Overvågning svampecellevækst og fænotype ved time-lapse mikroskopi er en kraftfuld tilgang til at vurdere cellulære adfærd i realtid og kvantitativt og præcist afgøre, om en bestemt lægemiddelbehandling og / eller genetisk intervention resulterer i påviselig cellevækst eller fænotypiske forskelle over tid.
I denne undersøgelse blev en pålidelig live-celle billeddannelsesmetode beskrevet til at måle og kvantitativt analysere svampeudvikling, herunder dynamikken i kimrør og hyphal...

Filamentøse svampe er af stor socioøkonomisk og økologisk betydning, idet de begge er afgørende som industrielle / landbrugsmæssige værktøjer til enzym- og antibiotikaproduktion1,2 og som patogener af afgrødeplanter3,skadedyrsinsekter4 og mennesker3. Desuden er filamentøse svampe som Aspergillus nidulans meget udbredt som modelorganismer til grundforskning, såsom undersøgelser i genetik, celle- og evolutionær biologi samt til undersøgelse af hyphalforlængelse5. Filamentøse svampe er stærkt polariserede organismer, der forlænger gennem kontinuerlig forsyning af membran lipider / proteiner og de novo syntese af cellevæg ved den forlængende spids6. En central rolle i hyphal tip vækst og polaritet vedligeholdelse er en specialiseret struktur ved navn 'Spitzenkorper' (SPK), en højt bestilt struktur, der hovedsagelig består af cytoskeletale komponenter og polariseret distribution af Golgi6,7,8.
Miljømæssige stimuli / signaler, sådan vand-luft interface, lys, CO2 koncentration, og den ernæringsmæssige status er ansvarlige for de udviklingsmæssige beslutninger truffet af disse forme9. I nedsænkede (flydende) kulturer er differentiering af A. nidulans undertrykt, og væksten opstår ved hyphal tip elongation6. Under vegetativ vækst spirer aseksuel sporer (conidia) ved apikal udvidelse og danner et udifferentieret netværk af indbyrdes forbundne hyphalceller, myceliet, som kan fortsætte med at vokse på ubestemt tid, så længe næringsstoffer og plads er tilgængelige. På den anden side, på solide medier hyphal tips forlænge og efter en defineret periode med vegetativ vækst (udviklingskompetence), aseksuel reproduktion er indledt og antenne conidiophore stilke strækker sig fra specialiserede fodceller i mycelium6. Disse giver anledning til specialiserede udviklingsmæssige flercellede strukturer kaldet conidiophores, som producerer lange kæder af haploid conidia10, der kan genstarte væksten under gunstige miljøforhold.
En udbredt metode til måling af filamentøs svampevækst er at vaccinere sporer på næringsstof agar indeholdt i en petriskål og makroskopisk måle diameteren af kolonien et par dage senere11. Diameteren / området af kolonien, mest afhængig af ændringer i mycelial vækstrate og mindre på conidiophore tæthed12, bruges derefter som en værdi af vækst. Selvom måling af svampepopulationens (koloni) størrelse, der vokser på faste overflader, er helt tilstrækkelig, er det på ingen måde det mest nøjagtige mål for vækst. Sammenlignet med befolkningsgennemsnit (gennemsnit af svampekolonistørrelse) kan målinger af en enkelt celle fange en cellepopulations heterogenitet og gøre det muligt at identificere nye underpopulationer af celler, stater13, dynamik, veje samt de biologiske mekanismer, hvormed celler reagerer på endogene og miljømæssige ændringer14,15. Overvågning svampe cellevækst og fænotype ved time-lapse mikroskopi er velsagtens den mest udbredte kvantitative enkelt celle observation tilgang.

quantitative analysis of aspergillus nidulans growth rate using live microscopy and open-source software

Det er veletableret, at koloni vækst af filamentøse svampe, for det meste afhængig af ændringer i hyphae / mycelia apikale vækstrate, er makroskopisk anslået på størknede medier ved at sammenligne koloni størrelse. Det er imidlertid en udfordring kvantitativt at måle vækstraten for genetisk forskellige svampestammer eller -stammer under forskellige miljø-/vækstbetingelser (pH, temperatur, kulstof- og kvælstofkilder, antibiotika osv.). Således bliver forfølgelsen af komplementære tilgange til kvantificering af vækst kinetiker obligatorisk for bedre at forstå svampecellevækst. Desuden er det velkendt, at filamentøse svampe, herunder Aspergillus spp., har forskellige former for vækst og differentiering under sub-antenne betingelser på solide medier eller nedsænket kulturer. Her beskriver vi en kvantitativ mikroskopisk metode til analyse af vækstkinetik af modelsvampen Aspergillus nidulans, ved hjælp af levende billeddannelse i både nedsænkede kulturer og solide medier. Vi tager billeder, analyserer og kvantificerer vækstrater for forskellige svampestammer på en reproducerbar og pålidelig måde ved hjælp af en open source, gratis software til biobilleder (f.eks. Fiji), på en måde, der ikke kræver nogen forudgående billedanalyseekspertise fra brugeren.

