werden echter beschouwd als ameloblasten op basis van ameloblastine-immunofluorescentie en / of amelogenine-expressie alleen11 zonder rekening te houden met hun langwerpige morfologie of gepolariseerde celvorm.
Naast de door NASA ontwikkelde rotating wall vessels (RWV) bioreactor, omvatten andere technologieën om 3D-aggregaten uit cellen te genereren magnetische levitatie, de random positioning machine (RPM) en de clinostat12. Om magnetische levitatie te bereiken, worden cellen gelabeld met magnetische nanodeeltjes zweven met behulp van een externe magnetische kracht, wat resulteert in de vorming van steigervrije 3D-structuren die zijn gebruikt voor de biofabricage van adipocytenstructuren 13,14,15. Een andere benadering om microzwaartekracht te simuleren is het genereren van multidirectionele G-krachten door gelijktijdige rotatie over twee assen te regelen, wat resulteert in een annulering van de cumulatieve zwaartekrachtvector in het midden van een apparaat genaamd clinostat16. Wanneer beenmergstamcellen werden gekweekt in een clinostat, werd nieuwe botvorming geremd door de onderdrukking van osteoblastdifferentiatie, wat een van de dedifferentiërende effecten van microzwaartekracht illustreert16.
In vitro systemen om de trouwe kweek van ameloblasten te vergemakkelijken, zouden een belangrijke stap voorwaarts zijn in de richting van tandglazuur tissue engineering17. Helaas is de cultuur van ameloblasten tot op heden een uitdagende onderneming geweest18,19. Tot nu toe zijn vijf verschillende ameloblastachtige cellijnen gemeld, waaronder de muis ameloblast-lijn cellijn (ALC), de rat tandheelkundige epitheelcellijn (HAT-7), de muis LS8 cellijn20, het varkensPABSo-E cellijn21 en de rat SF2-24 cellijn22. De meerderheid van deze cellen heeft echter hun kenmerkende gepolariseerde celvorm verloren in 2D-cultuur.
In de huidige studie hebben we ons gewend tot een Rotary Cell Culture Bioreactor System (RCCS) om de groei van ameloblastachtige cellen uit cervicale lus epithelia samen gekweekt met mesenchymale voorlopers te vergemakkelijken en om de uitdagingen van 2D-kweeksystemen te overwinnen, waaronder verminderde stroom van voedingsstoffen en cytoskeletale veranderingen als gevolg van zwaartekracht. Daarnaast heeft de RCCS nieuwe wegen geboden voor de studie van cel/scaffold interacties gerelateerd aan parodontale weefselmanipulatie en om de effecten van inflammatoire mediatoren op longalveolaire weefsels in vitro te onderzoeken. Samen benadrukken de resultaten van deze studies de voordelen van op microzwaartekracht gebaseerde rotatiecultuursystemen voor de voortplanting van gedifferentieerde epithelia en voor de beoordeling van milieueffecten op cellen die in vitro zijn gekweekt, inclusief cel/ scaffold-interacties en de weefselrespons op ontstekingsaandoeningen.

Zwaartekracht is een van de belangrijkste determinanten van menselijke celfunctie, proliferatie, cytoskeletale architectuur en oriëntatie. Roterende bioreactorsystemen (RCCSs) bootsen het verlies van zwaartekracht na zoals het in de ruimte optreedt en bieden in plaats daarvan een microzwaartekrachtomgeving door continue rotatie van gekweekte cellen of weefsels. Deze RCCS'en zorgen voor een ononderbroken toevoer van voedingsstoffen, groei- en transcriptiefactoren en zuurstof, en pakken enkele van de tekortkomingen van zwaartekrachten in onbeweeglijke 2D (tweedimensionale) cel- of orgaancultuurschotels aan. In de huidige studie hebben we RCS'en gebruikt om cervicale luscellen en tandpulpcellen te cokweken om ameloblasten te worden, om parodontale voorloper / steigerinteracties te karakteriseren en om het effect van ontsteking op longblaasjes te bepalen. De RCCS-omgevingen vergemakkelijkten de groei van ameloblastachtige cellen, bevorderden parodontale voorloperproliferatie als reactie op steigercoatings en maakten een beoordeling mogelijk van de effecten van inflammatoire veranderingen op gekweekte longblaasjes. Dit manuscript vat de omgevingsomstandigheden, materialen en stappen onderweg samen en belicht kritieke aspecten en experimentele details. Kortom, RCCS'en zijn innovatieve hulpmiddelen om de cultuur en 3D (driedimensionale) groei van cellen in vitro onder de knie te krijgen en om de studie van cellulaire systemen of interacties mogelijk te maken die niet vatbaar zijn voor klassieke 2D-cultuuromgevingen.

