糸状菌は、酵素および抗生物質生産1、2、作物植物3、害虫昆虫4およびヒト3の病原体のための産業/農業用具として重要であること、社会経済的、生態学的に重要な大きな社会経済的、生態学的重要性を持つ。また、アスペルギルスニデュランなどの糸状菌は、遺伝学、細胞、進化生物学の研究や、ヒュファル拡張5の研究などの基礎研究のモデル生物として広く使用されている。糸状菌は、膜脂質/タンパク質の連続供給と延伸先端部の細胞壁のデノボ合成を通じて伸びる高度に偏光した生物である6。催眠先端の成長と極性維持における中心的な役割は、細胞骨格成分とゴルジ6、7、8の偏光分布を主に構成する高度に順序付けられた構造である「スピッツェンコルパー」(SPK)という特殊な構造です。
環境刺激/信号、水と空気の界面、光、CO2 濃度、栄養状態は、これらの金型9によって行われた発達上の決定に責任を負う。水没(液体)培養では 、A.ニデュラン の分化が抑圧され、ヒファル先端伸長6によって成長が起こる。栄養成長の間、無性胞子(コニディア)は、補助的な拡張によって発芽し、相互接続された催眠細胞、菌糸体の未分化ネットワークを形成し、栄養素および空間が利用可能である限り無限に増殖し続ける可能性がある。一方、固体培地の催眠のヒントは、栄養成長(発達能力)の定義された期間(発達能力)の後に、無性生殖が開始され、空気共生茎が菌糸体6の特殊な足細胞から伸びる。これらは、有利な環境条件下で成長を再開することができるハプロイドコニディア10 の長い鎖を生成するコニジオフォアと呼ばれる特殊な発達多細胞構造を生み出す。
糸状菌の成長を測定するために広く使用されている方法は、ペトリ皿に含まれる栄養寒天上の胞子を接種し、数日後にコロニーの直径をマクロコスコープで測定する。コロニーの直径/面積は、最も、心の成長速度の変化に依存し、コニディオフォア密度12に少ない、成長の値として使用されます。しかし、固体表面上に成長する真菌集団(コロニー)サイズを測定することは非常に適切であるが、それは決して最も正確な成長の尺度ではない。集団レベル平均(真菌コロニーサイズの平均)と比較して、単一細胞測定は、細胞集団の不均一性を捕捉し、細胞の新しいサブ集団、状態13、ダイナミクス、経路、ならびに細胞が内因性および環境変化に応答する生物学的メカニズムを同定することを可能にする14、15。タイムラプス顕微鏡による真菌細胞の増殖および表現型をモニタリングすることは、おそらく最も広く使用されている定量的単一細胞観察アプローチである。
ここでは、透過光顕微鏡技術(位相コントラスト、微分干渉コントラスト(DIC)、偏光顕微鏡など)を用いたラベルフリーライブイメージングプロトコルを詳述し、蛍光顕微鏡の併用とは無関係に、水没培養物と固体培地の両方でのA. ニドゥラン 株の極性成長を分析し定量することができる画像を取り込む。
