F420生合成には少なくとも3つの経路があります2,6,24。初めに、生合成経路は5-デアザフラビン生合成および2-ホスホラク酸代謝分岐に分割される。F420分子の反応性部分は、フィロシンと5-アミノ-6-リビチルアミノ-2,4(1H, 3H)-ピリミジンジオンの基質を用いてFO-シンターゼを介して合成される。結果はリボフラビンレベルのクロモフォアFOです。現在受け入れられている乳酸代謝ブランチ内では、L-乳酸はL-乳酸キナーゼ(CofB)によって2-ホスホ-L-乳酸にリン酸化される。2-ホスホ-L-乳酸は、今度は、2-ホスホ-L-乳酸グアニリルトランスファーゼ(CofC)によってL-ラクチル-2-ジホスホ-5'-グアノシンにグアニル化される。次のステップでは、L-ラクチル-2-ジホスホ-5'-グアノシンは、F420-02を形成する2-ホスホ-L-乳酸トランスファーゼ(CofD)によってFOにリンクされています。最後に、酵素F420-0:ɣグルタミルリガーゼ(CofE)はグルタミン酸モノマーをF420-0にリゲートし、異なる数23,25で最終的な補因子6を形成する。異なる生物は、ミコバクテリア2,25,26よりも安価な尾がメサノゲンに見られる短い尾で、付着したグルタミン酸残基の数で異なるパターンを示す。一般的に、メタノゲンは2から3までの尾の長さを示し、アセトクラス性メタノゲン、メサノサルシナsp.で最大5つ、マイコバクテリウムspに見られる尾長は5〜7個のグルタミン酸残基2,25,26,27の範囲でした。しかし、最近の知見では、長鎖F420が短鎖F420よりも高い親和性を有するF420依存性オキシド還元酵素に結合することが示された。また、結合長鎖F420は基質親和性を高めるが、それぞれの酵素23の回転率を低下させる。
補因子F420の検出は、多くの場合、その蛍光に基づいています。それにより、そのオリゴグルタミン酸誘導体を逆相(RP)-HPLC27,28を用いて分離した。最近、Ney et al. は、負に荷電したグルタミン酸尾のイオン対向試薬として水酸化テトラブチラモニウムを使用し、RP-HLPC上での分離を正常に促進した5。ここでは、純粋な培養物からだけでなく、異なる環境サンプル(土壌および消化剤汚泥)からの補因子F420のサンプル、その後のリシス、抽出、精製、分離、定量の調製方法を提示する。