De binnenkamer van de bioreactor biedt een omgeving voor de cellen om zich te vermenigvuldigen en te differentiëren, zich aan een steiger te hechten of samen te komen om weefselachtige assemblages te vormen. Elk HARV-vat bevat tot 10 ml medium en vergemakkelijkt een constante circulatie van voedingsstoffen, zodat elke cel een uitstekende kans heeft om te overleven. Langwerpig en gepolariseerd de amelogenine-afscheidende ameloblast in de cultuur. Dit is ook het eerste protocol voor het kweken van ameloblastcellen in microzwaartekracht. De groei van ameloblasten in de cultuur is een van de grootste uitdagingen op het gebied van glazuur geweest.En voor ons is het gebruik van dit bioreactormodel, het is de eerste keer dat we dit kenmerk in glazuuronderzoek hebben bereikt. Dit model kan worden gebruikt om de biologie van ameloblastcellen te begrijpen. Dit model zal worden gedemonstreerd door Dr.Mirali Pandya.Begin met het plaatsen van het verzamelde weefsel van zesdaagse postnatale muizen onder de dissectiemicroscoop en ontleedde cervicale lussen. Stel een bioreactor in door de steriele kleppen aan de spuitpoorten te bevestigen. Voeg beide cellen, de cervicale lus en de tandpulp, samen met gecoate steiger toe aan de bioreactor en vul het bioreactorvat met 10 milliliter keratinocyten SFM-media aangevuld met groeifactoren en extracellulaire matrixeiwitten.Sluit en draai de vulpoortdop van het bioreactorvat dicht en draai deze vast en open vervolgens de steriele kleppen. Gebruik twee steriele spuiten van drie milliliter voor een mediawissel. Vul hiervoor één spuit met het verse medium en laat de andere spuit leeg.Open beide spuitkleppen en manoeuvreer de bubbels voorzichtig naar de lege spuitpoort. Zodra het verse medium van de volle spuit voorzichtig in het vat is geïnjecteerd, verwijdert u alle belletjes door de lege spuitpoort. Sluit de spuitpoorten met doppen.Bevestig elk vat aan de rotatorbasis. Schakel de stroom in en stel de snelheid in op 10,1 rotaties per minuut. Pas de snelheid aan om ervoor te zorgen dat de steigers in ophanging zijn en de vaatwand niet raken.Voor mediaverandering om de 24 uur, plaats de bloedvaten rechtop om ervoor te zorgen dat de cellen zich op de bodem nestelen. Open de spuitpoorten om de steriele spuiten op de poorten aan te sluiten. Aspirateer 75% van het voedingsarme medium en injecteer het verse medium door de andere poort zoals eerder aangetoond.De macrofoto's van grafeensteiger en collageensteiger zijn hier weergegeven. De grafeensteiger had een parallelle reeks oppervlakte-reliëfs, terwijl de collageensteiger de poreuze structuur vertoonde. Een geskeletiseerde muiskaak werd ontleed en het distale grootste deel van de onderste muissnijder werd blootgelegd.De precieze positie van de gereseceerde cervicale lus wordt afgebakend in de zesdaagse postnatale muissnijder, terwijl een vergelijkbaar gebied in een volwassen geskeletiseerde muiskaak als referentie wordt gegeven. De hemimandibles bestonden uit drie mandibulaire kiezen en een voortdurend groeiende snijtand, terwijl de cervicale luscelniche een gevarieerde celpopulatie bevatte die nodig was voor glazuurvorming. De succesvolle differentiatie van de cervicale luscellen in een op maat gemaakte micro-omgeving resulteerde in de vorming van ameloblasten met een typisch kenmerk van gepolariseerde cellen met de kern aan de ene kant en lange cellulaire processen aan de andere kant.De cultuur van cellen in media alleen zonder groeifactoren en steigercoating resulteerde in cervicale luscellen die belangrijke glazuureiwitten afscheidden, maar niet langer of polariseerden. De galanine-gecoate steigergroep vertoonde een significant hogere proliferatiesnelheid in vergelijking met de controlegroep met ongecoate steigers. De longweefselsegmenten geoogst van E15-muizen werden met succes gekweekt gedurende 10 dagen in een 3D-microzwaartekrachtomgeving in de bioreactor.De toevoeging van interleukine-6 aan het kweekmedium resulteerde in ontstekingsgerelateerde veranderingen in de alveolaire morfologie vergelijkbaar met die in vivo. In de inleiding vertelde ik je hoe moeilijk het was voor het emailleteam om met een ameloblastcelkweekmodel te komen. Nu hebben we deze uitdaging aangepakt door een zeer innovatieve procedure te gebruiken die zich richt op twee aspecten: sponscollageensteiger en ook het gebruik van matrix om de ameloblastcellen te differentiëren.Samen resulteerden deze twee innovaties in de polarisatie en groei van ameloblastachtige cellen in 3D-cultuur. Deze 3D-gekweekte cellen zijn een prachtig model om de biologie van ameloblasten te begrijpen. En om deze biologie te begrijpen, kunnen we een verscheidenheid aan technieken gebruiken, waaronder elektronenmicroscopie, proteomica en genomica.Ameloblasten zijn fantastische cellen. Ja, ze hebben het vermogen om prismatisch tandglazuur af te scheiden. Dit bioreactormodel brengt ons in de positie om te begrijpen hoe ameloblasten matrix afscheiden en apatietmineralen afscheiden om prismatisch tandglazuur te laten groeien.