Efter denne protokol fangede og analyserede vi forskellige billeder svarende til forskellige vækst / udviklingsstadier af filamentøs svamp A. nidulans. De data, der præsenteres i denne undersøgelse blev behandlet og analyseret ved hjælp af Fiji software. Målinger blev gemt som csv-filer, statistisk analyseret og udarbejdet som grafer ved hjælp af kommerciel statistisk software og / eller Python programmeringssprog ved hjælp af softwarebiblioteker som pandaer, numpy, statsmodeller, matplotlib og søfødt....

I denne video beskriver vi en etiketfri live imaging protokol til at fange billeder af modellen svamp aspergillus nidulans, ved hjælp af transmitteret lys mikroskopi teknikker. Vi bruger disse billeder til at analysere og kvantificere vækst kinetik af aspergillus nidulans i både flydende og faste medier. En visuel repræsentation af denne protokol giver brugerne mulighed for at blive fortrolige med vigtige trin i billedoptagelse og mikroskopi billedbehandling.En almindelig metode, der anvendes til at måle filamentøs svampevækst er to podede svampesporer i en petriskål indeholdende næringsstof agar og til at måle diameteren af den udviklende koloni et par dage senere. Denne tilgang er imidlertid ikke følsom nok til at kvantificere fine vækstforskelle. Begynd med at hælde 15 milliliter flydende næringsstof agar i petriskåle.Læg låget på toppen og lad det køle af. Brug UV-stråling til at sterilisere plader. Før striber ud svampestammen af interesse fra en bestand, varme den nikkende løkke i Bunsen brænderen, indtil det er rødglødende, afkøles løkken ved at stikke den ind i agaren.Vortex røret og streak løkken over overfladen af agar minimal medium suppleret med de relevante ernæringsmæssige krav. Inkuber plader i to til tre dage ved 37 grader Celsius. Begynd med at importere billederne til Fiji ved hjælp af plugins med et format.Vælg standardfarvetilstanden og den automatiske skalering. For at visualisere et tidsstempel og en skalalinje, når overlejring går til analyse, værktøjer, skaleringslinje. Angiv de ønskede parametre for bredden, højden, farven og placeringen af skalalinjen.Gå til billede, egenskaber, for at vise billedegenskaber i imageJ Fiji software. Overhold oplysninger om rammeinterval. Her bruger vi et tidsinterval på ca. 15 minutter og trykker på okay.Gå derefter til billede, stakke, etiket og indstil de korrekte oplysninger til interval. Angiv placeringen af etiketten ved at ændre X og Y.Angiv måleenheden i feltteksten, og tryk på Eksempel. Brug histogramme matching til belysning korrektion mellem forskellige rammer ved at vælge billede, justere, blegemiddel korrektion, histogramme matching.Der vises et nyt vindue med korrigeret belysning. Brug MtrackJ plugin på plugins, MtrackJ, til at spore voksende hyfal tip For at tilføje sporet skal du vælge tilføj-knappen på værktøjslinjen og placere det første punkt på hyphalspidsen ved hjælp af musens venstre klik. Det viser statistisk signifikante lavere vækstrate målinger i forhold til den vilde type stamme i nedsænket flydende kulturer. Denne forskel i vækst kan ikke påvises ved at måle koloniområdet af disse to stammer i fast minimal medium.Enkelt celle, langsigtet levende billeddannelse er af betydelig værdi i bestræbelserne på at opnå spacial og tidsmæssig information af cellulære proteiner dynamik. Denne protokol er også velegnet til udførelse af langsigtet levende cellebilleddannelse. Her ser vi spiring af conidia co-udtrykke GFP mærket kerne eisosomal protein PilA og mRFP histonen H1. Her præsenterer vi en protokol til analyse af svampevækst kinetik på en reproducerbar og pålidelig måde uden behov for nogen forudgående billedanalyse erfaring fra brugeren.Denne protokol giver mulighed for objektiv præcis kvantificering af svampevækst.
