De fleste af de excitatoriske synapser i centralnervesystemet er placeret på dendritiske rygsøjler - små fremspring af en neuronal membran. Disse fremspring danner begrænsede biokemiske rum, der styrer intracellulær signaltransduktion. Strukturel plasticitet af dendritiske rygsøjler og synapser er tæt forbundet med de funktionelle ændringer i synaptisk effekt, der ligger til grund for så vigtige processer som læring og hukommelse1,2. Det er vigtigt at bemærke, at elektronmikroskopi (EM) er den eneste teknik, der gør det muligt at afgøre, om en dendritisk rygsøjle har et præsynaptisk input. EM-opløsning er også nødvendig for at studere ultrastrukturelle detaljer såsom form af en postsynaptisk tæthed (PSD), der repræsenterer en postsynaptisk del af en synapse, eller dimensioner af en dendritisk rygsøjle, samt størrelsen og formen af en axonal bouton. Derudover er det med EM muligt at visualisere synapser og deres omgivelser.
Takket være fremskridt inden for billed- og computerteknologier er det muligt at rekonstruere hele neurale kredsløb. Volumenelektronmikroskopiteknikker, såsom seriel sektionstransmissionselektronmikroskopi (ssTEM), seriel blok-ansigtsscanning elektronmikroskopi (SBEM) og fokuseret ionstrålescanning elektronmikroskopi (FIB-SEM) er almindeligt anvendt til neuronale kredsløbsrekonstruktioner3.
I vores undersøgelser anvendes SBEM-metoden med succes til at undersøge den strukturelle plasticitet af dendritiske rygsøjler og PSD'er i prøver af musen hippocampus og organotypiske hjerneskiver 4,5. SBEM er baseret på installation af en miniature ultramikromitomi inde i scanningselektronmikroskopkammeret6,7,8,9. Toppen af prøveblokken afbildes, og derefter skæres prøven i en bestemt dybde af ultramikrotomen og afslører en ny blokflade, som igen afbildes, og derefter gentages processen8. Som følge heraf er det kun billedet af en blok-ansigt er tilbage, mens skiven, der er blevet skåret er tabt som snavs. Derfor kaldes SBEM en destruktiv teknik, hvilket betyder, at det ikke er muligt at afbilde det samme sted igen. Fordelen ved de destruktive on-block-metoder er imidlertid, at de ikke lider af vridningsproblemer og sektionstab, der kan påvirke datakvaliteten og dataanalysen3betydeligt . Desuden giver SBEM mulighed for at afbilde et relativt stort synsfelt ( > 0,5 mm × 0,5 mm) ved høj opløsning3.
For at anvende SBEM skal prøverne forberedes i henhold til en dedikeret, meget kontrasterende protokol på grund af den backscattered elektrondetektor, der bruges til at erhverve billeder. Vi viser her, hvordan man udfører prøveforberedelse i henhold til protokollen baseret på en procedure udviklet af Deerinck10 (National Center for Microscopy and Imaging Research (NCMIR) metode), ved hjælp af reducerede osmium-thiocarbohydrazide-osmium (rOTO) pletter udviklet i 1980'erne8,11. Derudover introducerer vi en to-trins fikseringsmetode med mild fikseringsperfusion, der gør det muligt at bruge den samme hjerne både til immunfluorescensundersøgelser med lysmikroskopi og SBEM.
I protokollen er en musehjerne primært fastgjort med et mildt fikseringsmiddel og skæres derefter i halve, og den ene halvkugle anbringes og forberedes til immunfluorescens (IF), mens den anden til EM-undersøgelser (Figur 1).

Tredimensionel elektronmikroskopi (3D EM) giver mulighed for at analysere morfologiske parametre for dendritiske rygsøjler med nanoskalaopløsning. Hver neuron i hjernen er forbundet med andre neuronale celler med ca. 7.000 synapser. Bortset fra, at disse synapser i pattedyrshjernen hovedsageligt er placeret på membranens små fremspring kaldet dendritiske rygsøjler. Plasticitet af synapser og dendritiske rygsøjler og lignende, så vigtige hjernefunktioner som læring og hukommelsesprocesser.Det er vigtigt at bemærke, at kun elektronmikroskopi giver et fuldt indblik i, om en dendritisk rygsøjle har et postsynaptisk input. Tag en aluminiumsnål, der er blevet affedtet med ethanol. Bland en ledende epoxy godt og bruge en lille mængde af det til at montere prøven til stiften.Cure ledende epoxy ved 70 grader Celsius i 10 minutter. Trim hver side af prøveblokken med diamantkniven, og puds derefter blokkens ansigt, indtil vævet udsættes. Jord prøven til stiften ved hjælp af ledende maling og helbrede det.For at minimere opladningen skal prøverne også ses belagt med et tyndt lag guld eller guldpaladium. Fastgør stiften med prøven i kammeret på det serielle blokbaserede scanningselektronmikroskop. Prøven justeres til kniven, og luk derefter kammeret, og angiv parametrene.Indsaml en stak billeder. Ved hjælp af den beskrevne metode blev der opnået billeder af musehjernevæv. Det store billede af hippocampus en region blev taget, og billeddannelsen blev sat i midten af stratum radiatum.En stak billeder blev erhvervet, og objekter af interesse, såsom dendritiske rygsøjler og synapser, blev segmenteret. God kvalitet væv morfologi blev opnået med klart synlige membraner og synaptiske vesikler og velbevarede mitokondrier. Immunfluorescerende undersøgelser af PSD 95 antistof bekræftede, at mild primær fiksering og traditionel fiksering med 4%PFA giver sammenlignelige resultater.Præsenteret protokol gør det muligt at erhverve hjernevævsbilleder ved hjælp af seriel blokbaseret scanningselektronmikroskopi, vævsmorfologi af høj kvalitet og 12 synlige membraner letter 10 rigtige segmentering og rekonstruktion. Min primære fiksering gjorde det muligt at bruge de samme hjerner til analyse af PSD 95 immunfluorescens og elektronmikroskopi imaging dendritiske rygsøjler. Her bruger vi PSE 9, 500 spirende.Den anden kan dog også bruges.
